Effects of temperature on the activity of flucycloxuron on larval stages of Panonychus ulmi (Koch), based on LC50 values, were highly significant (P < 0.001) with temperature coefficients of-1.7 in both the ranges of 15° to 25°C and 20° to 30°C. The slopes of probit regression lines at 15° and 20°C were significantly steeper than those at 25° and 30°C. As a consequence the temperature coefficients based on LC90 values were-4.4 and-2.2, for the 2 temperature ranges. The ovicidal activity of flucycloxuron on P. ulmi was low and was only statistically detectable at 20°C (LC90 of 84 mg a.i./l). In studies with larvae of Aedes aegypti (Linnaeus), Leptinotarsa decemlineata (Say), Plutella xylostella (Linnaeus), Spodeptera exigua (Hübner) and Spodoptera littoralis (Boisduval) probit regression lines were parallel over temperature. The activity of flucycloxuron on these five insect species was not affected by temperature. Based on LC50 values, diflubenzuron showed positive temperature coefficients on P. xylostella of + 2.1 at 15° to 25°C and + 2.5 at 20° to 30°C. For S. littoralis the temperature coefficient was positive (+ 2.4) at 15° to 25°C but negative (-1.9) at the 20° to 30°C range. Temperature coefficients of diflubenzuron were neutral for A. aegypti, L. decemlineata and S. exigua. In the design and analysis of these studies special allowance was made for date effects and variation in natural mortality over temperature.  相似文献

A new culture method for the injection of tobacco mesophyll protoplasts has been established. The protoplasts are embedded in a thin layer of alginate and are nourished from the medium in the underlying basislayer. In the alginate layer the protoplasts regenerate to calli at a frequency of up to 80%. Embedded protoplasts can be selected either with 50 mg l−1 kanamycin or 5 mg l−1 paromomycin. Single resistant cells can be recovered from about 10 000 sensitive cells in one alginate layer. Injection of theneo gene (coding for neomycin phosphotransferase II) into protoplast derived single cells in the alginate layer results in kanamycin resistant colonies that can be regenerated to mature plants. These plants express the neomycin phosphotransferase as shown by enzyme activity assay. The integration of the transgene into the plant genome could be proved by Southern hybridization to high molecular weight DNA. With this culture method 100 cells can be injected per hour. Transformation frequencies range from 2 to 20%. In crossing experiments, it was shown that the foreign gene is transmitted to the next generation in a Mendelian fashion.  相似文献

A sample of seventy-twoAphis gossypii Glover (Homoptera: Aphididae) clones was collected in south-eastern France. The efficiency of these clones to transmit a potyvirus (papaya ringspot virus T-strain) was assessed in controlled conditions. In a first screening, the virus was transmitted by all clones and a 3.5-fold difference between the most and least efficient clones was obtained. During subsequent trials, which were carried out to confirm the differences in the transmission efficiency of these clones, only one clone proved to be more efficient than the others. This difference appeared consistent over a 1-year period, and was also confirmed with 4 other related potyviruses.
Résumé Un échantillon de 72 clones deA. gossypii a été collecté dans le Sud-Est de la France. L'efficacité de transmission d'un potyvirus (PRSV-T) a été mesurée en conditions contr?lées pour chacun de ces clones. Un premier screening a permis de montrer que tous les clones transmettaient ce virus, et qu'un rapport de 3,5 existait entre l'efficacité de transmission du clone le plus efficace et celle du clone le moins efficace. Au cours des essais ultérieurs destinés à confirmer ces différences, un seul clone s'est montré significativement plus efficace que les autres. Cette différence s'est maintenue pendant la période d'essais (1 an). Elle s'est reproduite avec 4 autre potyvirus apparentés.

A full diallel was carried out with six diploid species of Allium section Cepa and A. roylei of section Rhizirideum , High isolation barriers were found between the related species A. cepa and A. oschaninii , between A. oschaninii and A. vavilovii , and between A. galanthum and A. pskemense . On the contrary, the species A. cepa and A. roylei , belonging to different sections, show only slight isolation barriers. The Wallace effect, which is the development of internal isolation barriers as such, is likely to have taken place in the evolution of A. oschaninii and A. vavilovii , and possibly also between A. galanthum and A. pskemense .  相似文献

The aim of this study was to investigate the ability of Pantoea agglomerans, a plant growth-promoting bacterium, to colonize various regions and tissues of the wheat plant (Triticum aestivum L.) by using different inoculation methods and inoculum concentrations. In addition, the enzyme-linked immunosorbent assay (ELISA) and transmission electron microscopy (TEM) were used to determine: (a) the ability of the bacterial cells to grow and survive both on the surface and within internal tissue of the plant and (b) the response of the plant to bacterial infection. After inoculation, cells of the diazotrophic bacterial strain P. agglomerans were found to be located in roots, stems and leaves. Colony development of bacterial cells was only detected within intercellular spaces of the root and on the root surface. However, single bacterial cells were observed in leaves and stems on the surface of the epidermis, in the vicinity to stomatal cells, within intercellular spaces of the mesophyll and within xylem vessels. Inoculated bacterial cells were found to be able to enter host tissues, to multiply in the plant and to maintain a delicate relationship between endophyte and host. The density of bacterial settlement in the plant in all experiments was about 106 to 107 cells per mL root or shoot sap. Establishment was confirmed by a low coefficient of variation of ELISA means at these concentrations.  相似文献

Model membranes with unsaturated lipid chains containing various amounts of M13 coat protein in the -helical form were studied using time-resolved fluorescence and ESR spectroscopy. The lipid-to-protein (L/P) ratios used were > 12 to avoid protein-protein contacts and irreversible aggregation leading to -polymeric coat protein. In the ESR spectra of the 12-SASL probe in dioleoyl phosphatidylcholine (DOPC) bilayers no second protein induced component is observed upon incorporation of M13 coat protein. However, strong effects are detected on the ESR lineshapes upon changing the protein concentration. The ESR lineshapes are simulated by assuming a fixed ratio between the parallel (D) and perpendicular (D) diffusion coefficients of 4, and an order parameter equal to zero. It is found that increasing the protein concentration from L/P to L/P 15 results in a decrease of the rotational diffusion coefficient D from 3.4 × 107 to 1.9 × 107 s–1. In the time-resolved fluorescence experiments with DPH-propionic acid as a probe, it is observed that increasing the M13 coat protein concentration causes an increase of the two fluorescent lifetimes, indicating an increase in bilayer order. Analysis of the time-resolved fluorescence anisotropy decay allows one to quantitatively determine the order parameters P2 and P4, and the rotational diffusion coefficient D of the fluorescent probe. The order parameters P2 and P4 increase from 0.34 to 0.55 and from 0.59 to 0.77, respectively, upon adding M13 coat protein to DOPC bilayers with an L/P ratio of 35. The rotational diffusion coefficient D of the DPH-propionic acid probe decreases on incorporating M13 coat protein, in accordance with the ESR results. It is concluded that M13 coat protein in the -monomeric state is not able to produce a long living lipid boundary shell and consequently an immobilization of the lipids. An overall effect on the lipids is induced, resulting in a reduction in the dynamics and an increase in average lipid order. The hydrophobic region of M13 coat protein is proposed to perfectly match the lipid bilayer, resulting in a relatively small distortion of the bilayer structure of the lipid system.  相似文献

This paper describes the effects of carbon dioxide anaesthesia on the ability of the planthopper Laodelphax striatellus to subsequently acquire and transmit maize rough dwarf reovirus (MRDV) and barley yellow striate mosaic rhabdovirus (BYSMV), both propagative in the vector. Anaesthesia reduced the acquisition of MRDV by about 45%, but did not affect acquisition of BYSMV. In serial transfer transmission tests, anaesthesia caused interruption of transmission of both MRDV and BYSMV in about 11% of infectious insects; the effect may be due to impaired ability to find the phloem. The effect was stronger on third instar nymphs than on adults. Transmission of MRDV was resumed 3 days after anaesthesia, but none of the insects that stopped transmitting BYSMV resumed it. The survival of insects which ceased to transmit BYSMV was significantly lower than that of hoppers that continued to transmit. No other effects on survival were detected. A possible interaction of carbon dioxide with BYSMV in L. striatellus is discussed, in the light of effects of carbon dioxide on Drosophila melanogaster infected by some rhabdoviruses.  相似文献

Phytoplankton primary production, nutrient concentrations and turbidity were monitored at three stations in the Oosterschelde during 1980–1984 as part of an ecosystem study.From comparisons of dissolved nutrient ratios with the nutrient requirements of phytoplankton, and of ambient nutrient concentrations with half-saturation constants for nutrient uptake by natural phytoplankton populations it was concluded that silicate was a limiting nutrient for diatoms after the spring bloom until the end of the summer. Dissolved inorganic nitrogen and phosphate were not considered to be limiting to phytoplankton growth.In general, the phytoplankton growing season started during the first fortnight of April and ended at the end of September. Column production in the whole Oosterschelde varied between 201 and 540 g C m–2 yr–1 and was, on average, 25% higher in the western part than in the eastern part. Basin production in the Oosterschelde varied between 120 and 466 g C m–2 yr–1 and was, on average, 55% higher in the western part than in the eastern part; this difference could be explained by differences in the ratio of euphotic depth to mean depth of the compartments.Estimated carbon-specific growth rates in the eastern part varied between < 0.1 and 3 d–1 and in the western part between < 0.1 and 1 d–1. This difference could be explained by the great differences in depth of the compartments. Carbon-specific growth rates are discussed in relation to phytoplankton loss rates. Our study of cultural transmission in Taiwan is based on a survey of 1000 students, their families, and friends, for characters ranging from religion to various customs and beliefs, as well as entertainment and hygienic habits. The effects of father, mother, and an older sib on a propositus are tested by an additive model of transmission, using a novel statistical procedure, and compared with correlations with friends. For many traits there exist significant influences; older sibs are almost as important as father and mother, with effects differing somewhat with their sex. Formulas for recurrences and equilibrium frequency of a cultural character for which father, mother, and sib are active in transmission are given in the appendices along with formulas for estimation of their effects from real data.This research was supported in part by grant NIH GM 20467 and NIH GM 20816. Summary An attempt to produce a set of addition lines of Aegilops sharonensis to the wheat variety Chinese Spring produced only one addition line. This was due to preferential transmission of one chromosome from Ae. sharonensis. This chromosome was studied in detail by established cytological methods of chromosome observation and by the newer techniques of C-banding and in situ hybridization of a cloned DNA sequence. The chromosome was found to be partially homologous to an Ae. sharonensis chromosome of similar behaviour in another wheat addition line. The incomplete homology of the two Ae. sharonensis chromosomes was due to the presence of a translocated segment of a wheat chromosome. — Substitution lines of the Ae. sharonensis chromosome for wheat homoeologous group 4 were produced and the Ae. sharonensis chromosome thereby designated 4 S l .  相似文献
