The stable isotope15N was added as (15NH4)2SO4 to throughfall water for one year, to study the fate of the deposited nitrogen at different levels of N deposition in two N saturated coniferous forests ecosystems in the Netherlands. The fate of the15N was followed at high-N (44–55 kg N ha–1 yr–1) 1) and low-N (4–6 kg N ha–1 yr–1) deposition in plots established under transparent roofs build under the canopy in a Douglas fir (Pseudotsuga menziesii (Mirb.) Franco.) and Scots pine (Pinus sylvestris L.) forest.The applied15N was detectable in needles and twigs, the soil and soil water leaching below the rooting zone (90 cm depth). Total15N recovery in major ecosystem compartments was 71–100% during two successive growing seasons after the start of a year-round15N application to throughfall-N. Nine months after the year-round15N application, the15N assimilated into tree biomass was 29–33% of the15N added in the Douglas fir stand and less than 17% in the Scots pine stand. At the same time total15N retention in the soil (down to 70 cm) of the high-N plots was about 37% of the deposited15NH4-N, whereas 46% and 65% of the15N was found in the soil of the low-N deposition plots at the Douglas fir and Scots pine stand, respectively. The organic layers accounted for 60% of the15N retained in the soil. The total N deposition exceeded the demand of the vegetation and microbial immobilization. Total15N leaching losses within a year (below 90 cm) were 10–20% in the high-N deposition plots in comparison to 2–6% in the lowered nitrogen input plots. Relative retention in the soil and vegetation increased at lower N-input levels.Species differences in uptake and tree health seem to contribute to lower15N recoveries in the Scots pine trees compared to the Douglas fir trees. The excessive N deposition and resulting N saturation lead to conditions were the health and functioning of biota were negatively influenced. At decreased N deposition, lower leaching losses together with increased soil and plant retention indicated a change in the fate of the15N deposited. This may have resulted from changes in ecosystem processes, and thus a shift along the continuum of N saturation to N limitation.  相似文献

Using a combined method of pulse-labelling trees and analysing detailed distribution of 13C tracer within tree rings, we studied how photo-assimilates incorporated on a given day are then distributed in a tree ring. A branch of a 4-year-old Cryptomeria japonica D.Don tree growing in Tsukuba, Japan was pulse-labelled with non-radioactive 13CO2 on two occasions: 29 May 2001 and 18 September 2001. Two discs were cut from the stem on 4 March 2002, one immediately under and the other 0.5 m below the branch and put through high-resolution δ 13C analysis. δ 13C peaks were observed in both the earlywood and latewood of the concerned tree ring, corresponding to each pulse-labelling date. The earlywood peaks was broader than the latewood peaks, possibly reflecting seasonal variation of the width of wood developing zone. Half-widths of the peaks were measured and used as indicators for the potential time resolution of tree-ring isotope analysis. The half-widths of the peaks indicated a time resolution no finer than 8.7–28 and 33–42 d in the early and latewood, respectively. Holocellulose extraction yielded only a slight change to the shape of the δ 13C peaks. 13C tracer pulse-labelled in May and September reached tangentially different locations in the lower disc, suggesting a seasonal change in the pathway of carbohydrates. Local consumption of spring assimilates and long-distance downward transport of autumn assimilates were also suggested.  相似文献

Apoplastic transport across young maize roots: effect of the exodermis   总被引:27,自引:0,他引:27  
The uptake of water and of the fluorescent apoplastic dye PTS (trisodium 3-hydroxy-5,8,10-pyrenetrisulfonate) by root systems of young maize (Zea mays L.) seedlings (age: 11–21 d) has been studied with plants which either developed an exodermis (Casparian band in the hypodermis) or were lacking it. Steady-state techniques were used to measure water uptake across excised roots. Either hydrostatic or osmotic pressure gradients were applied to induce water flows. Roots without an exodermis were obtained from plants grown in hydroponic culture. Roots which developed an exodermis were obtained using an aeroponic (=mist) cultivation method. When the osmotic concentration of the medium was varied, the hydraulic conductivity of the root (Lp r in m3 · m−2 · MPa−1 · s−1) depended on the osmotic pressure gradient applied between root xylem and medium. Increasing the gradient (i.e. decreasing the osmotic concentration of the medium; range: zero to 40 mM of mannitol), increased the osmotic Lp r. In the presence of hydrostatic pressure gradients applied by a pressure chamber, root Lp r was constant over the entire range of pressures (0–0.4 MPa). The presence of an exodermis reduced root Lp r in hydrostatic experiments by a factor of 3.6. When the osmotic pressure of the medium was low (i.e. in the presence of a strong osmotic gradient between xylem sap and medium), the presence of an exodermis caused the same reduction of root Lp r in osmotic experiments as in hydrostatic ones. However, when the osmotic concentration of the medium was increased (i.e. the presence of low gradients of osmotic pressure), no marked effect of growth conditions on osmotic root Lp r was found. Under these conditions, the absolute value of osmotic root Lp r was lower by factors of 22 (hydroponic culture) and 9.7 (aeroponic culture) than in the corresponding experiments at low osmotic concentration. Apoplastic flow of PTS was low. In hydrostatic experiments, xylem exudate contained only 0.3% of the PTS concentration of the bathing medium. In the presence of osmotic pressure gradients, the apoplastic flow of PTS was further reduced by one order of magnitude. In both types of experiments, the development of an exodermis did not affect PTS flow. In osmotic experiments, the effect of the absolute value of the driving force cannot be explained in terms of a simple dilution effect (Fiscus model). The results indicate that the radial apoplastic flows of water and PTS across the root were affected differently by apoplastic barriers (Casparian bands) in the exodermis. It is concluded that, unlike water, the apoplastic flow of PTS is rate-limited at the endodermis rather than at the exodermis. The use of PTS as a tracer for apoplastic water should be abandoned. A network model for the determination of tumor metabolic fluxes from 13C NMR kinetic isotopomer data has been developed and validated with perfused human DB-1 melanoma cells carrying the BRAF V600E mutation, which promotes oxidative metabolism. The model generated in the bonded cumomer formalism describes key pathways of tumor intermediary metabolism and yields dynamic curves for positional isotopic enrichment and spin-spin multiplets. Cells attached to microcarrier beads were perfused with 26 mm [1,6-13C2]glucose under normoxic conditions at 37 °C and monitored by 13C NMR spectroscopy. Excellent agreement between model-predicted and experimentally measured values of the rates of oxygen and glucose consumption, lactate production, and glutamate pool size validated the model. ATP production by glycolytic and oxidative metabolism were compared under hyperglycemic normoxic conditions; 51% of the energy came from oxidative phosphorylation and 49% came from glycolysis. Even though the rate of glutamine uptake was ∼50% of the tricarboxylic acid cycle flux, the rate of ATP production from glutamine was essentially zero (no glutaminolysis). De novo fatty acid production was ∼6% of the tricarboxylic acid cycle flux. The oxidative pentose phosphate pathway flux was 3.6% of glycolysis, and three non-oxidative pentose phosphate pathway exchange fluxes were calculated. Mass spectrometry was then used to compare fluxes through various pathways under hyperglycemic (26 mm) and euglycemic (5 mm) conditions. Under euglycemic conditions glutamine uptake doubled, but ATP production from glutamine did not significantly change. A new parameter measuring the Warburg effect (the ratio of lactate production flux to pyruvate influx through the mitochondrial pyruvate carrier) was calculated to be 21, close to upper limit of oxidative metabolism.  相似文献

PurposeThe use of a magnetic nanoparticle tracer and handheld magnetometer for sentinel lymph node biopsy (SLNB) was recently introduced to overcome drawbacks associated with the use of radioisotope tracers. Unlike the gamma probe, the used magnetometers are not only sensitive to the tracer, but also the diamagnetic human body. This potentially limits the performance of the magnetometer when used clinically.MethodsA phantom, mimicking the magnetic and mechanical properties of the human axilla, was constructed. The depth performance of two current generation magnetometers was evaluated in this phantom. LN-phantoms with tracer uptake ranging from 5 to 500 μg iron were placed at clinically relevant depths of 2.5, 4 and 5.5 cm. Distance-response curves were obtained to quantify the depth performance of the probes.ResultsThe depth performance of both probes was limited. In the absence of diamagnetic material and forces on the probe (ideal conditions) a LN-phantom with high uptake (500 μg iron) could first be detected at 3.75 cm distance. In the phantom, only superficially placed LNs (2.5 cm) with high uptake (500 μg iron) could be detected from the surface. The penetration depth was insufficient to detect LNs with lower uptake, or which were located deeper.ConclusionThe detection distance of the current generation magnetometers is limited, and does not meet the demands formulated by the European Association for Nuclear Medicine for successful transcutaneous SLN localization. Future clinical trials should evaluate whether the limited depth sensitivity is of influence to the clinical outcome of the SLNB procedure.  相似文献

PET imaging of α-synuclein (α-syn) deposition in the brain will be an effective tool for earlier diagnosis of Parkinson's disease (PD) due to α-syn aggregation is the widely accepted biomarker for PD. However, the necessary PET radiotracer for imaging is clinically unavailable until now. The lead compound discovery is the first key step for the study. Herein, we initially established an efficient biologically evaluation system well in high throughput based on SPR technology, and identified a novel class of N, N-dibenzylcinnamamide (DBC) compounds as α-syn ligands through the assay. These compounds were proved to have high affinities against α-syn aggregates (KD < 10 nM), which well met the requirement of binding activity for the PET probe. These DBC compounds were firstly reported as α-syn ligands herein and the preliminary obtained structure has been further modified into F-labeled ones. Among them, a high-affinity tracer (541) with 1.03 nM (KD) has been acquired, indicating its potential as a new lead compound for developing PET radiotracer.  相似文献

Electrical and tracer techniques were used to investigate the movement of Na+, K+ and Cl? through discs of a range of thicknesses cut from the root tissue of sugar beet, Beta vulgaris L. cv. Amono. At low external concentration the electrical resistance across a discs is less than that of an equivalent volume of solution. This does not appear to be due to a low resistance symplastic pathway but rather, to an enhanced concentration of cation in the apoplast. The resistance is proportional to the thickness of tissue. Although measurement of diffusion potential gives about 25 mV difference across the disc for a ten-fold change in cation concentration, there is little discrimination between K+ and Na+. The observed tracer kinetics of 86Rb through the disc are consistent with those of diffusion, with a coefficient of diffusion, D, of 0.19 × 10?9 m2 s?1 and a tissue partition coefficient, k, of 0.27 (or of 2.7 if referred to the cell wall phase only). 36Cl gives a similar value for D, but has a k reduced by a factor of 3.3, a result that is consistent with the diffusion potential observation. However, a much larger discrimination would have been expected from the chemically measured cation exchange capacity.  相似文献

选用15N同位素标记的新型回收塑料包膜控释肥和大颗粒尿素,采用池栽试验研究夏玉米-冬小麦轮作体系中肥料氮的去向及利用率。结果表明,整个轮作体系中,控释肥处理(PCU)作物吸收的肥料氮为241.03 kg/hm,高于尿素处理(Urea)的211.02 kg/hm。控释肥处理施用的肥料氮主要残留在0~40 cm土层,而尿素处理则残留在0~60 cm土层,控释肥延缓了肥料氮向土壤深层迁移的趋势。在夏玉米和冬小麦轮作体系中,控释肥处理的氮肥利用率(32.86%,32.47%)高于尿素处理(28.23%,30.16%)。在冬小麦季,控释肥处理损失率相比尿素处理从36.07% 降至28.75%,而夏玉米季,控释肥处理损失率相比尿素处理从37.17%降至29.50%。玉米季控释肥处理与尿素处理差异不显著,但在冬小麦季控释肥处理的产量显著高于尿素处理。因此,在玉米和小麦整个生长季,新型回收塑料包膜控释肥的养分释放与作物养分需求吻合,既提高氮肥利用率,也降低了肥料氮的损失。  相似文献
