Summary The permeability and porosity of xylem cell walls are believed to play a major role in defining the ability of a cell or tissue to exhibit deep supercooling. Lanthanum nitrate, was utilized to contrast the permeability of stem tissues inB. lenta, which exhibits deep supercooling, withB. papyrifera, which exhibits equilibrium freezing. Although the two species differed greatiy in their response to low temperature, distribution of lanthanum deposits was quite similar. Primary cell walls of all xylem cell types appeared permeable although lanthanum deposition was patchy. Secondary cell walls of fiber cells were also permeable to lanthanum whereas the secondary wall of vessel elements and xylem parenchyma appeared impermeable to the lanthanum. Pit membranes, in all cell types and the protective layer in xylem parenchyma frequently exhibited deposits of lanthanum. Results of this study indicate that the porosity and permeability of the pit membrane, rather than the entire cell wall may determine the rate of water loss from xylem parenchyma to sites of extracellular ice. Summary Thin section and freeze-fracture electron microscopy revealed that the terminal bars of the larval midgut epithelium ofAeshna cyanea consisted of extended smooth septate junctions (SSJ), multiple adhesive junctions and rare gap junctions. Freeze-fractures of native tissue suggested that the septal building units were anchored only in the external membrane leaflet by partially integrated proteins while the interseptal pegs were anchored partly in both leaflets by completely integrated proteins and partly by presumed peripheral proteins.Reversible depletion of the physiological Ca++ concentration had no apparent structural effect on the SSJ of the terminal bars, but led to a reversible formation of junctional septa between the foot processes concomitant with a rearrangement of IMPs in the basolateral plasma membranes. The basolateral SSJ assembly and disassembly induced by reversible Ca++ deprivation was interpreted as exaggerated response of an intrinsic capability normally related to the apical growth of regenerative cells and to the extrusion of degenerating cells. Lanthanum tracer ingested with hyperosmotic drinking solution was always found excluded from the basolateral intercellular spaces underneath the terminal bar, but there was a dual effect on the SSJ structure. Evidence for bacterial urea production in marine sediments   总被引:1,自引:0,他引:1  
Abstract The quantitative importance of bacteria in urea production and turnover in a defaunated sediment from a Danish estuary was studied. After collecting the sediment, benthic infauna was removed by sieving, followed by anoxic pre-incubation for a week. Yeast extract was added to half of the samples. The urea concentration, urea production/turnover rates and the net ammonium production rate, were followed during a 338 h incubation. The urea concentration was highest in the enriched sediment, although urea production and turnover rates were the same in the enriched and control sediment, indicating that the urea concentration was controlled by the urea turnover rate constant, k urea. Net ammonium production, urea concetrations and teh urea turnover declined towards the end of the experiment. Comparison of the net ammonium production rate and the urea turnover rate in the enriched and control treatments, showed that urea hydrolysis accounted for up to 100% of the ammonium produced, from all sources. More than 98% of the urea which was hydrolyzed, originated from production during incubation. This suggests that urea was a major nitrogen excretion product from bacteria in this sediment, and that bacteria could also be an important source of urea in other marine sediments.  相似文献

The seasonal population trends of cattle nematodes in tracer calves are described from 1981 to 1987. Successive worm-free calves were grazed with growing beef cattle for 20–30 days at 6 week intervals and then slaughtered for nematode counts 2 weeks after their removal from pasture. Ostertagia, Cooperia, Trichostrongylus and Haemonchus were the main genera recovered. O.ostertagi was the most often found and acquisition of inhibition-prone larvae began in late winter and reached a peak in spring, while maximum larval availability was in autumn. The seasonal inhibition and larval availability pattern for T. axei was similar to that of Ostertagia. Cooperia showed greatest inhibition during winter with maximum larval availability in autumn and spring. Haemonchus was more prevalent during summer to early autumn and no inhibition was observed. It was concluded that infective larval availability for tracer calves was highest during autumn, and most reduced in summer. Summary The occurrence of tight junctions between Sertoli cells, providing the structural basis of the blood-testis barrier, has been studied using hypertonic fixative and lanthanum tracer in the testes of seven species of vertebrates having different testicular organization. In all cases inter-Sertoli tight junctions, establishing an effective barrier, appear either when meiosis is complete (teleosts and amphibians, both with cystic testes) or immediately after the onset of meiosis (reptiles and birds, both having testes consisting of seminiferous tubules). In the cystic testes, tight junctions are regularly associated with desmosomes, whereas in testes with seminiferous tubules, cisternae of the endoplasmic reticulum are present beneath the junctions (subsurface cisternae). The avian testes examined have, in addition, septate-like junctions between the Sertoli cells but before the tight junctions.Dedicated to Prof. Dr. H. Summary Tracer and freeze-fracture replication techniques show that there are two morphologically and topographically distinct permeability barriers in the epidermis of the grass snake. Tight junctions interconnect the apico-lateral plasma membranes of the uppermost living cells, establishing an ionic or osmotic gradient between the stratum germinativum and alpha layer. The second barrier is formed by intercellular lipid sheets in the overlying mesos layer, which are very similar to the barrier found in the stratum corneum of mammals.  相似文献

The effects of nymph size and temperature on the ingestion rate ofHexagenia limbata were investigated using14C labelled algae mixed into sediment. Ingestion rate increased with nymph length and temperature. Daily ingestion rates of large 19 mm nymphs burrowing in silt averaged 58 and 192% of the dry body weight at 16° and 21 °C, respectively. Ingestion of seston due to filter feeding in artificial burrows was insignificant compared to that ingested by burrowing nymphs.  相似文献

The retina, a thin tissue in the back of the eye, has two apparent sources of cholesterol: in situ biosynthesis and cholesterol available from the systemic circulation. The quantitative contributions of these two cholesterol sources to the retinal cholesterol pool are unknown and have been determined in the present work. A new methodology was used. Mice were given separately deuterium-labeled drinking water and chow containing 0.3% deuterium-labeled cholesterol. In the retina, the rate of total cholesterol input was 21 μg of cholesterol/g retina • day, of which 15 μg of cholesterol/g retina • day was provided by local biosynthesis and 6 μg of cholesterol/g retina • day was uptaken from the systemic circulation. Thus, local cholesterol biosynthesis accounts for the majority (72%) of retinal cholesterol input. We also quantified cholesterol input to mouse brain, the organ sharing important similarities with the retina. The rate of total cerebral cholesterol input was 121 μg of cholesterol/g brain • day with local biosynthesis providing 97% of total cholesterol input. Our work addresses a long-standing question in eye research and adds new knowledge to the potential use of statins (drugs that inhibit cholesterol biosynthesis) as therapeutics for age-related macular degeneration, a common blinding disease.  相似文献

The effects of increased reactive nitrogen (N) deposition in forests depend largely on its fate in the ecosystems. However, our knowledge on the fates of deposited N in tropical forest ecosystems and its retention mechanisms is limited. Here, we report the results from the first whole ecosystem 15N labeling experiment performed in a N‐rich old‐growth tropical forest in southern China. We added 15N tracer monthly as 15NH415NO3 for 1 year to control plots and to N‐fertilized plots (N‐plots, receiving additions of 50 kg N ha?1 yr?1 for 10 years). Tracer recoveries in major ecosystem compartments were quantified 4 months after the last addition. Tracer recoveries in soil solution were monitored monthly to quantify leaching losses. Total tracer recovery in plant and soil (N retention) in the control plots was 72% and similar to those observed in temperate forests. The retention decreased to 52% in the N‐plots. Soil was the dominant sink, retaining 37% and 28% of the labeled N input in the control and N‐plots, respectively. Leaching below 20 cm was 50 kg N ha?1 yr?1 in the control plots and was close to the N input (51 kg N ha?1 yr?1), indicating N saturation of the top soil. Nitrogen addition increased N leaching to 73 kg N ha?1 yr?1. However, of these only 7 and 23 kg N ha?1 yr?1 in the control and N‐plots, respectively, originated from the labeled N input. Our findings indicate that deposited N, like in temperate forests, is largely incorporated into plant and soil pools in the short term, although the forest is N‐saturated, but high cycling rates may later release the N for leaching and/or gaseous loss. Thus, N cycling rates rather than short‐term N retention represent the main difference between temperate forests and the studied tropical forest.  相似文献
