1. Lipids have more negative delta(13)C values relative to other major biochemical compounds in plant and animal tissues. Although variable lipid content in biological tissues alters results and conclusions of delta(13)C analyses in aquatic food web and migration studies, no standard correction protocol exists. 2. We compared chemical extraction and mathematical correction methods for freshwater and marine fishes and aquatic invertebrates to better understand impacts of correction approaches on carbon (delta(13)C) and nitrogen (delta(15)N) stable isotope data. 3. Fish and aquatic invertebrate tissue delta(13)C values increased significantly following extraction for almost all species and tissue types relative to nonextracted samples. In contrast, delta(15)N was affected for muscle and whole body samples from only a few freshwater and marine species and had a limited effect for the entire data set. 4. Lipid normalization models, using C : N as a proxy for lipid content, predicted lipid-corrected delta(13)C for paired data sets more closely with parameters specific to the tissue type and species to which they were applied. 5. We present species- and tissue-specific models based on bulk C : N as a reliable alternative to chemical extraction corrections. By analysing a subset of samples before and after lipid extraction, models can be applied to the species and tissues of interest that will improve estimates of dietary sources using stable isotopes.  相似文献

The physiological importance of connexin-26 (Cx26) gap junctions in regulating auditory function is indicated by the finding that autosomal recessive DFNB1 deafness is associated with mutations of the Cx26 gene. To investigate the pathogenic role of Cx26 mutation in recessive hearing loss, four putative DFNB1 Cx26 mutants (V84L, V95M, R127H, and R143W) were stably expressed in N2A cells, a communication-deficient cell line. In N2A cells expressing (R127H) Cx26 gap junctions, macroscopic junctional conductance and ability of transferring neurobiotin between transfected cells were greatly reduced. Despite the formation of defective junctional channels, immunoreactivity of (R127H) Cx26 was mainly localized in the cell membrane and prominent in the region of cell-cell contact. Mutant (V84L), (V95M), or (R143W) Cx26 protein formed gap junctions with a junctional conductance similar to that of wild-type Cx26 junctional channels. (V84L), (V95M), or (R143W) Cx26 gap junctions also permitted neurobiotin transfer between pairs of transfected N2A cells. The present study suggests that (R127H) mutation associated with hereditary sensorineural deafness results in the formation of defective Cx26 gap junctions, which may lead to the malfunction of cochlear gap junctions and hearing loss. Further studies are required to determine the exact mechanism by which mutant (V84L), (V95M), and (R143W) Cx26 proteins, which are capable of forming functional homotypic junctional channels in N2A cells, cause the cochlear dysfunction and sensorineural deafness.  相似文献

Summary The occurrence of tight junctions between Sertoli cells, providing the structural basis of the blood-testis barrier, has been studied using hypertonic fixative and lanthanum tracer in the testes of seven species of vertebrates having different testicular organization. In all cases inter-Sertoli tight junctions, establishing an effective barrier, appear either when meiosis is complete (teleosts and amphibians, both with cystic testes) or immediately after the onset of meiosis (reptiles and birds, both having testes consisting of seminiferous tubules). In the cystic testes, tight junctions are regularly associated with desmosomes, whereas in testes with seminiferous tubules, cisternae of the endoplasmic reticulum are present beneath the junctions (subsurface cisternae). The avian testes examined have, in addition, septate-like junctions between the Sertoli cells but before the tight junctions.Dedicated to Prof. Dr. H. Residence time distribution studies of gas through a rotating drum bioreactor for solid-state fermentation were performed using carbon monoxide as a tracer gas. The exit concentration as a function of time differed considerably from profiles expected for plug flow, plug flow with axial dispersion, and continuous stirred tank reactor (CSTR) models. The data were then fitted by least-squares analysis to mathematical models describing a central plug flow region surrounded by either one dead region (a three-parameter model) or two dead regions (a five-parameter model). Model parameters were the dispersion coefficient in the central plug flow region, the volumes of the dead regions, and the exchange rates between the different regions. The superficial velocity of the gas through the reactor has a large effect on parameter values. Increased superficial velocity tends to decrease dead region volumes, interregion transfer rates, and axial dispersion. The significant deviation from CSTR, plug flow, and plug flow with axial dispersion of the residence time distribution of gas within small-scale reactors can lead to underestimation of the calculation of mass and heat transfer coefficients and hence has implications for reactor design and scale-up.  相似文献

The term 'RNA silencing' describes a process that results in the specific degradation of an RNA target. In plants, silenced tissues can initiate the spreading of the process into non-silenced regions by a mobile signal that can be transmitted over long distances. In the present work, we made use of a modified grafting approach to elucidate the driving force behind long-distance transport of the silencing signal. We made reciprocal grafts of two GFP-transgenic Nicotiana benthamiana lines, the non-silenced line 16c (sensor) and the silenced line 6.4 (inducer). We show that the direction of systemic spread of silencing from inducer to sensor can be manipulated by altering sink/source relations in the plant. Using radioactive phosphate as a phloem tracer, we demonstrated that plants that transmitted silencing from silenced scion to non-silenced rootstock had developed a persisting phloem flow from scion to rootstock. These data provide experimental proof of what has been hypothesized so far, that the silencing signal travels via phloem from source to sink. We present here evidence that the appearance of systemic silencing is not an accidental stochastic process, but can be predicted on the basis of the direction of phloem flow.  相似文献

Protein production of mammalian-cell culture is limited due to accumulation of waste products such as lactate, CO(2), and ammonia. In this study, the intracellular fluxes of hybridoma cells are measured to determine the amount by which various metabolic pathways contribute to the secretion of waste products derived from glucose. Continuously cultured hybridoma cells are grown in medium containing either 1-(13)C-, 2-(13)C-, or 6-(13)C-glucose. The uptake and production rates of amino acids, glucose, ammonia, O(2), and CO(2) as well as the cellular composition are measured. In addition, the (13)C distribution of the lactate produced and alanine produced by the hybridomas is determined by (1)H-NMR spectroscopy, and the (13)CO(2)/(12)CO(2) ratio is measured by on-line mass spectrometry. These data are used to calculate the intracellular fluxes of the glycolysis, the pentose phosphate pathway, the TCA cycle, and fluxes involved in amino acid metabolism. It is shown that: (i) approximately 20% of the glucose consumed is channeled through the pentose shunt; (ii) the glycolysis pathway contributes the most to lactate production, and most of the CO(2) is produced by the TCA cycle; (iii) the pyruvate-carboxylase flux is negligibly small; and (iv) the malic-enzyme flux is estimated to be 10% of the glucose uptake rate. Based on these flux data suggestions are made to engineer a more efficient glucose metabolism in mammalian cells.  相似文献

Streams are important sites of nutrient transport and transformation in the landscape but little is known about the way in which individual taxa or individual habitats (riffles and pools) influence nutrient dynamics within stream reaches. We used 5-week additions of a stable isotope (15NH4Cl) tracer to investigate nitrogen dynamics in pools and riffles of two New Zealand streams, one with native fish (Galaxias depressiceps) and the other with invasive brown trout (Salmo trutta). In New Zealand, brown trout initiate a trophic cascade leading to increased algal biomass that we predicted would lead to higher N uptake and retention. Uptake of NO3, but not ammonium, was greater in the trout stream. Rather than causing a large increase in N demand, trout may induce a reallocation of N uptake and retention among food web compartments in different habitats. The largest differences between streams were apparent in riffles, where most uptake and retention of N occurred. In the trout stream, uptake rate by epilithon in riffles was more than six times greater than uptake rates of any other compartment. In the Galaxias stream, several compartments in both habitats had similar uptake rates. Epilithon also accounted for a larger percentage of the 15N retained in the study reach in the trout stream (51%) than the Galaxias stream (34%). Our results show that an individual predatory taxon (in our case an invader) can influence N dynamics in streams but that the magnitude and location of the impact depend on a range of abiotic and biotic factors involved in N dynamics in streams.  相似文献

在桂西北喀斯特洼地,用地统计学方法研究了旱季初期湿润和干旱条件下表层(0~5和5~10 cm)土壤水分的空间结构及其分布特征.结果表明,表层土壤水分存在明显的空间异质性和各向异性,呈现差异显著的斑块状分布格局.湿润条件下,土壤水分具有中等和较强的空间相关性,变程分别为33.15 m和15.75 m,其中0~5 cm层具有明显的趋势效应;干旱条件下,土壤水分则呈现出强烈的空间相关性,而且相似斑块的空间尺度有所减小,变程最小仅为8.22m;在平均含水量较低时(干旱条件)其变异程度较大,实际应用中应根据平均含水量水平采取不同的取样设计.实验区表层土壤水分空间变异及其分布格局的显著差异,主要是受地貌、平均含水量(降水)和地形等因素的影响.  相似文献

Soil fertilization is a common practice in modern agriculture, undertaken to prevent nutrient deficiency in crops. However, fertilization is costly and causes environmental pollution. The cultivation of plants that tolerate low nutrient supplies may circumvent this problem. Here, we report the generation of Arabidopsis thaliana plants that tolerate boron (B)-deficient conditions due to the overexpression of BOR1, an efflux B transporter that is required for efficient xylem loading of B. In several independently generated transgenic plants expressing BOR1 or BOR1-GFP under the control of the cauliflower mosaic virus 35S RNA promoter, root-to-shoot translocation of B was enhanced and shoot growth was greater under B-limiting conditions compared with wild-type plants. In addition, the transgenic plants showed increased translocation of B, especially to the shoot apex, and set seed normally under B-limiting conditions, under which wild-type plants failed to set seed. This study therefore reports plants that show improved seed yields compared with wild-type under nutrient-deficient conditions as a result of increased production of an essential mineral nutrient transporter.  相似文献

The seasonal population trends of cattle nematodes in tracer calves are described from 1981 to 1987. Successive worm-free calves were grazed with growing beef cattle for 20–30 days at 6 week intervals and then slaughtered for nematode counts 2 weeks after their removal from pasture. Ostertagia, Cooperia, Trichostrongylus and Haemonchus were the main genera recovered. O.ostertagi was the most often found and acquisition of inhibition-prone larvae began in late winter and reached a peak in spring, while maximum larval availability was in autumn. The seasonal inhibition and larval availability pattern for T. axei was similar to that of Ostertagia. Cooperia showed greatest inhibition during winter with maximum larval availability in autumn and spring. Haemonchus was more prevalent during summer to early autumn and no inhibition was observed. It was concluded that infective larval availability for tracer calves was highest during autumn, and most reduced in summer.
