A benign-by-design method for the determination of boron (B) in microsamples of biological tissues was developed. This is a simple, automated, microdigestion method. Use of reagents and generation of waste are minimized, and the use of toxic/hazardous reagents is eliminated as compared to currently available B methodology. Microsamples are accommodated by the method; 100–400 mg sam ples were used in this study. B is determined by inductively coupled plasma atomic emission spectrometry (ICPAES) at 249.678 nm. The instrument detection limit for B is 0.01 Μg/mL. Interference studies have been investigated for 21 common elements. Over 250 analyses of standard reference materials were analyzed during the study dura tion. Recoveries for a series of biological tissues, both plant and ani mal, ranged from 82–104%.  相似文献

Five groups of individually housed albino rats (n=7, initial average weight=48 g) were fed diets based on egg albumen and cornstarch (basal diet 8.2 g Ca, 6.0 g P, 0.7 g Mg, 225 mg Zn, 150 mg Fe, 60 mg Mn, 8 mg Cu, and 5 mg Cd) over a 4-wk period. Group I (control) was fed the basal diet free of phytic acid (PA). In groups II, III, IV, and V, cornstarch was replaced by 3.5, 7.0, 10.5, and 14.0 g sodium phytate/kg diet, respectively. Daily gain, feed efficiency, Zn status (Zn in plasma, femur, testes, liver and kidneys, activity of the plasma alkaline phosphatase) and apparent absorption of Zn, Fe, Cu, and Mn remained unchanged by the different dietary treatments. PA decreased apparent Mg absorption significantly and apparent absorption of Ca in tendency. Increasing the amount of phytate caused a corresponding enhancement of amount of the digestible P. Cd accumulation in the liver was not significantly altered, and kidney Cd accumulation slightly increased owing to PA. In conclusion, it was shown that under conditions of high dietary Zn, PA had only little effect on the carryover of Cd in growing rats.  相似文献

PROJECT: The results of our recent studies prove the stimulating effect of aqueous peat extract (APE) on the spontaneous contractile activity (SCA) of the smooth musculature. Only substances with a molecular weight of <3,000 Dalton are able to evoke any effect. As we know from the corresponding literature, trace elements as for instance copper, manganese, lead and cadmium do also influence the SCA even when they appear in low concentrations (micromol-range). The purpose of this study therefore is to examine the influence of the trace-elements on the inspected stimulating effects which aqueous peat extract has on the SCA. PROCEDURE: During in-vitro experiments with smooth muscles in organbaths, it has been examined--under application of a standardized method--if trace-elements are the cause for the stimulating effect of aqueous peat extract on the SCA of the smooth musculature. RESULTS: The results have shown that--independent from their concentration within the peat - the trace-elements do not influence the SCA of the smooth muscles. CONCLUSION: The results can be explained by the chelating capacity of the peat-components, that leads to the absorption of the trace-elements. Additionally we can conclude that organic substances are the exclusive reason for the described effects.  相似文献

Catalase (CAT) is an enzyme that is involved in antioxidant defense, cell growth, and is possibly associated with tumoral processes. In this paper, the results of experiments designed to determine the influence of metallic carcinogens such as nickel (Ni), lead (Pb), mercury (Hg), and cadmium (Cd), on CAT activity are reported. CAT activity was measured in erythrocytes from three groups: a group of colon cancer patients, a group of gastric cancer patients before clinical treatment, and a control group of healthy blood donors. Concentrations of this enzyme are significantly higher than controls in the colon cancer group, but lower in gastric neoplasia. By generating highly reactive oxygenated species, Ni, Pb, Hg, and Cd alter catalase activity. Solutions of Ni, Cd, and Pb at 0.2 mM concentrations inhibit CAT activity in colon cancer, but increase it in gastric neoplasia. Hg activates CAT in colon cancer, and causes a slightly increased activity in gastric cancer. No complete deactivation of the enzyme was observed.  相似文献

Accurate and precise analytical data of the concentrations of bioanalytes in bioclinical studies are of fundamental importance. Quality assurance procedures should always be performed to check the overall analytical work. This can be conveniently performed if appropriate standard reference materials with known concentrations of the analyte object of study are available. This paper underlines the key points related to the production and use of biological standard materials for trace element analysis. In particular, the present situation in the field of trace element determination in human biological fluids and the related problems are illustrated. The considerations given in this work may contribute to the preparation of the new biomarker standard materials.  相似文献

Substitution of nitrilotriacetic acid (NTA) for polyphosphates in detergents has brought questions concerning its potential toxicity and impact on trace metal distribution in the environment. A calculation based upon metal ligand equilibria, known environmental concentrations of NTA following extensive detergent usage, and the presence of competitive metal-binding ligands and trace elements demonstrates that NTA will be present almost completely as the calcium and magnesium chelates. An analogous estimate of the speciation of NTA in various toxicity studies demonstrates that the onset of chronic toxicity in feeding studies is coincident with the presence of significant concentrations of “free” NTA in the gastrointestinal tract. Massive doses of NTA over long periods of time cause reproducible renal tumors in rats, but dosages of 7500 ppm administered indefinitely are without measurable effect.  相似文献

Abstract

In this study, the fractionation and ecotoxic potential of Cr, Ni, Cu and Pb in water and sediments of the river Yamuna (Delhi stretch) is presented. The river water is used for various purposes such as irrigation, industrial, domestic and drinking. The investigations suggest that the availability of metals in water follows the sequence Ni>Cr>Pb>Cu whereas a different pattern (Pb>Cu>Ni>Cr) is observed in sediments. Statistically significant spatial variations are indicated by two-way ANOVA in different geochemical forms of the metals and physicochemical parameters of water and sediments of the river Yamuna. Principal component analysis (PCA) suggests a similar source for all four metals probably from the sewage from municipal drains. PCA also indicates some independent sources of Cu and Pb. The hot spots were identified using hierarchical cluster analysis considering bioavailable forms of the metals in water and sediments as objects and the sampling sites as variables. Risk assessment code analysis indicates that metals generally pose medium to high risk at different sites. However, at a few locations either Ni or Pb cross the level of very high ecological risk.  相似文献

Sample preparation is the first crucial step in the speciation analysis, bioavailability and risk assessment of trace metals in plant samples such as herb and vegetables. Two bionic technologies titled ‘in vitro digestion’ and ‘extraction with biomembrane’ were developed for pre‐treatment of herbal decoction. The decoctions of Aconiteum carmichaeli and Paeonia lactiflora were digested at body temperature, at the acidity of the stomach or intestine and with inorganic and organic materials (digestive enzymes were included for whole‐bionic and excluded for semi‐bionic) found in the stomach or intestine. Being similar to the biomembrane between the gastrointestinal tract and blood vessels, monolayer liposome was used as a biomembrane model. Affinity‐monolayer liposome metals and water‐soluble metals were used for speciation analysis and bioavailability assessment of copper and zinc in herbal decoction. In the decoction of Aconiteum carmichaeli and Paeonia lactiflora, Zn was mainly absorbed in the intestine and Cu was mainly absorbed by both stomach and intestine. Abstract

In many arid and semi-arid regions, farmers are often obligated to informally use raw wastewater for irrigating their crops. The impacts of wastewater irrigation on soil, crops, and human health were investigated, regarding trace metals and bacteria. Cr, Cu, Fe, Ni, and Zn were detected in wastewater. Cr, Cu, and Zn accumulated in soil and crops in the order rocket?>?clover?>?cabbage. The Health Risk Index reported risk from Cr and Zn in rocket. Fecal coliforms in wastewater and crops were detected along with Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa. The frequency (%) was 86.7% (cabbage), 66.7% (rocket), and 43.3% (clover). The multiple antibiotic resistance index (0.503) rendered crops high risk source for contamination. The comprehensive pollution index classified wastewater severely polluted (≥2.01). Conclusions deduced crops alternative reservoirs for trace metals and human pathogens. Recommendations included implementation of low cost treatment methods, holding irrigation 5–15?days before harvest, assuming citric and acetic acids reducing solutions for trace metals, and suggested ofloxacin, amoxycillin/clavulanate, and amikacin antibiotics against reported bacteria.  相似文献
