Fermentative production of squalene under anaerobic conditions using commercially available compressed baker's yeast (Saccharomyces cerevisiae), and a strain of Torulaspora delbrueckii isolated from molasses was studied. Yield of squalene from S. cerevisiae and T. delbrueckii were found to be 41.16 and 237.25 g g–1 respectively, dry weight of yeast cells. Isolation and purification of squalene from the lipid extracts obtained by cell lysis of either strain were achieved chromatographically. The purified squalene was characterized spectroscopically against an authentic standard.  相似文献

Bacillus thuringiensis (Bt) is a grampositive, spore forming bacterium, which is principally distinguishedfrom other bacilli by the production of large, insecticidal,protein crystals (Insecticidal Crystal Proteins, or ICPs). Theseproteins are usually thought to act only on the actively feedinglarvae of susceptible species by a mechanism which involvesconsumption and proteolytic processing of the protein followed bybinding to, and lysis of, midgut epithelial cells. However, few authorshave reported Bt toxicity to adult insects. In the followingpaper, we expand on previous reports of toxicity to adult insects andpresent data which demonstrate that: (1) proteolytically activated ICPssignificantly reduce the lifespans of adult Heliothis virescensand Spodoptera exigua at concentrations of 500 g/ml, butnot 167 or 25 g/ml, (2) individual activated ICPs are differentiallytoxic to adult H. virescens and S. exigua, and (3)adult S. exigua are sensitive to Cry1C protoxin at aconcentration of 1 mg/ml.Deceased  相似文献

Ten distinct monoclonal antibodies (MAbs) against alpha toxin of Clostridium perfringens were produced by the fusion of SP2/O with spleen cells of mice immunized with alpha toxoid, and alpha toxin mixed with or without ethylenediamine-tetraacetate (EDTA). The antibody activity was evaluated by antigen-binding activity in an enzyme linked immunosorbent assay (ELISA), by phospholipase C (PLC)-neutralizing activity using both egg yolk lecithin and p-nitrophenylphosphoryl-choline (PNPPC) hydrolysis reactions and by anti-lethal activity in mice. Since the toxin-neutralizing activities of each MAb were not parallel, it has been suggested that the three biological activities may not be located in the same site in the toxin molecule. This report also describes the development of a simple purification of the toxin by affinity chromatography and a sensitive immunoassay for quantitation of the toxin using the monoclonal antibody.  相似文献

为获得一定规模的小麦单倍体植株以构建小麦DH(Double Haploid)群体,采用小麦(Triticum aestivum,2n=42)与玉米(Zeamays,2n=20)远缘杂交诱导小麦单倍体胚,经胚拯救产生单倍体植株,研究了1/2MS培养基中幼胚取材时期、幼胚大小、4℃处理时间、暗处理时间对单倍体胚培养再生成苗的影响,结果表明:授粉后12天~16天取材的幼胚经胚拯救后成苗率无明显差异;0.5mm~1.0mm大小的幼胚成苗率显著高于0mm~0.5mm和1.0mm~1.5mm大小的幼胚成苗率;1天~3天短期4℃处理对胚萌发具有一定促进作用,但处理3天后,出愈率和成苗率降低;胚培养过程中12天左右的24h暗处理能有效提高成苗率。  相似文献

以大量的室内模拟培养实验,以内蒙古温带草甸草原土壤为研究对象,利用AIM乙炔抑制法,模拟野外条件对原样土壤样品进行N2O产生过程进行研究。实验结果表明:内蒙古温带草甸草原土壤N2O产生过程以硝化作用为主。其中异养硝化作用起主导作用,自养硝化潜势和反硝化潜势在草原植物不同生长季节变化不同,总体上异养硝化潜势>自养硝化潜势>反硝化潜势。由于自养硝化作用在不同季节的发生,使得草甸草原土壤N2O的产生潜势也高、低起伏变化。从而揭示了内蒙古温带草原土壤以异养硝化作用过程为主产生N2O和N2O排放通量较低的微生物学机理。  相似文献

Changes in the content of cyclic heptapeptide hepatotoxins called microcystins were investigated during batch culture of two Microcystis species using high performance liquid chromatography. After adsorption to ODS-silica gel cartridges and elution with methanol, the toxins were analyzed and quantified by HPLC. 35 μg per 100 mg dry cells of microcystin-RR, 34 μg of -YR and 43 μg of -LR were present at the beginning of the exponential growth phase of M. viridis. Microcystin-RR increased markedly towards the end of the exponential phase with the maximum content of 112 μg per 100 mg cells was measured at the late stage of the exponential phase. A remarkable increase of microcystin-YR from 130 μg per 100 mg cells to 1020 μg was observed during the exponential phase of a highly toxic strain of M. aeruginosa. However no clear differences were found in the pattern of change among the three toxins during the growth course.  相似文献

Fungal chitinases are hydrolytic enzymes responsible for degradation of chitin. Chitinases are involved in several aspects of fungal biology, including cell wall remodelling during hyphal growth, conidial germination, autolysis, mycoparasitism and nutrient acquisition. They are divided into three distinct phylogenetic groups; A, B and C. Chitinases from the C group show structural similarities with the killer toxin zymocin produced by the yeast Kluyveromyces lactis and it is speculated that they have a similar function in filamentous ascomycetes, by facilitating penetration of toxins into cells of competing individuals. Genome analyses show that certain fungal species with a mycoparasitic lifestyle contain high numbers of killer toxin-like chitinases, compared with specialized saprotrophs and plant pathogens. Recent developments within this research field have revealed considerable variation in the modular structure and regulation of killer toxin-like chitinases, suggesting more diverse roles than merely fungal-fungal interactions. In this review, we summarize the current knowledge about this intriguing class of chitinases, including their modular structure, evolution, gene regulation, and functional analyses in mycoparasitic as well as in saprotrophic species. Two types of amphoteric glycosphingolipid found in the earthworm Pheretima hilgendorfi, PC(-->6)-beta-d-Galp-(1-->6)-beta-d-Galp-(1-->1)Cer (1) and PC(-->6)-beta-d-Galp-(1-->6)-beta-d-Galp-(1-->6)-beta-d-Galp-(1-->1)Cer (2), and their derivatives (4, 5) were synthesized. These were examined for their ability to enhance production of interleukin-8 (IL-8), a potent inflammatory cytokine involved in neutrophil chemotaxis, in a TNFalpha-stimulated granulocytic HL-60 cells. 1. Side‐arms connected to the main stem of the river are key areas for biogeochemical cycling in fluvial landscapes, exhibiting high rates of carbon processing. 2. This work focused on quantifying autochthonous and allochthonous carbon pools and, thereby, on comparing transport and transformation processes in a restored side‐arm system of the River Danube (Regelsbrunn). We established a carbon budget and quantified carbon processing from March to September 2003. In addition, data from previous studies during 1997 to 1999 were assessed. 3. Gross primary production (GPP) and community respiration were estimated by diel oxygen time curves and an oxygen mass balance. Plankton primary production was determined to estimate its contribution to GPP under different hydrological conditions. 4. Based on the degree of connectivity, three hydrological phases were differentiated. Most of the organic matter, dominated by allochthonous carbon, was transported in the main channel and through the side‐arm during floods, while at intermediate and low flows (and thus connectivity), transformation processes became more important and autochthonous carbon dominated the carbon pool. The side‐arm system functioned as a sink for particulate matter [total suspended solids and particulate organic carbon (POC)] and a source of dissolved organic carbon (DOC) and chlorophyll‐a. 5. Autochthonous primary production of 4.2 t C day?1 in the side‐arm was equivalent to about 20% of the allochthonous inputs of 20 t C day?1 (POC and DOC) entering the area at mean flow (1% of the discharge of the main channel). Pelagic photosynthesis was generally high at mean flow (1.3–3.8 g C m?2 day?1), and contributed up to 90% of system productivity. During long stagnant periods at low discharge, the side‐arm was controlled by biological processes and a shift from planktonic to benthic activity occurred (benthic primary production of 0.4–14 g C m?2 day?1). 6. The transformation of the organic matter that passes through the side‐arm under different hydrological conditions, points to the importance of these subsystems in contributing autochthonous carbon to the food web of the main channel.  相似文献

A Chlamydomonas reinhardtii chloroplast expression vector, pACTBVP1, containing the fusion of the foot and mouth disease virus (FMDV) VP1 gene and the cholera toxin B subunit (CTB) gene was constructed and transfered to the chloroplast genome of C. reinhardtii by the biolistic method. The transformants were identified by PCR, Southern blot, Western blot and ELISA assays after selection on resistant medium and incubation in the dark. The CTBVP1 fusion protein was expressed in C. reinhardtii chloroplast and accounted for up to 3% of the total soluble protein. The fusion protein also retained both GM1-ganglioside binding affinity and antigenicity of the FMDV VP1 and CTB proteins. These experimental results support the possibility of using transgenic chloroplasts of green alga as a mucosal vaccine source.  相似文献
