In extracts of flax seedlings 4 days after imbibition, isocitrate lyase activity is unstable in comparison to that in extracts from 2.5-day seedlings or to malate syntheses analysed at several stages of development. This instability in extracts of 4-day seedlings is especially pronounced when a large number of seedlings is homogenized per unit volume of Tris-Mg2+-EDTA-dithioerythritol buffer. However, isocitrate lyase can be stabilized when the resultant homogenate is diluted soon after seedling breakage. The pronounced instability in more concentrated extracts is not due to inadequate buffering by the homogenization medium, nor can it be due to polyphenols because added polyvinylpyrrolidone has no effect. Mixing of a heated supernatant from concentrated extract with dilute unheated extract yields the units of stable isocitrate lyase expected in the dilute extract, ruling out stoichiometric inactivation by a heat-stable component. The pronounced instability is attributed to the action of proteinases. A theoretical model assuming a decay process that is first order in isocitrate lyase and first-order in one or more proteinases is in reasonable agreement with the results. Malate synthase and NADP+-isocitrate dehydrogenase are much more stable in concentrated extracts prepared from 4-day flax seedlings. Isocitrate lyase is stable in concentrated extracts of 5-day watermelon seedlings, which is a developmental stage analogous to that for 4-day flax seedlings. 相似文献

The in vivo amounts of UDPG, UTP, UDP and UMP, metabolites known to influence the activity of sucrose phosphate synthase (SPS) and sucrose synthase (SS), were measured throughout 5 hr incubations of scutellum slices in fructose or water, i.e. under conditions of sucrose synthesis or breakdown. Cytosolic concentrations were estimated assuming that these metabolites were confined to the cytosol. Within the estimated in vivo concentration ranges, UDPG, UTP and UDP had little effect on the in vitro SS activity, but glucose (100 mM) inhibited SS in the synthesis direction by 63–70% and in the breakdown direction by 86–93%. Glucose inhibition of SS was considerably less when saturating levels of substrates were used. Sucrose did not inhibit SS. It is concluded that during germination the glucose produced from starch breakdown in the maize endosperm enters the scutellum and inhibits SS, preventing a futile cycle and limiting SS participation in sucrose synthesis. 相似文献

Two species of jute plants Corchorus capsularis L. (cv. JRC 212) and C. olitorius L. (cv. JRO 632) were subjected to water stress for 2 and 4 days by withholding water. The relative water content (RWC) decreased in both plants under water stress but to a greater extent in C. olitorius . The C. olitorius seedlings also showed greater membrane injury than C. capsularis seedlings under water stress as was evident from injury index data. Water stress increased glycolate oxidase (EC 1.1.3.1.) activity more in C. olitorius than in C. capsularis . The activity of superoxide dismutase (SOD, EC 1.15.1.1.) and catalase (EC 1.11.1.6.) decreased under water stress and their decrease was higher in C. olitorius than in C capsularis . The level of hydrogen peroxide and lipid peroxidation also increased in both plants under water stress and the increase was higher in C. olitorius than in C. capsularis seedlings. Under comparable external water stress, C. capsularis seedlings showed lower membrane damage, lower H2 O2 accumulation and lower lipid peroxidation than C. olilorius which may be taken as indicative of higher water stress tolerance capacity of the former. 相似文献

The major sesquiterpenes in the foliage of Dacrydium cupressinum are α-longipinene, longifolene, longibornyl acetate, caryophyllene, caryophyllene oxide, humulene, α- and β-selinene, β- and δ-elemene, aromadendrene and the rare 9βH-caryophyllene. Sesquiterpene levels vary greatly from tree to tree. As this variation is largely independent of environmental factors, genetic control is proposed. Six-day-old tobacco seedlings rapidly incorporated and metabolized exogenously supplied [4-14C]-sitosterol, but none of the radioactivity was recovered from stigmasterol. However, exogenously supplied [2-14C]-mevalonic acid was incorporated into both sitosterol and stigmasterol. The total lipids of Pinus halepensis pollen were separated into individual classes of neutral and polar lipids and the components of each class were identified and determined quantitatively. Free fatty acids, waxes and triacylglycerols were found as the main constituents of neutral lipids and phosphatidylcholine and phosphatidylethanolamine of polar lipids. The role of endogenous ethylene during germination of non-dormant seeds of Amaranthus caudatus L. was investigated. The seeds readily germinated in water and darkness at 24°C. Application of ethylene or of its precursor I-aminocyclopropane-I-carboxylic acid (ACC) slightly increased the rate of germination. Both compounds effectively antagonized osmotic inhibition by polyethyleneglycol. Application of aminoethoxyvinylglycine (AVG) reduced ethylene production by 90% but did not inhibit germination. However, germination was inhibited by 2,5-norbornadiene, a competitive inhibitor of ethylene action. This inhibition was counteracted by ethylene, ethephon or ACC and enforced by AVG. It is concluded that the action of endogenous ethylene is an indispensable factor during germination of non-dormant seeds of A. caudatus. Ethylene action is required from the start of imbibition on. In water, low levels of endogenous ethylene are sufficient for this action. PEG increased the ethylene requirement considerably. 相似文献

Analysis of total nitrogen, chlorophyll content, ribulose-1,5-bisphosphate carboxylase/oxygenase activity and net photosynthesis rate was carried out on the leaves that support the developing pods in pigeon pea [ Cajanus cajan (L.) Millsp. cv. Prabhat] at several stages during pod filling. A continuous loss in all the above-mentioned parameters was observed during the course of pod development. When no pods were allowed to develop by continuous flower removal treatment, there was a considerable delay in loss of all these metabolic parameters. Excision of pods after their mid-development resulted not only in no further loss, but also in a significant recovery both of total nitrogen and of other investigated characteristics. 相似文献
