Apolipoprotein F (ApoF) modulates lipoprotein metabolism by selectively inhibiting cholesteryl ester transfer protein activity on LDL. This ApoF activity requires that it is bound to LDL. How hyperlipidemia alters total plasma ApoF and its binding to LDL are poorly understood. In this study, total plasma ApoF and LDL-bound ApoF were quantified by ELISA (n = 200). Plasma ApoF was increased 31% in hypercholesterolemic plasma but decreased 20% in hypertriglyceridemia. However, in donors with combined hypercholesterolemia and hypertriglyceridemia, the elevated triglyceride ameliorated the rise in ApoF caused by hypercholesterolemia alone. Compared with normolipidemic LDL, hypercholesterolemic LDL contained ~2-fold more ApoF per LDL particle, whereas ApoF bound to LDL in hypertriglyceridemia plasma was <20% of control. To understand the basis for altered association of ApoF with hyperlipidemic LDL, the physiochemical properties of LDL were modified in vitro by cholesteryl ester transfer protein ± LCAT activities. The time-dependent change in LDL lipid composition, proteome, core and surface lipid packing, LDL surface charge, and LDL size caused by these factors were compared with the ApoF binding capacity of these LDLs. Only LDL particle size correlated with ApoF binding capacity. This positive association between LDL size and ApoF content was confirmed in hyperlipidemic plasmas. Similarly, when in vitro produced and enlarged LDLs with elevated ApoF binding capacity were incubated with LPL to reduce their size, ApoF binding was reduced by 90%. Thus, plasma ApoF levels and the activation status of this ApoF are differentially altered by hypercholesterolemia and hypertriglyceridemia. LDL size is a key determinate of ApoF binding and activation.  相似文献

【目的】通过对极端环境耐受的耐辐射奇球菌Deinococcus radiodurans R1全基因组进行序列比对分析,获得具有铁储备蛋白Ferritin类似功能基序的未知功能蛋白DRA0258,采用分子生物学技术对该蛋白的功能和性质进行了验证和分析。【方法】首先对DRA0258进行克隆表达和纯化,并经络合物显色法测定蛋白上铁结合含量;通过三段连接敲除法构建dra0258突变株,检测突变株在双氧水协迫下的生存率、总抗氧化活性及过氧化氢酶活性;利用实时定量PCR检测突变株内抗氧化酶类及铁转运相关性调控蛋白的基因转录水平。【结果】经体内外蛋白铁含量检测证实DRA0258具有一定的铁结合能力;双氧水生存率实验表明dra0258的缺失导致细胞的抗氧化能力显著下降;过氧化氢酶活性、总抗氧化活性检测及抗氧化酶类的基因转录水平检测证实dra0258基因的缺失导致细胞内一些抗氧化基因转录水平下调,细胞的抗氧化应激系统受到损伤,并影响了一些铁调控网络蛋白的基因转录水平。【结论】本研究证实DRA0258是一种铁结合蛋白,该编码基因的缺失影响胞内铁转运系统并使细胞抗氧化能力下调。  相似文献

摘要 目的:探讨基于"经筋理论"针刀治疗对早中期膝骨关节炎(KOA)患者骨代谢指标和血清金属蛋白酶抑制物-1(TIMP-1)、基质金属蛋白酶(MMP)-3、MMP-13的影响。方法:根据随机数字表法,将2021年1月至2023年1月期间就诊于新疆医科大学附属第一医院的120例早中期KOA患者分为对照组(n=60,常规治疗)和研究组(n=60,对照组的基础上接受"经筋理论"针刀治疗)。对比两组疗效、量表评分[疼痛视觉模拟评分(VAS)、西安大略和麦克马斯特大学骨关节炎调查表(WOMAC)]、骨代谢指标[抗酒石酸盐酸性磷酸酶异构体(TRACP-5b)、骨特异性碱性磷酸酶(BALP)、骨钙素(BGP)]和血清TIMP-1、MMP-3、MMP-13。结果:与对照组相比,研究组的临床总有效率更高(P<0.05)。与对照组相比,研究组治疗后WOMAC、VAS评分和血清MMP-3、MMP-13、TRACP-5b水平更低,TIMP-1、BALP、BGP水平更高(P<0.05)。结论:基于"经筋理论"针刀治疗早中期KOA患者,可有效减轻疼痛症状,提高临床治疗效果,可能与改善骨代谢指标和血清TIMP-1、MMP-3、MMP-13水平有关。  相似文献

摘要 目的:探讨不同剂量艾司氯胺酮对腹腔镜全子宫切除术患者术后疼痛和情绪状态的影响。方法:按照随机数表法,选择2022年1月至2023年4月我院行腹腔镜全子宫切除术患者120例分为对照组(A组,n=40)、艾司氯胺酮0.25 mg/kg组(B组,n=40)、艾司氯胺酮0.5 mg/kg组(C组,n=40)。比较三组组围术期指标、术后1、4、8、24 h和48 h的疼痛评定数字量表(NRS)评分及术前1天(PRE1)、术后第1天(POD1)、术后第2天(POD2)的焦虑自评量表(SAS)评分、抑郁自评量表(SDS)评分、血流动力学指标及不良反应。结果:B组和C组术后24 h舒芬太尼镇痛药物用量、补救镇痛例数小于A组,A组、B组苏醒时间、拔管时间短于C组(P<0.05)。B组、C组术后1 h的NRS评分低于A组(P<0.05),B组术后4 h、8 h、24 h的NRS评分低于A组,术后4 h、8 h的NRS评分低于C组(P<0.05)。与组内PRE1相比,三组POD1的SAS、SDS评分均升高(P<0.05)。B组、C组POD1、POD2的SAS、SDS评分均低于A组,且C组低于B组(P<0.05)。与组内PRE1相比,A组POD2的SAS、SDS评分升高(P<0.05)。与组内PRE1相比,B组POD2的SDS与C组POD2的SAS、SDS均降低(P<0.05)。在T3时刻,B组和C组的平均动脉压(MAP)、心率(HR)低于A组(P<0.05)。B组和C组术后恶心呕吐发生率低于A组(P<0.05);A组和B组术后头晕、苏醒延迟的发生率低于C组(P<0.05);三组术后苏醒期躁动和呼吸抑制的差异无统计学意义(P>0.05)。结论:0.25 mg/kg的艾司氯胺酮对减轻腹腔镜全子宫切除术患者术后疼痛,减少阿片类药物用量,改善负面情绪的临床效果更佳。  相似文献

摘要 目的:观察通滞苏润江胶囊联合塞来昔布对早期膝骨关节炎(KOA)患者血清炎症因子和疼痛介质的影响。方法:选择2021年6月-2022年12月期间石家庄市人民医院收治的早期KOA患者80例,采用随机数字表法将患者分为对照组(塞来昔布,40例)和观察组(通滞苏润江胶囊联合塞来昔布,40例)。对比两组疗效、疼痛视觉模拟评分(VAS)、西安大略和麦克马斯特大学骨关节炎指数(WOMAC)评分、炎症因子和疼痛介质。结果:观察组临床总有效率高于对照组(P<0.05)。两组治疗1个月后VAS、WOMAC评分下降,且观察组低于对照组同时间点(P<0.05)。两组治疗1个月后肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)下降,且观察组低于对照组同时间点(P<0.05)。两组治疗1个月后前列腺素E2(PGE2)、P物质(SP)下降,且观察组低于对照组同时间点(P<0.05)。两组不良反应发生率对比未见统计学差异(P>0.05)。结论:塞来昔布和通滞苏润江胶囊联合治疗早期KOA患者,可缓解疼痛,提高临床治疗效果,改善关节功能,同时还可改善血清炎症因子和疼痛介质水平。  相似文献

The glycans of well characterized, [6-3H]galactose-labelled glycopeptides, GC-4 from bovine IgG1 as well as GP-V-2 and GP-V-5 from α1-acid glycoprotein, were liberated by hydrazinolysis. Molecular weights close to the expected values were observed by gel filtration. Desialated glycans of Semliki Forest virus proteins were likewise liberated by hydrazinolysis and subjected to gel filtration. Metabolically labelled [1-3H]galactose-oligosaccharides of the mixed viral proteins revealed an apparent molecular weight of 1800. The bi-antennary glycan liberated from the reference glycopeptide GC-4 was of 1750 daltons. A mixture of [2-3H]mannose-labelled E1-and E2-proteins of the virus contained L-type glycans of 1800 daltons (formerly called A-type), and M-type glycans of 1200 daltons (formerly called B-type). A fraction of the E3-glycans isolated by affinity chromatography on Concanavalin A-Sepharose showed an average molecular weight of 2150, a value intermediate between the three- and four-antennary glycans liberated from the reference glycopeptides GP-V-5 and GP-V-2. The rest of the E3-glycans were of 1850 daltons, a value close to the bi-antennary GC-4 glycan. We suggest that the comparatively large size of the E3-glycans and the exposed position of E3-proteins on the viral surface may be interrelated.  相似文献

Abstract

Context: The kidney plays a central role in detoxification and excretion of toxic metabolites, and therefore, is susceptible to toxicity by xenobiotics.

Objective: To investigate the protective effect of Rosmarinus officinalis (rosemary) powder and its essential (volatile) oil against diethylnitrosamine (DEN)-induced renal injury in rats.

Materials and methods: Phenolic and flavonoid components were characterised in rosemary powder using HPLC-UV instrument while rosemary essential oil (E.O) was investigated via GC-MS technique. In rat model, rosemary was administrated orally (in diet) for two months. Lipid profile, antioxidant biomarkers, kidney functions and histopathological examinations were assessed.

Results: Hesperidin (4878.88?ppm) and ellagic acid (403.57?ppm) are among the major phenolic and flavonoid constituents in rosemary powder. Camphor (18.36%) and α-pinene (12.74%) represent the main E.O active ingredients. Rats treated with rosemary E.O showed a significant elevation in serum HDL (28.28%) accompanied by a decrease in LDL (115.47%). A significant decrease in serum creatinine and urea was also reported (69.72 and 109.89%, respectively). Moreover, serum glutathione peroxidise (GSH-Px) activity has been significantly increased. Kidney histopathological examinations confirmed the protective effect against DEN-induced abnormalities.

Conclusion: Rosemary (powder/E.O) was able to reduce or even prevent the severity of diethylnitrosamine-induced renal dysfunction.  相似文献

Length weight relationships (LWR) of five marine fishes were estimated from the Bay of Bengal, Bangladesh. Fish samples were collected occasionally from fisher's catch with gill net (mesh size: 3 cm) during November 2019 to October 2020. Fishing was done overnight. Total length (0.1 cm) and body weight (0.01 g) of each individual were measured. We recorded maximum total length for P. maculatus (55.0 cm) and P. parmatus (39.0 cm).  相似文献

红孢囊藻(Rhodosorus sp.)可以积累高含量多糖,在医药保健和生物能源领域具有潜在开发价值。为进一步评估红孢囊藻(Rhodosorus sp.)产业化潜力,研究了一株红孢囊藻SCSIO-45730在盐度不同的4种培养基中生长及总糖和β-葡聚糖积累特性,结果表明:改良的ASW培养基相对于其他培养基(f/2、Conway和Erdschreiber)更有利于红孢囊藻SCSIO-45730生物量和总糖的积累。以ASW培养基为基础培养基,研究该藻盐度适应性,结果显示:该藻可在18‰-58‰盐度范围内正常生长,随着盐度增加,藻细胞体积逐渐增大且出现聚团现象;在盐度为38‰时,获得最大生物量、总糖含量和产率以及β-葡聚糖产率,分别为13.70 g/L、48.52%DW、412.58 mg/L·d和119.70 mg/L·d,β-葡聚糖含量随着盐度增加而降低;该藻在户外平板光生物反应器中总糖和β-葡聚糖单位体积产量分别为0.73 g/L和0.23 g/L。结果表明,红孢囊藻SCSIO-45730具有较广的盐度适应性、高产总糖和β-葡聚糖潜力,是生产生物乙醇和β-葡聚糖的理想原料。  相似文献

This work is concerned with the lubrication analysis of artificial knee joints, which plays an increasing significant role in clinical performance and longevity of components. Time-dependent elastohydrodynamic lubrication analysis for normal total knee replacement is carried out under the cyclic variation in both load and speed representative of normal walking. An equivalent ellipsoid-on-plane model is adopted to represent an actual artificial knee. A full numerical method is developed to simultaneously solve the Reynolds and elasticity equations using the multigrid technique. The elastic deformation is based on the constrained column model. Results show that, under the combined effect of entraining and squeeze-film actions throughout the walking cycle, the predicted central film thickness tends to decrease in the stance phase but keeps a relatively larger value at the swing phase. Furthermore, the geometry of knee joint implant is verified to play an important role under its lubrication condition, and the length of time period is a key point to influence the lubrication performance of joint components.  相似文献
