The pre- and postdispersal spatial patterns of the germinable seed bank of Festuca pallescens were evaluated in semiarid grasslands of Patagonia disturbed by sheep grazing. Before dispersal, the seed bank showed spatial heterogeneity and was significantly smaller than the postdispersal seed bank. The postdispersal seed bank was uniformly distributed in patches of bare soil. The size of the germinable seed bank varied with the topographic condition of sites. On slopes, bunches exhibited more panicles and the germinable seed bank was larger after dispersal and smaller before dispersal as compared with upland sites. Losses of the germinable seed bank during autumn and spring are partially accounted for by germination rates. Predominant westerly winds influenced the patterns of seed dispersal, but other agents such as predation or sheet wash may also be active, especially on slopes. The low persistence of the seed bank of F. pallescens, particularly at canopy gaps, indicates that this may constitute a primary control of the regeneration of this species in semiarid grasslands of Patagonia. Management of these grasslands for restoration should accordingly include practices oriented to an adequate replenishment of the germinable seed bank at these places.  相似文献

The surface topography of the rat egg was examined during fertilization in vitro and in vivo. Using phase optics, 348 in vitro fertilized and 50 in vivo fertilized eggs were continuously monitored throughout the 7-hour period of sperm incorporation. A myriad of different surface configurations were seen, with each egg exhibiting one or more of the following changes. A small number of eggs (4–6%) formed surface elevations over the sperm head after its detachment from the flagellum, 15–30 min after sperm-egg fusion; 1 to 1.5 hr after fusion, 40–50% of the eggs produced the so-called incorporation cone, a prominent surface elevation over the decondensing sperm nucleus. The vast majority of eggs (74–82%) formed surface elevations over the proximal tip of the flagellum 2–3 hr after sperm-egg fusion. These had no association with the decondensing sperm nucleus. A few eggs (11–12%) exhibited multiple protrusions that were distributed randomly about the egg surface, whereas 14–20% did not manifest any surface elevations and remained spherical throughout the sperm incorporation period. Regardless of the type of surface change, all of the eggs resumed a spherical shape by the time sperm incorporation was complete. These observations are in contrast to the conclusions by previous authors that formation of the so-called incorporation cone over the decondensing sperm nucleus is a ubiquitous event.  相似文献

A cleavable cross-linking reagent, dithiobis(succinimidyl propionate), DSP, was used to study the topography of the proteins in the endoplasmic reticulum membrane of rat liver. Reaction of untreated (control), phenobarbital- or 3-methylcholanthrene-induced microsomes with 0.5 mM DSP for 30 min at 0°C resulted in the cross-linking of a protein with a molecular weight of about 52 000 to form an apparent dimer. In phenobarbital microsomes, a smaller amount of a 52 000-dalton protein also appeared in a dimer in the absence of DSP if N-ethylmaleimide was not included during homogenization. In phenobarbital and 3-methylcholanthrene microsomes, a 48 000-dalton protein was cross-linked by DSP to a protein of about 57 000. In all three types of microsomes, a protein with an MI of about 52 000 was also cross-linked to a protein of about 79 000. In phenobarbital and control microsomes, cross-linking resulted in an oligomeric protein of approximate molecular weight 180 000 which contained three proteins, two with Mr of about 52 000 and one about 79 000. Under the cross-linking conditions, little or no denaturation of cytochrome P-450 and NADPH-cytochrome c reductase was observed. The aryl hydrocarbon hydroxylase activity was significantly inhibited by the bifunctional cross-linking reagent, DSP, but not by the monofunctional reagent N-succinimidyl-3-(4-hydroxyphenyl) propionate. However, attempts to regenerate the aryl hydrocarbon hydroxylase activity by cleavage of the disulfide linkage with 2-mercapto-ethanol or dithiothreitol were not successful.  相似文献

The mechanisms by which macromolecular impurities degrade the diffraction properties of protein crystals have been investigated using X-ray topography, high-resolution diffraction line shape measurements, crystallographic data collection, chemical analysis, and two-photon excitation fluorescence microscopy. Hen egg-white lysozyme crystals grown from solutions containing a structurally unrelated protein (ovotransferrin) and a related protein (turkey egg-white lysozyme) can exhibit significantly broadened mosaicity due to formation of cracks and dislocations but have overall B factors and diffraction resolutions comparable to those of crystals grown from uncontaminated lysozyme. Direct fluorescence imaging of the three-dimensional impurity distribution shows that impurities incorporate with different densities in sectors formed by growth on different crystal faces, and that impurity densities in the crystal core and along boundaries between growth sectors can be much larger than in other parts of the crystal. These nonuniformities create stresses that drive formation of the defects responsible for the mosaic broadening. Our results provide a rationale for the use of seeding to obtain high-quality crystals from heavily contaminated solutions and have implications for the use of crystallization for protein purification. Inhibitory and excitatory connections of remarkably precise topographic order are characteristic features of the mammalian auditory system, particularly within the superior olivary complex (SOC). Little is known about the requirements for the correct development of these specific connections. Previous in vivo experiments have demonstrated a high expression of calcium-binding proteins in this system during development, pointing to the need for precise calcium regulation. Here, we have employed an organotypic slice culture from the above neuronal network and analyzed the requirements for the maintenance and development of this system in vitro. When slices from neonatal rats were incubated in standard culture medium for up to 7 days, we found no organotypic features. Only if 25 mM KCl was added to the culture medium, the cytoarchitecture of the nuclei, the neuronal morphology, and the specificity and topography of internuclear connections were indistinguishable from that in vivo. The addition of calcium channel blockers (MgCl2 and nifedipine) to the high-KCl medium reduced organotypicity drastically, indicating that a depolarization-induced increase of intracellular calcium is indispensable. Furthermore, the temporal course of the expression of the calcium-binding protein parvalbumin in culture under high KCl mimics that in vivo, demonstrating developmental processes during incubation. The need for calcium influx into neurons of this auditory network in vitro (which is not seen in other slice culture systems) strengthens the hypothesis that an optimal calcium concentration is exceptionally important in auditory neurons. The effect of KCl in the slice cultures may substitute for input activity regulating intracellular calcium in auditory neurons in vivo. © 1998 John Wiley & Sons, Inc. 中国西南亚高山云冷杉林中大熊猫主食竹的地上生物量及其生物和非生物影响因子 作为大熊猫(Ailuropoda melanoleuca)的主食竹之一,缺苞箭竹(Fargesia denudate)广泛分布在中国西南山地云冷杉林的林下。然而,缺苞箭竹生物量及其分布的驱动因素仍不清楚。本研究基于对王朗大样地(25.2 ha) 209个样方(投影面积为20 m × 20 m)中的乔木层和灌木层(包括竹子等木质草本)的系统调查,探讨非生物因素(地形和土壤理化性质)和生物因素(乔木密度、乔木总胸高断面积(TBA)和灌木盖度等) 对缺苞箭竹地上生物量的影响。研究结果表明,样方缺苞箭竹地上生物量平均为1.17 ton/ha,在209 个 调查样方中差异很大(0–4.88 ton/ha,95%置信区间)。缺苞箭竹生物量随海拔、坡度和乔木平均胸径的增 加而显著增加,随乔木密度、灌木盖度和土壤pH的增加而显著降低,而与乔木TBA、坡向、土壤有机质或总氮含量无显著线性关系。随机森林模型表明,地形和生物因素对缺苞箭竹生物量的影响比土壤理化性质更大。具体来说,地形主要通过改变乔木密度和土壤理化性质来影响缺苞箭竹的生物量。本研究结果可为大熊猫的保护和亚高山云冷杉林的管理提供科学参考。  相似文献

Having the ability to control cell behaviour would be of great advantage in tissue engineering. One method of gaining control over cell adhesion, proliferation, guidance and differentiation is use of topography. Whilst it has be known for some time that cells can be guided by micro‐topography, it is only recently becoming clear that cells will respond strongly to nano‐scale topography. The fact that cells will take cues from their micro‐ and nano‐environment suggests that the cells are in some way ‘spatially aware’. It is likely that cells probe the shape of their surroundings using filopodia, and that this initial filopodia/topography interaction may be critical to down‐stream cell reactions to biomaterials, or indeed, the extracellular matrix. One intriguing question is how small a feature can cells sense? In order to investigate the limits of cell sensing, high‐resolution scanning electron microscopy has been used to simultaneously view cell filopodia and 10 nm high nano‐islands. Fluorescence microscopy has also been used to look at adhesion formation. The results showed distinct filopodial/nano‐island interaction and changes in adhesion morphology.  相似文献

Plants of spring wheat (Triticum aestivum L. cv. Saxana) were grown during the autumn. Over the growth phase of three leaves (37 d after sowing), some of the plants were shaded and the plants were grown at 100 (control without shading), 70, and 40 % photosynthetically active radiation. Over 12 d, chlorophyll (Chl) and total protein (TP) contents, rate of CO2 assimilation (P N), maximal efficiency of photosystem 2 photochemistry (FV/FP), level of lipid peroxidation, and activities of antioxidative enzymes ascorbate peroxidase (APX) and glutathione reductase (GR) were followed in the 1st, 2nd, and 3rd leaves (counted according to their emergence). In un-shaded plants, the Chl and TP contents, P N, and FV/FP decreased during plant ageing. Further, lipid peroxidation increased, while the APX and GR activities related to the fresh mass (FM) decreased. The APX activity related to the TP content increased in the 3rd leaves. The plant shading accelerated senescence including the increase in lipid peroxidation especially in the 1st leaves and intensified the changes in APX and GR activities. We suggest that in the 2nd and 3rd leaves a degradation of APX was slowed down, which could reflect a tendency to maintain the antioxidant protection in chloroplasts of these leaves.  相似文献

Based on the integration of geological information, the pollen record, radiocarbon data, and modern mangrove distribution, this paper proposes a model of evolutionary development for mangroves on the Brazilian Bragança Peninsula driven by relative sea-level changes from the middle to late Holocene. After a postglacial relative sea-level rise (RSL), the mangrove habitats on the Bragança Peninsula began to develop at about 5100 years BP close to the current RSL, originating in the middle of the peninsula. Between 1800 and 1400 years BP, this littoral area underwent a maximum RSL fall of 1 m below the current one, followed by a gradual RSL rise until 1000 years BP, when the modern RSL was reached. Between 5100 and 1000 years BP, the RSL of the Bragança coastline was probably never higher than 0.6 m above the present level. The first Holocene mangrove forest (5100 years BP) in this region disappeared from the plains due to RSL fall and was replaced by herbaceous vegetation (Cyperaceae and Poaceae).  相似文献
