This protocol provides multiple methods for the analysis and quantification of predatory feeding behaviors in nematodes. Many nematode species including Pristionchus pacificus display complex behaviors, the most striking of which is the predation of other nematode larvae. However, as these behaviors are absent in the model organism Caenorhabditis elegans, they have thus far only recently been described in detail along with the development of reliable behavioral assays 1. These predatory behaviors are dependent upon phenotypically plastic but fixed mouth morphs making the correct identification and categorization of these animals essential. In P. pacificus there are two mouth types, the stenostomatous and eurystomatous morphs 2, with only the wide mouthed eurystomatous containing an extra tooth and being capable of killing other nematode larvae. Through the isolation of an abundance of size matched prey larvae and subsequent exposure to predatory nematodes, assays including both "corpse assays" and "bite assays" on correctly identified mouth morph nematodes are possible. These assays provide a means to rapidly quantify predation success rates and provide a detailed behavioral analysis of individual nematodes engaged in predatory feeding activities. In addition, with the use of a high-speed camera, visualization of changes in pharyngeal activity including tooth and pumping dynamics are also possible.  相似文献

A growing number of studies have assessed the personality of pet owners. However, although there is a large number of people who own exotic pets, their personalities have seldom been examined. Furthermore, studies of personality of pet owners have focused almost exclusively on typical personality traits, ignoring associations with “dark” traits. Here, we assessed both traditional and some dark personality features in association with pet ownership and attachment in 325 pet owners via an online survey. We predicted that individuals scoring higher on narcissism and borderline personality features would be a) more likely to own exotic pets, and b) less attached to their pets compared with people scoring lower on narcissism and traditional pet owners. Additionally, we theorized that neurotic pet owners would be more attached to their pets compared with less neurotic pet owners. We did not find an association between personality and exotic pet ownership but we found that those high in grandiose narcissism were actually more attached to their traditional pets. Those high in vulnerable narcissism were more attached only if their pets were exotic. Those high in borderline features were less attached to both kinds of pets. Personality assessments including “dark” features of personality may therefore be useful in predicting attachment to pets during the matching process of potential adopters to pets.  相似文献

Orthodontic tooth movement (OTM) is an adaptive biomechanical response of dentoalveolar components to orthodontic forces, in which remodeling of the alveolar bone occurs in response to changes in the surrounding mechanical environment. In this study, we developed a framework for OTM simulation by combining an image-based voxel finite element method, with a surface-tracking level set method using three-dimensional computer models. For a case study to demonstrate its capability of expressing clinical tooth movement, we observed displacement and rotation of the tooth under three types of force conditions. The simulation results demonstrate that the proposed simulation method has the potential to predict clinical OTM.  相似文献

In orthodontic treatment, the locations of the centre of resistance (CR) of individual teeth and the applied load system are the major determinants for the type of tooth movement achieved. Currently, CR locations have only been specified for a relatively small number of tooth specimen for research purposes. Analysing cone beam computed tomography data samples from three upper central incisors, this study explores whether the effort to establish accurate CR estimates can be reduced by (i) morphing a pre-existing simplified finite element (FE) mesh to fit to the segmented 3D tooth-bone model, and (ii) individualizing a mean CR location according to a small parameter set characterising the morphology of the tooth and its embedding. The FE morphing approach and the semi-analytical approach led to CR estimates that differ in average only 0.04 and 0.12 mm respectively from those determined by very time-consuming individual FE modelling (standard method). Both approaches may help to estimate the movement of individual teeth during orthodontic treatment and, thus, increase the therapeutic efficacy.  相似文献

The purpose of this study is to measure the failure risk of a crown depending on the cusp angle. Three all-ceramic crown models consisting of CH (high incline), CM (middle incline), and CL (low incline) are designed. Stress is applied to the crown with Loading case-1 (top of cusp tip) and Loading case-2 (middle of cusp ridge) with the use of FEA software. In Loading case-1 and case-2, the CH showed the highest Maximum Principal Stress (MPS) while the CL showed the lowest MPS. The cusp angle is an influential factor affecting stress distribution in dental crowns.  相似文献

N-ethylmaleimide sensitive factor (NSF) is an ATPases associated with various cellular activities protein (AAA), broadly required for intracellular membrane fusion. NSF functions as a SNAP receptor (SNARE) chaperone which binds, through soluble NSF attachment proteins (SNAPs), to SNARE complexes and utilizes the energy of ATP hydrolysis to disassemble them thus facilitating SNARE recycling. While this is a major function of NSF, it does seem to interact with other proteins, such as the AMPA receptor subunit, GluR2, and beta2-AR and is thought to affect their trafficking patterns. New data suggest that NSF may be regulated by transient post-translational modifications such as phosphorylation and nitrosylation. These new aspects of NSF function as well as its role in SNARE complex dynamics will be discussed.  相似文献

Mast cells are important players in innate immunity and mediate allergic responses. Upon stimulation, they release biologically active mediators including histamine, cytokines and lysosomal hydrolases. We used permeabilized rat basophilic leukaemia cells as model to identify R-SNAREs (soluble NSF (N-ethylmaleimide-sensitive fusion protein)) mediating exocytosis of hexosaminidase from mast cells. Of a complete set of recombinant mammalian R-SNAREs, only vesicle associated membrane protein (VAMP8)/endobrevin consistently blocked hexosaminidase release, which was also insensitive to treatment with clostridial neurotoxins. Thus, VAMP8, which also mediates fusion of late endosomes and lysosomes, plays a major role in hexosaminidase release, strengthening the view that mast cell granules share properties of both secretory granules and lysosomes.  相似文献
