Experiments were conducted to test the possibility that organic amines inhibit ethylene production by inhibiting transport of the ethylene precursor, 1-aminocyclopro-pane-1-carboxylic acid (ACC), into the vacuole. α-Aminoisobutyric acid (αAIB) was used as a model substrate to study ACC uptake into the vacuole in relationship to ethylene production in pericarp slices of Lycopersicon esculentum Mill. cv. Liberty treated with and without organic amines and related substances. Organic amines (polyamines and other basic amines) inhibited αAIB uptake into the vacuole. These amines also enhanced ACC accumulation in the tissue and reduced the passive efflux of αAIB from the vacuole. Overall, ethylene production was inhibited. The inhibition of αAIB transport and of ethylene production followed a polyvalent cationic progression in the order polyamines > diamines> basic 1-amino acids. Ca2+, but not Mg2+, strongly stimulated αAIB uptake into the vacuole and ethylene production. At equal concentrations, Ca2+ counteracted the inhibitory effects of polyamines on both αAIB uptake and ethylene production. Competitive and irreversible inhibitors of polyamine biosynthesis stimulated αAIB uptake into the vacuole and ethylene production. The results indicate an apparent relationship between polyamines, ACC uptake into the vacuole and ethylene production.  相似文献

The influence of nicotianamine (NA) on formation and elongation of adventitious roots in hypocotyls of de-rooted NA-less mutant seedlings of Lycopersicon esculentum Mill, was examined in relation to the iron supply [ferric N-N'-ethylenediaminedi-(2-hydroxyphenylacetate) (FEDDHA), ferric ethylenediaminetetracetate (FeEDTA), ferric N-(2-hydroxyethyl)-ethylenediaminetriacetate (FeHEDTA, Fe-citrate and FeCl3] in the nutrient solution. The initiation of root primordia in hypocotyl cuttings was independent of NA and occurred with about the same frequency in both, mutant and wild-type. In the mutant the development of primordia to adventitious roots was blocked at all iron sources used, except FeEDTA. Addition of NA (5x 10−6 to 2 × 10−5 M ) to the rooting medium resulted in a fast growth of adventitious roots in mutant cuttings with all iron sources tested. Rooting of wild-type cuttings was independent from NA application and iron sources. We suppose that NA is involved in the intracellular transport of iron. Its function is possibly linked with chelation of ferrous iron in the cell.  相似文献

利用热偶湿度计(thermocouplepsychrometer)研究了野生型、GA-缺陷型和ABA-缺陷型番茄发育过程中果实种子的水分关系,发现除ABA-缺陷型种子胶囊和果肉水势变化特殊外,3种类型果实水分状况变化基本一致;在整个发育时期内.前期种子胶囊和果肉水分流向种子,中期种子水分流向种子胶囊和果肉,后期种子和果实间的水势达到平衡。鉴于种胚脱水是一种主动过程,种胚水势一直低于整个种子、种子胶囊和果肉。内源赤霉素可明显增加果实和种子的重量,但对增加种胚溶质的作用不大。由于内源脱落酸可以促使果实成熟和衰老,促进果实细胞解体,大大降低种子胶囊和果肉水势,因而抑制成熟种子在果实内萌发。  相似文献

We investigated the effects of brushing on the chilling tolerance and metabolism of nonstructural carbohydrates (soluble sugars and starch) in tomato leaves before, during and after a chilling stress. Tomato plants ( Lycopersicon esculentum Mill. cv. Caruso) were cultivated either without mechanical stress application (control plants) or with daily brushing treatments for 15 days (brushed plants), prior to a 7-day chilling treatment (8/5°C day/night). Brushing resulted in shorter plants with a 34% reduction in leaf dry weight per area and a 59% reduction of soluble sugars and starch, on a dry weight basis. The sugar to starch ratio was not affected by brushing. A greater chilling tolerance in the brushed plants was demonstrated by the maintenance of a significantly higher PSII efficiency in brushed plants (42%) compared to that of the control plants (30%) after 7 days of chilling treatment, less visible damage to the leaf tissue, and a more rapid resumption of growth during 3 days of recovery as compared to control plants. During the chilling treatment levels of soluble sugars per leaf dry weight increased 15-fold in the brushed plants and 5-fold in control plants. In the present study we have demonstrated that brushing can increase chilling tolerance in tomato plants. The observed differences in chilling tolerance and concentration of soluble sugars in the leaves may indicate an involvement of soluble sugar levels in acclimation to chilling.  相似文献

Development of galactomannan-hydrolyzing activity, that is involved in the weakening of the mechanical restraint of the endosperm, was followed at pre-germinative stages in tomato ( Lycopersicon esculentum ) seed. Prior to germination the activity developed exclusively in the endosperm portion just adjacent to the radicle tip. In other parts of the endosperm, the activity developed only after germination occurred. Under the conditions where germination was suppressed (far-red light- or ABA-treatment). no activity was detected in the endosperm at the pre-germinative stages. Under the conditions where the inhibition of germination was alleviated (far-red + red or ABA + GA3), the activity developed prior to germination in the endosperm part in front of the radicle tip. Thus, a clear parallel relationship was observed between germinability of the seed and the pre-germinative development of activity in the part of the endosperm portion adjacent to the radicle tip.  相似文献

I considered the possibility that changes in fruit photosynthesis obscure the occurrence of the climacteric rise in respiration in tomato fruits attached to the plant. Internal CO2 and ethylene concentrations in tomatoes ( Lycopersicon esculentum Mill. cv. OH 7814) were analyzed after direct sampling through polyethylene tubes implanted in the external pericarp. Fruits which were shaded with aluminium foil contained up to 60 ml 1−1 CO2, until the internal ethylene concentration exceeded 1 μl l−1, when CO2 concentration declined to below 40 ml l−1; the CO2 concentration in fruits exposed to light only occasionally exceeded 40 ml 1−1. The internal CO2 concentration of detached fruits first declined and then increased along with ethylene concentration, as expected for the climacteric. Detached green fruits under continuous low photosynthetic photon flux density (100 μmol m−2 s−1) contained almost no internal CO2 and produced no CO2. Changes in photosynthesis and an associated CO2-generating system in green fruits are thought to obscure the climacteric rise in tomato fruits developing on the plant.  相似文献

The nucleotide sequence of a cDNA prepared from poly(A)+ RNA from Lycopersicon esculentum fruit codes for a protein, M r 20812, with features representative of the protein core of arabinogalactan proteins. The deduced amino acid sequence resembles that of peptides of arabinogalactan proteins isolated from carrot and rose and is most similar to the sequence of tryptic peptides from Lolium multiflorum (Gleeson et al., Biochem J 264 (1989) 857–862). The similar sequences include a number of Ala-Pro repeats, a feature considered distinctive of arabinogalactan proteins. The amino acid composition is similar to that of the peptide core of the Lolium multiflorum arabinogalactan protein; alanine, serine and proline account for 57% of the polypeptide. The mRNA corresponding to the cDNA sequence was detected in roots, leaves and fruit. The levels of mRNA are reduced in older leaves, in fruit that have commenced ripening and in leaves and fruit that have been wounded.  相似文献

A simple and sensitive chemical assay was developed for 1-aminocyclopropane-1-carboxylic acid (ACC), a precursor of ethylene. The assay is based on the liberation of ethylene from ACC at pH 11.5 in the presence of pyridoxal phosphate, MnCl2 and H2O2. This assay was used to detect ACC in extracts of tomato fruits (Lycopersicon esculentum Mill.) and to measure the activity of a soluble enzyme from tomato fruit that converted S-adenosylmethionine (SAM) to ACC. The enzyme had a Km of 13 M for SAM, and conversion of SAM to ACC was competitively and reversibly inhibited by aminoethoxyvinylglycine (AVG), an analog of rhizobitoxine. The Ki value for AVG was 0.2 M. The level of the ACC-forming enzyme activity was positively correlated with the content of ACC and the rate of ethylene formation in wild-type tomatoes of different developmental stages. Homogenates of tomato fruits catalysed the enzymic conversion of linoleic and linolenic acids (but not oleic acid) to C6 aldehydes in low (3–5%) molar yield. Hexanal was formed from linoleic acid; cis-3-hexenal and smaller amounts of trans-2-hexenal were formed from linolenic acid. With the fatty acids as substrates, the major products were fatty acid hydroperoxides (50–80% yield) and the ratio of 9- to 13-hydroperoxides as isolated from an incubation with linoleic acid was at least 95:5 in favour of the 9-hydroperoxide isomer. When the 9- and 13-hydroperoxides of linoleic acid were used as substrates with tomato homogenates, the 13-hydroperoxide was readily cleaved to hexanal in high molar yield (60%) but the 9-hydroperoxide isomer was not converted to cleavage products. Properties of the hydroperoxide cleavage system are described. The results indicate that the C6 aldehydes are formed from C18 polyunsaturated fatty acids in a sequential enzyme system involving lipoxygenase (which preferentially oxygenates at the 9-position) followed by a hydroperoxide cleavage system which is, however, specific for the 13-hydroperoxy isomers.  相似文献
