The effects of five antipathogenic plant peptides, wheat α-thionin, potato PTH1 defensin, barley LTP2 lipid transfer protein, and potato tuber DL1 and DL2 defensins, have been tested against phospholipid vesicles (liposomes). Wheat thionin very actively induces aggregation and leakage of negatively charged vesicles. LTP2 displays the same activities, although to a limited extent. Under certain conditions PTH1 and DL2 induce vesicle aggregation, but not leakage. Potato defensin DL1 failed to show any effect on liposomes. The same peptides have been assayed against a plant pathogenic bacterium, both the membrane-active and -inactive compounds having efficient antibacterial action.  相似文献

A model of single-species growth in the chemostat on two non-reproducing, growth-limiting, noninhibitory, perfectly substitutable resources is considered. The medium in the growth vessel is enriched by increasing the input concentration of one of the resources. Analytical methods are used to determine the effects of enrichment on the asymptotic behaviour of the model for different dilution rates. It is shown that there exists a threshold value for the dilution rate which depends on the maximal growth rate of the species on each of the resources. Provided the dilution rate is below the threshold, enrichment is beneficial in the sense that the carrying capacity of the environment is increased, regardless of which resource is used to enrich the environment. When the dilution rate is increased beyond the threshold, it becomes important to consider which resource is used for enrichment. For one of the resources it is shown that, while moderate enrichment can be beneficial, sufficient enrichment leads to the extinction of the microbial population. For the other resource, enrichment leads from washout or initial condition dependent outcomes to survival, and is thus beneficial. There are important implications of these results to the management of natural aquatic ecosystems. For example, while enrichment may be beneficial to the microbial species during the summer months, it can lead to their decimation during spring run-off, when the natural dilution rate is higher.Research partially supported by an Ontario Graduate Scholarship. Summary The effect of experimental and integration errors on the calculations in interproton distances from NOE intensities is examined. It is shown that NOE intensity errors can have a large impact on the distances determined. When multiple spin (spin diffusion) effects are significant, the calculated distances are often underestimated, even when using a complete relaxation matrix analysis. In this case, the bias of distances to smaller values is due to the random errors in the NOE intensities. We show here that accurate upper and lower bounds of the distances can be obtained if the intensity errors are properly accounted for in the complete relaxation matrix calculations, specifically the MARDIGRAS algorithm. The basic MARDIGRAS algorithm has been previously described [Borgias, B.A. and James, T.L. (1990) J. Magn. Reson., 87, 475–487]. It has been shown to provide reasonably good interproton distance bounds, but experimental errors can compromise the quality of the resulting restraints, especially for weak cross peaks. In a new approach introduced here, termed RANDMARDI (random error MARDIGRAS), errors due to random noise and integration errors are mimicked by the addition of random numbers from within a specified range to each input intensity. Interproton distances are then calculated for the modified intensity set using MARDIGRAS. The distribution of distances that define the upper and lower distance bounds is obtained by using N randomly modified intensity sets. RANDMARDI has been used in the solution structure determination of the interstrand cross-link (XL) formed between 4-hydroxymethyl-4,5,8-trimethylpsoralen (HMT) and the DNA oligomer d(5-GCGTACGC-3)2 [Spielmann, H.P. et al. (1995) Biochemistry, 34, 12937–12953]. RANDMARDI generates accurate distance bounds from the experimental NOESY cross-peak intensities for the fixed (known) interproton distances in XL. This provides an independent internal check for the ability of RANDMARDI to accurately fit the experimental data. The XL structure determined using RANDMARDI-generated restrains is in good agreement with other biophysical data that indicate that there is no bend introduced into the DNA by the cross-link. The process of labyrinth determination has been studied in three urodelean and seven anuran species by means of homoplastic transplantation of ear region epidermis, defined as the piece of epidermal layer containing the material of the prospective ear vesicle (labyrinth rudiment). The ear region epidermis was grafted onto the abdominal wall of embryos of the same developmental stage. The earliest stage of operation resulting in ectopic ear vesicle formation was determined, suggesting the appearance of organ-specific properties in the ear ectoderm. These properties were enhanced in the further course of development, as indicated by the frequency of ear vesicle formation and by the volume and degree of complexity that the vesicles reached. The data obtained allowed us to arrange the species studied in a sequence, ranging from most Ranidae and Bufo viridis, in which organ-specific properties appear earlier and are most strongly expressed, to Triturus vulgaris in which their expression is least pronounced. Comparison of properties of the material giving rise to the ear vesicle or to several other ectodermal derivatives led to the conclusion that species-specific differences in determination of different ectodermal rudiments are due to species specific properties of the whole ectoderm. These differences appear to be determined by an evolutionary shift of the beginning of gastrulation towards later cleavage cycles.Deceased in November 1993  相似文献

以3d龄Sprague-Dawley大鼠腹腔注射苯丙氨酸(Phe)诱导高苯丙氨酸血症,荧光法测定大脑皮层及其突触体中去甲肾上腺素(NE)、多巴胺(DA)和5-羟色胺(5-HT)含量;Y型电迷宫法测其学习记忆能力.结果显示:高苯丙氨酸血症大鼠大脑皮层NE、DA及5-HT含量降低38.6%~67.4%,突触体中NE、DA和5-HT含量降低51.9%~70.2%,学习记忆能力明显低于对照组.结果提示,苯酮尿症智力障碍可能与大脑皮层及其突触体中某些单胺类递质含量降低相关.  相似文献

采用局限性损毁和刺激垂体的方法,以行为测痛为指标,观察大鼠垂体在痛觉调节中的作用以及地塞米松(Dex)对其影响。实验结果显示,损毁垂体中间叶(IL)及邻近的前叶(AL),大鼠痛阈明显低于手术前的痛阈(P<0.01)。电刺激垂体的上述同样部位,大鼠痛阈明显高于手术基础值及自身假刺激值(P<0.001)。经Dex处理的动物,电刺激垂体不再引起痛阈升高。结果表明,大鼠垂体IL及靠近AL与痛调节有关,这种  相似文献

A photomicrobial sensor consisting of immobilized Chlorella vulgaris and an oxygen electrode has been developed for selective determination of phosphate. When 40 mM phosphate was added to the sensor system, the photocurrent increased to a maximum under light irradiation with a response time of 1 min. The current increased with increasing phosphate concentration in the range 8–70 mM. Selectivity of the sensor was satisfactory. Good agreement was obtained between the phosphate concentrations in lake water determined by the photomicrobial sensor and by conventional colorimetry (correlation coefficient 0.96).  相似文献

Roche Susceptibility Test (RST) Medium represents the most completely optimized and convenient fully defined medium described. It requires no post-autoclaving supplementation with vitamins, supports good growth of all common aerobic and anaerobic pathogens and may be used as a broth or agar gel on which the swarming of Proteus spp. is virtually eliminated. The broth as a superior buffering capacity to most complex media and an osmolality and pH close to those of human serum. RST is highly satisfactory for the susceptibility testing of commonly used antibiotics and meets the requirements of the National Committe for Clinical Laboratory Standards of the U.S.A. in almost every respect.  相似文献

A highly sensitive fluorimetric assay using 3-O-methylfluorescein phosphate as substrate was used in the determination of K+-dependent phosphatase activity in preparations of rat skeletal muscle. The gastrocnemius muscle was chosen because of mixed fibre composition. Crude, detergent treated homogenate was used so as to avoid loss of activity during purification. K+-dependent phosphatase activities in the range 0.19–0.37 μmol · (g wet weight)−1 · min−1 were obtained, the value decreasing with age and K+-deficiency. Complete inhibition of the K+-dependent phosphatase was obtained with 10−3 M ouabain. Using a KSCN-extracted muscle enzyme the intimate relation between K+-dependent phosphatase activity and (Na+ + K+)-activated ATP hydrolysis could be demonstrated. A molecular activity of 620 min−1 was estimated from simultaneous determination of K+-dependent phosphatase activity and [3H]ouabain binding capacity using the partially purified enzyme preparation. The corresponding enzyme concentration in the crude homogenates was calculated and corresponded well with the number of [3H]ouabain binding sites measured in intact muscles or biopsies hereof.  相似文献

In reproducing apterae of Megoura viciae, parturition is often completely arrested during periods of isolation from the host plant. In contrast, surgical removal of the rostrum (including the stylets), amputation of the extremities of the legs, or decapitation, all stimulate parturition away from the plant. These operations also induce alata-producing aphids to revert immediately to the production of apterae, but have no detectable effect on aptera-producers. Carbon dioxide or ether anaesthesia and nitrogen narcosis have a similar action on this maternally controlled response. Although the rostrum and tibio-tarsus bear sensilla whose removal might well be involved in inducing parturition, the influence on morph change is probably indirect and is to a great extent associated with the delay in the resumption of parturition. The effect can be reproduced by isolating individual aphids away from the food plant. The morph change cannot, however, be attributed to starvation since it also occurs when the genital pore of an actively feeding aphid is temporarily occluded. The change in physiology appears to be associated with the retention of embryos at a time when there is no sensory input from crowding.
Résumé Chez les Megoura viciae aptères, la parturition est souvent complètement arrêtée pendant les périodes de séparation de la plante hôte. Par opposition, l'amputation chirurgicale du rostre (y compris les stylets), de l'extrémité des pattes (tarse et une partie du tibia), ou la décapitation, stimulent toutes la parturition en l'absence de la plante. Les pucerons induits à produire des ailés (élevage antérieur en groupe) retournent, après ces opérations, immédiatement vers la production d'aptères. Le dioxyde de carbone ou l'anesthésie à l'éther et la narcose à l'azote ont une action semblable sur ce déterminisme maternel. Bien que l'élimination des sensilla portées par le rostre et par l'ensemble tarse-tibia puisse être déterminante dans l'induction de la parturition, l'action sur le changement de type semble être principalement associée au retard consécutif dans la reprise de la parturition. Les anesthésiants qui, eux aussi, retardent l'apparition ou la reprise de la parturition, ont probablement une action indirecte du même type.Les pucerons groupés, isolés de la plante hôte pendant plus de 24 h, ont aussi tendance à retourner immédiatement à la production d'aptères. Ce changement de type ne peut, cependant, être attribué au jeûne puisqu'il se produit aussi quand, chez un puceron s'alimentant activement, le pore génital est momentanément bouché. Le changement physiologique semble être associé à la rétention des embryons à un moment où il n'y a pas l'influence sensorielle du groupement.Aucun de ces traitements, à l'exception du groupement, n'induit des pucerons antérieurement isolés à devenir des producteurs d'ailés.
