A multi component based synthesis involving palladium catalyzed C-C bond forming reaction has been developed as a new strategy to access systematically modified functionalized 2-aminochromenes. This MCR involves the use of bromobenzaldehyde as a key component and is highlighted by generating a new compound library. Many of these compounds showed Mycobacterium tuberculosis H37Rv chorismate mutase inhibiting properties in vitro representing the lead example of chorismate mutase inhibition by heteroarene based compounds. 相似文献

Acylation of Pseudomonas cepacia lipase with Pyromellitic dianhydride to modify 72% of total amino groups was carried out. Different organic solvents were screened for precipitation of modified lipase. It was found that 1,2-dimethoxyethane was the best precipitant which precipitated 97% protein and complete activity. PCMC (protein coated microcrystals), CLPCMC (crosslinked protein coated microcrystals), EPROS (enzyme precipitated and rinsed with organic solvents) and pH tuned preparations of modified and unmodified lipase were prepared and used for carrying out transesterification reaction with n-octane and dimethyl formamide (DMF) as reaction medium. In n-octane, among all the preparations, CLPCMC of modified lipase gave highest rate (1970 nmol min−1 mg−1) as compared to unmodified pH tuned lipase (128 nmol min−1 mg−1). In DMF, with both 1% (v/v) and 5% (v/v) water content, CLPCMC showed highest initial rate of 0.72 and 7.2 nmol min−1 mg−1, respectively. 目的:观察左旋门冬酰胺酶两种常用剂型在急性淋巴细胞性白血病患儿联合化疗中的治疗反应.方法:本院2010-3-2010.12行长春新碱+吡柔比星+门冬酰胺酶+强的松方案化疗的急性淋巴细胞白血病患儿,使用含有聚乙二醇化的左旋门冬酰胺酶剂型培门冬酶即VDPAP者作为A组(40例)与既往使用含大肠杆菌剂型即VDLP者B组(60例)对比,观察不良反应.化疗前后检测血象,肝功能,凝血功能,观察过敏情况等,记录化疗后第28天的各项指标.结果:A组过敏发生2例(5%)而B组过敏发生13例(21.67%),P值0.045,有统计学意义.既往对大肠杆菌剂型发生过敏的13例患儿首剂使用PEG-Asp均无过敏,2例于第二剂时出现皮试阳性.A组(>2级)白细胞减少16例,中性粒细胞减少26例,血红蛋白降低0例,血小板减少5例,纤维蛋白原降低1例,部分凝血活酶时间延长0例;B组(>2级)白细胞减少28例,中性粒细胞减少46例,血红蛋白降低2例,血小板减少16例,纤维蛋白原降低2例,部分凝血活酶时间延长0例.A组(未分级)抗凝血酶Ⅱ降低23例,D二聚体升高4例,谷丙转氨酶升高5例:B组(未分级)抗凝血酶Ⅱ降低33例,D二聚体升高8例,谷丙转氨酶升高5例..血液学不良反应差异无统计学意义.A组平均住院日11.14天.B组平均住院日18.47天.结论:左旋门冬酰胺酶两种剂型不良反应相当,但培门冬酶具有使用次数少,使用方便,过敏率低,缩短住院目的优点. 相似文献

The results of investigations into performing DNA sequencing chemistry on a picoliter-scale electrowetting digital microfluidic platform are reported. Pyrosequencing utilizes pyrophosphate produced during nucleotide base addition to initiate a process ending with detection through a chemiluminescence reaction using firefly luciferase. The intensity of light produced during the reaction can be quantified to determine the number of bases added to the DNA strand. The logic-based control and discrete fluid droplets of a digital microfluidic device lend themselves well to the pyrosequencing process. Bead-bound DNA is magnetically held in a single location, and wash or reagent droplets added or split from it to circumvent product dilution. Here we discuss the dispensing, control, and magnetic manipulation of the paramagnetic beads used to hold target DNA. We also demonstrate and characterize the picoliter-scale reaction of luciferase with adenosine triphosphate to represent the detection steps of pyrosequencing and all necessary alterations for working on this scale. 相似文献

The encounter between anisotropic agents in diffusion-controlled reactions is a topic of very general relevance in chemistry and biology. Here we introduce a simplified model of encounter of an isotropic molecule with a pair of partially reacting agents and apply it to the encounter reaction between an antibody and its antigen. We reduce the problem to the solution of dual series relations, which can be solved iteratively, yielding the exact solution for the encounter rate constant at any desired order of accuracy. We quantify the encounter effectiveness by means of a simple indicator and show that the two binding centers systematically behave in an anti-cooperative fashion. However, we demonstrate that a reduction of the binding active sites allows the composite molecule to recover binding effectiveness, in spite of the overall reduction of the rate constant. In addition, we provide a simple formula that enables one to calculate the anti-cooperativity as a function of the size of the binding site for any values of the separation between the two active lobes and of the antigen size. Finally, some biological implications of our results are discussed. 相似文献

The synthesis, modification, structure, and biological activity in vivo of the 16,17- cycloalkanoprogesterone (pregna-D-pentarane) analogues of progesterone are described. A possibility of separation of their biological functions has been demonstrated. A systematic synthesis of a set of uniform compounds that differ in a limited number of alterable parameters was developed. A number of Leuconostoc spp. strains were screened for their ability to produce glucansucrases and carry out acceptor reactions with maltose. Acceptor products were analyzed by thin-layer chromatography (TLC) and it was discovered that they could be grouped into four distinct categories based on oligosaccharide product patterns. These patterns corresponded with structural features of the dextrans each strain is reported to produce. Strains that produced a typical dextran—characterized by a predominantly linear (16)-linked d-glucan chain with a low to moderate degree of branching—produced a homologous series of isomaltooligosaccharides via acceptor reactions. Strains that produced dextrans with moderate to high levels of (12) branch points, exemplified by NRRL B-1299, synthesized the same isomaltodextrins as well as another series of oligosaccharides migrating slightly faster in our TLC system. Strains that produced dextrans with higher levels of (13)-branches, such as NRRL B-742, synthesized isomaltodextrins plus a series of oligosaccharides that migrated slightly more slowly on TLC. And finally, strains known to produce alternansucrase produced isomaltodextrins plus oligoalternans. Within a given type, variability exists in the relative proportions of each product. The data presented here may be useful in selecting strains for the production of specific types of oligosaccharides, for example as prebiotics. 相似文献
