Aim To identify biogeographical boundaries which are obscured by faunal overlap and habitat modification. Location KwaZulu-Natal in south-east, South Africa beyond the southern tip of the Moçambique Coastal Plain. Methods Species abundance data for dung beetles (Scarabaeidae: Scarabaeinae) were collected at six levels from the coastal escarpment (30°16′S – 500 m) to the top of the nearby Drakensberg (29°35′S – 2850 m). Cross-altitudinal boundaries were identified using clustering techniques, beta-diversity indices, and range edge analysis. Biogeographical data for the species were drawn from an extensive reference collection and used to classify the biogeographical affinities of the assemblages. Results Three discrete communities are defined (<10% similarity) from species abundance distributions. These communities occur in coastal forest (500 m), coastal to highveld grassland (500–1500 m), and montane grassland (1900–2850 m). Two of these communities are biogeographically homogeneous comprising >89% east coast endemics (coastal forest) or >84% South African montane endemics (montane grassland) in terms of abundance. The third community in coastal to highveld grassland is biogeographically more heterogeneous. Predominant biota of this community comprise both South African highveld endemics and elements with distributions extending into the tropics. At highveld levels (1500 m), there are proportionately more highveld endemics whereas at lowland levels (500 m), there are proportionately more tropical elements. At 1000 m, there was a change in the balance between these two groups across an anthropogenic gradient due to a decline in the proportion of endemics in favour of temperate/tropical generalists. This gradient from a natural grassland fragment to improved pastures of Kikuyu grass also parallels a decline in species richness and abundance. Species turnover analyses showed three different cross-altitudinal patterns. Range-edge analysis showed a trimodal pattern of species turnover (peaks in forest and the Drakensberg foothills as in the community analysis but also at 1000 m). Five beta-diversity indices showed either a bimodal pattern of turnover (forest/grassland and foothills/middle Drakensberg slopes) or a trimodal turnover pattern (forest/grassland, highveld/Drakensberg foothills, Drakensberg peaks). Main conclusions Clear altitudinal zonation is revealed by community and biogeographical analysis but one natural biogeographical boundary may be obscured by the process of habitat modification. This boundary at 1000 m is revealed by range-edge analysis and is supported by findings for plant communities. Beta-diversity, species turnover patterns diverged slightly from those suggested by the community and range-edge analyses. 相似文献

Often regeneration in orchids is only achieved through protocorms, i.e., the juvenile stage. In order to produce directly shoots via bud regeneration both rapidly and with a high frequency, a transverse thin cell layer (tTCL) method is extended to Rhynchostylis gigantea. Transverse thin cell layer (tTCL) explants (0.3--0.5 mm) excised along the stem from the basis to the shoot tip of one-year-old plantlets were cultured on Murashige and Skoog (1962) medium supplemented with different combinations of benzyladenine (BAP), 1-naphthaleneacetic acid (NAA), thidiazuron (TDZ) and 3% sucrose. The optimal combination for maximal bud regeneration was 3 µM BAP and 3 µM TDZ, giving rise to 11.7 buds per tTCL. Roots were obtained with 10 µM forchlofenuron (CPPU) and 1% sucrose. The in vitro plants (> 3 cm long) obtained 4 to 6 weeks after the tTCLs culture were transferred to the greenhouse; their morphology was normal. Efficient micropropagation of direct production of shoots without passing through protocorm stage of orchid species can be achieved using the thin cell layer (TCL) method. 相似文献

2-methyellipticinium (NSC 226137) does not exhibit any spectral interaction with cytochrome p-450; however, it is transformed by microsomes from livers of phenobarbital induced rats. This transformation is NADPH dependant. According to presently available analytical criteria (HPLC and chromatographic behavior, UV and mass spectra), its product is very likely 2-methyl-9-hydroxyellipticinium (NSC 264137) which is an active antitumor drug in man. Two minor metabolites are also present. The same major product is found in the bile of non-induced rats after intravenous administration of 2-methylellipticinium. 相似文献

以白及(Bletilla striata)假鳞茎为外植体,根据上、中、下部位切取薄片,探索假鳞茎不同部位BA、NAA和TDZ对假鳞茎薄片诱导不定芽的影响,比较假鳞茎薄片不同厚度对褐化率和出芽数的影响,采用正交试验,研究了BA和NAA对不定芽增殖的效果,并对组培苗进行壮苗、生根和移栽。结果表明:假鳞茎的部位对诱导不定芽作用极显著,下部的出芽率显著高于上部和中部,BA和TDZ对诱导不定芽作用显著,NAA对诱导不定芽作用不显著。最佳诱导不定芽的方式为假鳞茎下部薄片在基本培养基+2.0 mg·L^-1BA+1.0 mg·L^-1TDZ的培养基上培养4周,出芽率为93.3%,出芽数为15个,厚度为1.6~2.0 mm的假鳞茎薄片其褐化率最低。最佳的增殖培养基为基本培养基+1.5 mg·L^-1BA+0.3 mg·L^-1NAA,增殖系数达4.3,平均苗高为7.8 cm。本研究成功建立了白及假鳞茎薄片诱导芽为关键技术的快繁技术体系,为白及种质资源创新奠定了基础。 相似文献

Small molecule stimulation of β-cell regeneration has emerged as a promising therapeutic strategy for diabetes. Although chemical inhibition of dual specificity tyrosine-phosphorylation-regulated kinase 1A (DYRK1A) is sufficient to enhance β-cell replication, current lead compounds have inadequate cellular potency for in vivo application. Herein, we report the clinical stage anti-cancer kinase inhibitor OTS167 as a structurally novel, remarkably potent DYRK1A inhibitor and inducer of human β-cell replication. Unfortunately, OTS167’s target promiscuity and cytotoxicity curtails utility. To tailor kinase selectivity towards DYRK1A and reduce cytotoxicity we designed a library of fifty-one OTS167 derivatives based upon a modeled structure of the DYRK1A-OTS167 complex. Indeed, derivative characterization yielded several leads with exceptional DYRK1A inhibition and human β-cell replication promoting potencies but substantially reduced cytotoxicity. These compounds are the most potent human β-cell replication-promoting compounds yet described and exemplify the potential to purposefully leverage off-target activities of advanced stage compounds for a desired application. 相似文献

Females of the fungus gnat species Bradysia difformis , Bradysia optata and Bradysia tilicola are able to produce sexual attractants. In males, the female extracts elicit species-specific courtship behaviour and attraction in an olfactometer bioassay. Attractiveness of the sex pheromone extracts seems to differ in the investigated species, being highly attractive for males in B. difformis and only moderately attractive in B. tilicola . Female B. difformis are attractive from late pupal stages until death of the imago. In contrast, female B. optata and B. tilicola are not attractive to males before emergence from the pupa and attractiveness decreases in older females. The pheromones seem to be located on the surface of several body compartments of freshly emerged females. Sexual pheromones of all three species represent multicomponent blends and can be separated in several fractions via thin layer chromatography (TLC). Males of all species show courtship behaviour when placed on developed TLC sheets, indicating the position of the biologically active compounds. In B. difformis and B. optata , the most non-polar TLC fraction is attractive to males, whereas the single fractions of B. tilicola -pheromone elicited almost no attraction of males. 相似文献

Protocorm-like bodies (PLBs) and callus were induced in epiphytic hybrid Cymbidium Twilight Moon ‘Day Light’, where induction capacity was strongly explant dependent. Following the use of various explant
sources (PLB, leaf tip or base, root tip or base, cell and tissue ‘suspension’), highest PLB formation and callus induction
occurred when we used whole PLBs, PLB segments or PLB transverse thin cell layers (tTCLs) or longitudinal TCLs (lTCLs). Plantlet
growth and photosynthetic state from whole or bisected PLBs, as well as from tTCLs were not significantly different, after
analysis of chlorophyll content. However plantlets generated from lTCLs showed lower values for growth and photosynthetic
parameters. All resultant plants were shown to be cytogenetically identical using RAPD and mtDNA analysis despite cytological
variation and endopolyploidy occuring between different plant parts. Acclimatization and survival rate was shown to be 100%

Abstract Mulga (Acacia aneura Mimosaceae) and spinifex (Triodia spp. Poaceae) habitats together characterize a large part of arid central Australia. Often very abrupt boundaries form between these two habitats, giving rise to a mosaic pattern of contrasting shrub‐grass alterations across the landscape. Reasons for such patterning remain poorly understood though current niche‐based views relate species' distributions to spatial resource gradients or to fire effects. Field survey work was conducted on central Australian mountain ranges to further quantify floristic, regeneration traits, and structural patterning across mulga‐spinifex transitions and to test resource‐ and disturbance‐models that explain these patterns. Compositional analysis demonstrated variability in transition type – in certain cases boundaries denoted true floristic discontinuity and in others, somewhat more of a structural shift. Moreover, it was shown that minimal between‐habitat floristic overlap coincided with the occurrence of distinct edaphic changes, while greater compositional commonality occurred when soil gradients were more diffuse. This indicated that floristic patterning cannot be ascribed to any one single process. In the case of strong soil gradients, between‐habitat segregation most likely resulted from resource‐based niche differentiation; for weaker gradients, fire‐frequency assumed greatest importance. Disturbance theory most readily accounted for the distribution of woody species' post‐fire regeneration traits across habitat boundaries. The results also suggested that biotic factors –viz competition, facilitation and animal‐mediated dispersal – may be of additional consequence for mulga‐spinifex coexistence. Overall, the study served to emphasize the importance of multi‐factor explanation for within‐ and between‐habitat patterning in these mosaics. Cloned cells of a myoblast line show the presence of GM3, GM2, GM1 and GD1a gangliosides. The amount of GM3, GM2 and GM1 gangliosides does not vary significantly during the differentiation of myoblasts to myotubes. However, the concentration of GD1a transiently increases almost 3-fold just prior to the fusion of myoblasts and returns to the basal levels in the myotubes. Mutant myoblasts selected for 5-azacytidine resistance and unable to fuse produce only GM3 and traces of GM2. We conclude that GD1a probably participates in the fusion process through yet unknown mechanism. 相似文献

Human fibroblast interferon (Hu IFN beta) was directly introduced with glass micropipets into the cytoplasm of Hela cells. Such an injection of more than 10(4) molecules per cell failed to induce any antiviral state when challenged with vesicular stomatitis virus (VSV).
