The purpose of this study was to evaluate thermal and near‐infrared (NIR) reflectance imaging methods for the assessment of the activity of root caries lesions. In addition, changes in the lesion structure were monitored with polarization sensitive optical coherence tomography (PS‐OCT). Artificial bovine and natural root caries lesions were imaged with PS‐OCT, and their dehydration rate was measured with thermal and NIR cameras. The lesion activity of the natural root caries samples was also assessed by two clinicians by conventional means according to ICDAS II guidelines. The thickness of the highly mineralized transparent surface layer measured using PS‐OCT increased and the area enclosed by the time‐temperature curve, ΔQ, measured with thermal imaging decreased significantly with longer periods of remineralization in simulated dentin lesions, but the NIR reflectance intensity differences, ΔI, failed to show any significant relationship with the degree of remineralization. The PS‐OCT algorithm for the automated assessment of remineralization successfully detected the highly mineralized surface layer on both natural and simulated lesions. Thermal imaging provided the most accurate diagnosis of root caries lesion activity. These results demonstrate that thermal imaging and PS‐OCT may be ideally suited for the nondestructive root caries lesion activity during a clinical examination.
Threshold‐based thermal time models provide insight into the physiological switch from the dormant to the non‐dormant germinating seed.
This approach was used to quantify the different growth responses of the embryo of seeds purported to have morphophysiological dormancy (MPD) through the complex phases of dormancy release and germination. Aquilegia barbaricina seeds were incubated at constant temperatures (10–25 °C) and 25/10 °C, without pre‐treatment, after warm+cold stratification (W+C) and GA3 treatment. Embryo growth was assessed and the time of testa and endosperm rupture scored. Base temperatures (Tb) and thermal times for 50% (θ50) of embryo growth and seed germination were calculated.
W+C enabled slow embryo growth. W+C and GA3 promoted rapid embryo growth and subsequent radicle emergence. The embryo internal growth base temperature (Tbe) was ca. 5 °C for W+C and GA3‐treated seeds. GA3 treatment also resulted in similar Tb estimates for radicle emergence. The thermal times for embryo growth (θe50) and germination (θg50) were four‐ to six‐fold longer in the presence of GA3 compared to W+C.
A. barbaricina is characterised by a multi‐step seed germination. The slow embryo growth during W+C reflects continuation of the maternal programme of development, whilst the thermal kinetics of both embryo and radicle growth after the removal of physiological dormancy are distinctly different. The effects of W+C on the multiphasic germination response in MPD seeds are only partially mimicked by 250 mg·l?1 GA3. The thermal time approach could be a valid tool to model thermal kinetics of embryo growth and radicle protrusion.
A model is described which predicts those aspects of wheat canopy development and growth which are influential in determining the development of epidemics of foliar pathogens, the efficacy of foliar applied fungicides and the impact of disease on yield; specifically the emergence, expansion and senescence of upper culm leaves in relation to anthesis date. This focus on upper leaves allowed prediction of leaf emergence dates by reference to anthesis, rather than sowing. This avoided the step changes in flag leaf emergence date with temperature, reported with earlier models, without the additional complexity of a stochastic approach. The model is designed to be coupled to models of foliar disease, where the primary effect on yield is via reduction in green canopy area and hence interception of photosynthetically active radiation. Mechanisms were incorporated to allow observations of crop development during the growing season to update state variables and adjust parameters affecting future predictions. The model was calibrated using experimental data, and validated against independent observations of crop development on four wheat cultivars across seven contrasting sites in the UK. Anthesis date and upper culm leaf emergence were always predicted within one week of their observed dates. 相似文献
Starches from exotic corn lines were screened by using differential scanning calorimetry (DSC) to find thermal properties that were significantly different from those exhibited by starches from normal Corn Belt lines. Two independent gelatinization transitions, one corresponding to the melting of a peak at 66 °C and the other to a peak melting at 69 °C, were found in some starches. The melting characteristics were traced to two separate types of granules within the endosperm. Strong correlations were found between DSC properties and proportion of large granules with equivalent diameter ≥17 μm. Starches with a lower peak onset gelatinization temperature (ToG), had a lower normalized concentration of chains with a degree of polymerization (dp) of 15–24 and/or a greater normalized concentration of chains with a dp of 6–12. These studies will aid in understanding structure–thermal property relationships of starches, and in identifying corn lines of interest for commercial breeding. 相似文献
Thermal inactivation kinetics with extrapolation were used to model the survival probabilities of spores of various Bacillus species over time periods of millions of years at the historical ambient temperatures (25–40 °) encountered within the 250 million-year-old Salado formation, from which the putative ancient spore-forming bacteriumSalibacillus marismortui strain 2-9-3 was recovered. The model indicated extremely low-to-moderate survivalprobabilities for spores of mesophiles, but surprisingly high survival probabilities for thermophilic spores. The significance of the results are discussed in terms of the survival probabilities of (i) terrestrial spores in ancient geologic samples and (ii) spores transported betweenplanets within impact ejecta. 相似文献
Centaurea maculosa Lam. (spotted knapweed), a Eurasian perennial forb, has invaded disturbed and undisturbed semiarid grasslands in the western United States. In the past, success in controlling C. maculosa and restoring invaded areas has been limited. Most research has addressed chemical aspects of invasive species interactions with soils, while potential impacts of altered soil physical properties on C. maculosa's success has not been studied. We hypothesized that the persistence of C. maculosa in semiarid rangelands might reflect an ability to alter site conditions. The objective of this study was to compare selected soil physical properties under C. maculosa-dominated and native perennial grass-dominated areas on semiarid grassland. We used six field sites in western Montana containing adjacent plots dominated by C. maculosa and by native perennial grasses. Soil physical properties including particle size fractions, bulk density, and hydraulic and thermal properties, as well as total organic carbon content, of near-surface soils were measured for each vegetation type. Soil physical properties seldom differed between C. maculosa- and native grass-dominated areas. When soil physical properties differed, the differences were inconsistent within and among sites. Presence of C. maculosa did not alter surface soil characteristics at our six sites, thus its persistence on these semi-arid grasslands cannot be explained by an ability to alter near-surface soil characteristics. 相似文献
SUMMARY 1. From measurements at several weir pool sites along the turbid and freshwater Barwon‐Darling River, Australia, the development of persistent stratification (for periods of >5 days) was related to river discharge. For the sites examined, the required discharge to allow the development of persistent stratification was between 100 and 450 ML day?1 during the hotter months. High discharge during the hotter months did not allow the formation of persistent stratification, although diel stratification did occur. Low discharge through the cooler months resulted in diel stratification, although persistent stratification lasting for a few days could occur at times. 2. The growth and dominance of Anabaena circinalis at these sites was closely related to the establishment and maintenance of persistent and strong thermal stratification. Growth only occurred during extended periods (>5 days) of persistent stratification. These conditions not only restrict the displacement of A. circinalis downstream, they also allowed the alga to accumulate in surface waters. 3. The discharge levels required to suppress the formation of persistent stratification at the study sites were variable because of large differences in channel cross‐sectional area. To compensate for this variation, the discharges were converted to flow velocities. A critical velocity of 0.05 ms?1 was sufficient for the suppression of persistent thermal stratification and concurrent A. circinalis growth for all sites. The turbulent velocity (u*) under weak wind mixing at the study locations varied between 2.66 × 10?3 and 2.91 × 10?3 ms?1 at the critical flow velocities. These values may have potential to be applied to other rivers in similar climatic zones to suppress nuisance cyanobacterial growth. 相似文献
The glucose transporter GLUT 1 was isolated from human erythrocytes and reconstituted into endogenous membrane lipids. Results from thermal denaturation studies, using differential scanning calorimetry, indicate that the thermal denaturation temperature of GLUT 1 is significantly lower in the presence of ATP. The lowering of this transition temperature is very dependent on pH. At more acidic pH, ATP has a greater effect of lowering the thermal denaturation temperature of the protein. For example, with 4.8 mM ATP, the denaturation endotherm is lowered by over 10 degrees at pH 4.3, whereas at pH 7.4, ATP does not alter this transition temperature. However, a change in pH alone, in the absence of ATP, has very little effect on the denaturation temperature. Both glucose and salt partially reverse the lowering of the temperature of thermal denaturation caused by ATP. Studies of acrylamide quenching of the Trp residues of GLUT 1 indicate that at neutral pH, ATP increases the Stern-Volmer quenching constant, while glucose lowers it. The results indicate that ATP binds to GLUT 1 and destabilizes the native structure, leading to a lowering of the thermal denaturation temperature and an increase in acrylamide quenching. The effects of ATP are reversed in part by glucose and are also partly electrostatic in nature. 相似文献
