A new restriction endonuclease, EcoO109, has been isolated from Escherichia coli H709c by polyethyleneimine (PEI) precipitation, DEAE-cellulose chromatography and heparin agarose chromatography. The yield was high, more than 3000 units/g of wet cells. The EcoO109 endonuclease recognizes and cleaves a nucleotide sequence of (formula: see text), in the presence of 10 mM Mg2+. The enzyme will be useful for structural analysis and molecular cloning of DNA because of the stability, high yield and easy handling of the producer strain.  相似文献

Abstract Acetobacterium woodii was continuously grown on 3,4,5-trimethoxybenzoate as pure culture or in commensalistic combination with Pelobacter acidigallici and Desulfobacter postgatei . Under pure culture conditions the following growth parameters were determined: μ max= 0.112 h−1, K s= 1.07 mM, Y max= 35 g/mol, and m = 0.22 mmol·g−1·h−1. In coculture with P. acidigallici the affinity for the substrate increased and the K s value was found to be 135 μM. Under batch culture conditions mixed populations of A. woodii, P. acidigallici , and D. postgatei completely mineralized 3,4,5-trimethoxybenzoate to CO2, whereas under continuous culture conditions more than 3 mM acetate remained unused.  相似文献

Abstract A most-probable-number technique was used to quantify the abundance of myxomycetes (myxogastrids) in soil samples taken from 23 'Sites of Special Scientific Interest' and one other site in the West of England. Associated organisms and soil conditions were also recorded. Sixteen of the 24 sites yielded myxomycete plasmodium-forming units (PFUs) in numbers averaging 230 cm−3 fresh soil. Where detectable in samples of grassland soil, the numbers ranged from 20–2750 cm−3, in woodland soil from 20–800 cm−3, and in sand dunes from 80–400 cm−3. Repeated sampling revealed changing numbers at single sites. The abundance of PFUs was correlated positively with numbers of soil amoebae, ciliates and nematodes, and with levels of magnesium and soil density, and negatively with organic matter content and levels of NH4-nitrogen and phosphate. Each PFU probably corresponded to one or several uninucleate cells in the soil rather than to a plasmodium.  相似文献

In an attempt to show that the open field can still be used as a valid measure of fear, Jones (1983) has reported a failure to replicate some of our findings. The present studies show that this was due to procedural and methodological differences. For instance, we found that birds tested in a novel environment behaved quite differently from those, as in Jones' case, which were placed in one resembling the home cage. Moreover, birds housed in isolation for two days prior to testing reacted differently than those, as again in Jones' case, which were reared in isolation from hatching to the time of testing. The results were interpreted as being consistent with our view that open-field behaviour reflects a conflict between the need to reinstate contact with conspecifics on the one hand, and evade predation on the other.  相似文献

Abstract Glycine added to the growth medium of Caulobacter crescentus was found to substitute Cterminal alanine in the peptide side chains of the murein of this species. Murein synthesized in vivo and in vitro in the presence of glycerine was poorly crosslinked as was new murein formed in the presence of the amino acid. The reduced cross-linkage seems to be due to the effect of glycine on the formation of trimeric muropeptides as revealed by high-performance liquid chromatography (HPLC) muropeptide analysis of murein formed in the presence and absence of the amino acid.  相似文献

Abstract Listeriolysin, an SH-activated haemolysin probably involved in Listeria pathogenicity, has been cloned into the cosmid vector pHC79 and was expressed in Escherichia coli HB101 cells. Chromosomal DNA of Listeria monocytogenes serovar 1/2 was partially digested with Mbo I and ligated to the Bam HI cleaved cosmid. From 2000 recombinant clones examined, 12 (0.6%) produced haemolysin in solid and liquid media. All of them contained chromosome fragments of Listeria of about 40 kb. The cloning of the listeriolysin determinant will lead to a better understanding of the basis of Listeria pathogenicity.  相似文献

It is characterized by its terricolous habitat, perithecioid closed ascomata, relatively small spores, cylindrical asci, and the presence of diploschistesic and orsellinic acids in addition to lecanoric acid. Synopsis Aspects of the life history of Barbus anoplus were studied in Lake le Roux, a turbid man-made lake on the Orange River, South Africa. This minnow underwent a population explosion and successfully colonized the shoreline of the newly-formed lake during the early phases of reservoir filling. Male and female B. anoplus reach sexual maturity in one year at about 40 mm fork length. They have a multiple spawning habit with the first spawning in November–January and the second in February–March. The growth of the two resulting cohorts is discussed. It is proposed that the offspring from the second spawning not only acts as a ‘back-up’ but is capable of prolonging the life of that year-class into an additional reproductive season. Most of the minnows die after their second summer, but more offspring from the second spawning, especially females, live into a third summer. Females attain a larger maximum size (73 mm FL) and age (3–4 years) than males (60 mm FL, 2–3 years). B. anoplus is small and short-lived with a high seasonal reproductive potential, which is in contrast to the larger Barbus species in the Orange River system. These life-history traits enable the species to colonize and successfully inhabit unstable environments and probably account for its widespread distribution.  相似文献
