Summary In flagellate green algae two types of fibrous flagellar roots can be distinguished: system I fibres, cross-striated bundles of 2nm filaments (striation periodicity about 30 nm), which are associated with flagellar root microtubules, and system II fibres, contractile bundles of 4–8 nm filaments which are often cross-striated (striation periodicity variable but greater than 80 nm). The major protein of system II fibres is centrin, a Ca2+-modulated phosphoprotein, which is a member of the EF-hand protein family. The major protein of system I fibres (of severalChlamydomonas-type green algae) is a 34 kDa phosphoprotein, named assemblin. Using confocal laser scanning and conventional light microscopy, the morphology and organization of the muscle fibres in a proprioceptor, the thoracic coxal muscle receptor organ (TCMRO), and the associated 'extrafusal' promotor muscle were investigated in two species of decapod crustacea, the crayfish Cherax destructor and the mud crab Scylla serrata . The diameter of the TCMROs was shown to increase distally, with an increase up to 350% recorded for the crayfish. The tapered shape of the crayfish TCMRO was demonstrated to amplify movements mechanically at the transducer region where the afferent nerves attach. Serial sectioning of the TCMROs, showed that the fibre number increased in the proximal to distal direction from 14 to 30 fibres in the crayfish and from 7 to 20 in the crab. Optical sectioning with the laser scanning confocal microscope revealed that the increase in fibre numbers was the result of muscle fibres branching in the distal third section of the TCMRO. The percentage of muscle tissue in the cross-sectional area in the TCMRO was found to be only 35.2% and 64.6% in the crayfish and crab, respectively. Longitudinal sectioning using laser scanning confocal microscopy revealed the average sarcomere length of the TCMRO muscle fibres of both species to be in the intermediate range for crustacean muscle fibres (4.1 ± 0.1 µm and 4.55 ± 0.34 µm for the crayfish and crab) compared with the long sarcomere muscle fibres in the associated promotor muscles (7.87 ± 0.2 and 10.6 ± 0.6 µm). The distinct morphology of the TCMRO muscle fibres – smaller diameter, intermediate sarcomere length and branching of fibres compared to the larger, long sarcomere promotor fibre muscle fibres – suggest that the TCMRO muscle fibres are specialized in their role of proprioception.  相似文献

This work is an investigation of the influence of ABA and calcium on soluble and wall-bound activities of peroxidase and O-diphenol oxidase of cotton fibres at the stages of primary and secondary wall development, during incubation of intact fibres for 3h at 28°C. ABA (10 M) caused marked inhibition of enzyme activities in both the fractions, whereas calcium (1 mM) was promotory. The incorporation of 1 mM EGTA (a calcium chelator) and chlorpromazine (10 g cm–3) (a calmodulin antagonist) resulted in decreased enzyme activities suggesting regulation of enzyme synthesis and/or secretion to the cell wall by calcium-calmodulin. As a general trend, the relative effect of Ca2+ on the activity of peroxidase in the wall was much greater than on the soluble activity, but this was not true of O-diphenol oxidase. Summary The distribution of monoamines in the diencephalon and pituitary of the dogfish, Scyliorhinus canicula, has been investigated using the histochemical fluorescence technique of Falck and Hillarp (Falck and Owman, 1965). Terminals of monoamine-containing axons were found in the neurointermediate lobe of the pituitary and the axons were traced, by means of nialamide and L-dopa treatment and lesions, to the nucleus medius hypothalamicus. A separate hypothalamic system converging on the anterior median eminence and the occurrence of aminergic cells in the nuclei lobi inferiores and nucleus medius hypothalamicus were similarly demonstrated. Normal fish show a bilateral uncrossed tegmental tract and two areas of catecholamine-containing neurones in modified ependymal organs. The organum vasculosum hypothalami includes both primary catecholamine and 5-hydroxytryptamine-containing cell types whilst the organum vasculosum praeopticum has only the former type. Both organs contain cells which send club-like processes into the third ventricle. The subcommissural organ does not contain monoamines.The role of hypothalamic catecholamine systems in the regulation of pituitary function is discussed.  相似文献

Summary Muscle spindles contain two types of intrafusal muscle fibre, nuclear bag fibres and nuclear chain fibres. The intrafusal fibres of rabbit and guinea pig spindles have been studied using quantitative stereological techniques at the ultrastructural level. The crosssectional areas occupied by myofilaments have been measured in the polar and equatorial regions of both types of intrafusal fibre. There are considerably fewer myofilaments in the equatorial regions of both types of fibre compared with their polar regions.This work was carried out with the aid of grants from the Medical Research Council and the Science Research Council of Great Britain.  相似文献

Summary The distribution of adrenergic and peptidergic (Gomori-positive) structures of the hypophysial neuro-intermediate complex in Acipenseridae has been studied by means of light, fluorescence, and electron microscopy. Adrenergic fibres (B-fibres) and their terminals have been detected in the neurohypophysis of these fishes. The terminal swellings of B-fibres as well as the terminals of the neurosecretory peptidergic fibres (A1 and A2) make contact with the basement membrane of the connective tissue layer separating the neurohypophysis from the intermediate lobe. Capillaries are situated within this layer and, therefore, the main part of the fibre terminals is in contact with the pericapillary space. The release of catecholamines from the adrenergic terminals into the capillaries connected with the general circulation is supposed. The diffusion of catecholamines through the connective tissue layer into the parenchyma of the intermediate lobe is also suggested. Hence, the glandular activity of the intermediate lobe seems to be under the dual control of adrenergic and peptidergic elements of the hypothalamus.The authors wish to express their deep appreciation to G. AMP-deaminase activity was measured in white muscle from a wide range of fish, including one cyclostome, 13 chondrosteans, and one teleost to elucidate the pattern of the AMP-deaminase activity in white muscle of fish. Compared to a mammalian (rat) muscle extract, low enzyme activities are found in the cyclostome and two elasmobranchs from two families (Scyliorhinidae, Hexanchidae). In contrast, higher AMP-deaminase activities, similar to mammals, are expressed in Squalidae, all families of skates, Chimaeridae and in the teleostean fish. We then compared AMP-deaminase activities in red and white muscles from two representative elasmobranch fish, the dogfish (Scyliorhinus canicula) and the thornback ray (Raja clavata). The fibre type composition and distribution of the locomotory musculature were determined in these two elasmobranchs to establish a relationship between the morphology, the type of fibres of the locomotion-implicated muscles and the AMP-deaminase activity. Experimental data are discussed with respect to the layout of fibres in the myotome. In both species, three fibre types were identified. In the two fish myotomes, most of the axial muscles are white fibres while red fibres constitute a thin sheet. Some differences were observed between the two species in the distribution of intermediate fibres: in dogfish, these are located between the red and white fibres; in thornback ray, some are dispersed within the white fibre region, while others form an intermediary layer like in dogfish. These results suggest that in the course of evolution, an amplification of the AMP-deaminase activity in muscle was coupled with increase of complexity of the muscular structure.  相似文献

The infectious agent of transmissible spongiform encephalopathies (TSE) has been considered to be PrP(SC), a structural isoform of cellular prion protein PrP(C). PrP(SC) can exist as oligomers and/or as amyloid polymers. Nucleic acids induce structural conversion of recombinant prion protein PrP and PrP(C) to PrP(SC) form in solution and in vitro. Here, we report that nucleic acids, by interacting with PrP in solution, produce amyloid fibril and fibres of different morphologies, similar to those identified in the diseased brains. In addition, the same interaction produces polymer lattices and spherical amyloids of different dimensions (15-150 nm in diameters). The polymer lattices show apparent morphological similarity to the two-dimensional amyloid crystals obtained from linear amyloids isolated in vivo. The spherical amyloids structurally resemble "spherical particles" observed in natural spongiform encephalopathy (SE) and in scrapie-infected brains (TSE). We suggest that spherical amyloids, PrP(SC)-amylospheroids, are probable constituents of the coat of the spherical particles found in vivo and the latter can act as protective coats of the SE and TSE agents in vivo.  相似文献

FMRFamide-related peptides are common to a wide variety of invertebrate species, including helminths and arthropods. In arthropods, five distinct FMRFamide-related peptide subfamilies are recognised: the myosuppressins, extended-FLRFamides, -FMRFamides, -RFamides, and sulfakinins, members of which induce potent and diverse myotropic effects. Whilst >80 FMRFamide-related peptides have been identified in nematodes, only four FMRFamide-related peptides have been characterised from flatworms. The Ascaris suum ovijector/body wall bioassay and the Procerodes littoralis muscle fibre bioassay have proved both reliable and sensitive systems for assessing the functional activities of FMRFamide-related peptides in vitro, and data describing the effects of native FMRFamide-related peptides in these systems are rapidly accumulating. This is the first study to determine the cross-phyla activities of non-native FMRFamide-related peptides in both nematode and flatworm species. In the present study, the effects of 10 arthropod FMRFamide-related peptides (leucomyosuppressin [pQDVDHVFLRFamide], schistoFLRFamide [PDVDHVFLRFamide] and truncated analogues [HVFLRFamide and VFLRFamide], lobster peptide I [TNRNFLRFamide], lobster peptide II [SDRNFLRFamide], manducaFLRFamide II [GNSFLRFamide], manducaFLRFamide III [DPSFLRFamide], calliFMRFamide 4 [KPNQDFMRFamide] and perisulfakinin [EQFDDY(SO(3)H)GHMRFamide]), representing the five subfamilies, were examined on the body wall and ovijector of the parasitic porcine nematode, A. suum and dispersed muscle fibres from the free-living turbellarian, P. littoralis. The muscle activity of the ovijector was found to be modulated significantly by each of the arthropod FMRFamide-related peptides tested; the effects were concentration-dependent, reversible and repeatable. All but one (perisulfakinin) of the 10 arthropod FMRFamide-related peptides examined modulated significantly the activity of A. suum body wall muscle. In addition, all of the arthropod FMRFamide-related peptides examined induced potent concentration-dependent contractions of P. littoralis muscle fibres. These results reveal similarities in the ligand requirement(s) between FMRFamide-related peptide receptors within the Phyla Arthropoda, Nematoda and Platyhelminthes, and indicate significant receptor promiscuity, which highlights the potential of FMRFamide-related peptide receptors as legitimate targets for novel endectocidal agents.  相似文献
