The mesoderm-derived segmental somite differentiates into dermomyotome and sclerotome, the latter of which undergoes vertebrogenesis
to spinal cartilage and ultimately to vertebral bones. However, analysis and manipulation of the developing mammalian vertebrae
in the same embryo has been infeasible because of their placental-dependent embryogenesis. Here, we report a novel culture
system of the mouse embryonic tailbud, by which the developmental processes of mammalian vertebral cartilage are traceable
and manipulatable in the same sample. The anaplastic segmental somites/sclerotomes in the tailbud of 13 gestational day (g.d.)
embryo that are structurally continuous to the vertebral column underwent progressive vertebrogenesis when (1) the ectoderm-derived
nascent epidermis was microsurgically removed prior to cultivation, and (2) the sample was incubated at the air-medium interface.
After cultivation for 5 days, the size and shape of the instructed vertebral cartilage showed features comparable to well-differentiated
body vertebra along with the expression of the cartilage marker collagen type II, suggesting that aggressive differentiation
of the sclerotomal cell lineage was achieved. In the presence of recombinant bone morphogenic protein (BMP) and Noggin, or
adenoviral particles for extracellular epimorphin, dramatic alteration of the vertebral morphology ensued in the explants.
Thus, this model system provides an approach to study the detailed molecular mechanisms of mammalian vertebrogenesis and enables
pretreatment strategies of precartilagious fragments for improving the efficacy of subsequent transplantation. 相似文献

Helianthus petiolaris andH. niveus are polytypic species which are morphologically distinct at the periphery of their ranges but intergrade in areas of sympatry.Helianthus niveus includes both annual and perennial members, whereasH. petiolaris is strictly annual. Chloroplast DNA and nuclear ribosomal DNA restriction site data were used to reconstruct the evolutionary history of populations of the two species. Cladistic analyses reveal the following: (1) neither species is monophyletic; (2) the annual habit is derived once in this complex; and (3) the region of morphological intergradation appears to be primary in origin. Bacillus circulans MTCC 7906, an extracellular alkaline protease producer was genetically characterized. B. circulans genomic DNA was isolated, oligonucleotide primers specific to alkaline protease gene of B. circulans were designed and its PCR amplification was done. The purified PCR product and pTrcHisA vector were subjected to restriction digestion with NcoI and HindIII and transformed into Escherichia coli DH5-α competent cells. The recombinant expression of alkaline protease gene studied by inducible expression and analysis by SDS-PAGE, established that the alkaline protease protein had an estimated molecular size of 46 kDa. Gene sequencing of the insert from selected recombinant clone showed it to be a 1329 bp gene encoding a protein of 442 amino acids. The sequence was blasted and aligned with known alkaline protease genes for comparison with their nucleotide and amino acid sequences. This identified major matches with three closely related subsp. of B. subtilis (B. subtilis subsp. subtilis strain 168, B. subtilis BSn5 and B. subtilis subsp. spizizenii strain W23). The insert also showed a number of substitutions (mutations) with other sp. of Bacillus which established that alkaline protease of B. circulans MTCC 7906 is a novel gene. The phylogenetic analysis of alkaline protease gene and its predicted amino acid sequences also validated that alkaline protease gene is a novel gene and the same has been accessioned in GenBank with accession number . The Phalaenopsis sp. cv. SM 9108 flower provides a good system to isolate ovule-specific genes. A cDNA library at mature ovule stage has been constructed. A differential screening approach was used to identify cDNAs representing genes which are expressed in a stage-specific manner during ovule development. The authors have demonstrated that the expression of a cDNA (0138) was regulated stage-specifically and tis-sue-specifically using, Northern blot, and also have analyzed the full sequence of this cDNA. Animals produce a wide array of sounds with highly variable acoustic structures. It is possible to understand the causes and consequences of this variation across taxa with phylogenetic comparative analyses. Acoustic and evolutionary analyses are rapidly increasing in sophistication such that choosing appropriate acoustic and evolutionary approaches is increasingly difficult. However, the correct choice of analysis can have profound effects on output and evolutionary inferences. Here, we identify and address some of the challenges for this growing field by providing a roadmap for quantifying and comparing sound in a phylogenetic context for researchers with a broad range of scientific backgrounds. Sound, as a continuous, multidimensional trait can be particularly challenging to measure because it can be hard to identify variables that can be compared across taxa and it is also no small feat to process and analyse the resulting high-dimensional acoustic data using approaches that are appropriate for subsequent evolutionary analysis. Additionally, terminological inconsistencies and the role of learning in the development of acoustic traits need to be considered. Phylogenetic comparative analyses also have their own sets of caveats to consider. We provide a set of recommendations for delimiting acoustic signals into discrete, comparable acoustic units. We also present a three-stage workflow for extracting relevant acoustic data, including options for multivariate analyses and dimensionality reduction that is compatible with phylogenetic comparative analysis. We then summarize available phylogenetic comparative approaches and how they have been used in comparative bioacoustics, and address the limitations of comparative analyses with behavioural data. Lastly, we recommend how to apply these methods to acoustic data across a range of study systems. In this way, we provide an integrated framework to aid in quantitative analysis of cross-taxa variation in animal sounds for comparative phylogenetic analysis. In addition, we advocate the standardization of acoustic terminology across disciplines and taxa, adoption of automated methods for acoustic feature extraction, and establishment of strong data archival practices for acoustic recordings and data analyses. Combining such practices with our proposed workflow will greatly advance the reproducibility, biological interpretation, and longevity of comparative bioacoustic studies. 相似文献

Acoustic signals consist of pressure changes over time and can thus be analyzed in the frequency- or in the time-domain. With
behavioural experiments we investigated which frequency components (FC) are necessary for the recognition of the periodic
envelope of the conspecific song by females of the grasshopper Chorthippus biguttulus. Further, we determined up to which frequency component phase information is required which would indicate processing in
the time domain. Responses of females revealed that signals composed of FC between 10 and 50 Hz are sufficient for recognition
of the song envelope. A systematic reduction in the number of FC showed that no single frequency component was required; signals
without the fundamental frequency were still highly attractive and only three FC may be sufficient for song recognition. Phase
changes for frequencies up to 40 Hz strongly changed the attractiveness of song signals but only little at 50 Hz. Females
were also tested with rectangular signals in which pause duration was varied. Evidently, and despite the high attractiveness
of song signals with a “missing fundamental”, females evaluated the attractiveness of signals in the time-domain, since the
selectivity for pause duration predicted the responses to signals composed from FC well. 相似文献
