Settlement of male great reed warblers in the breeding ground was highly asynchronous at Lake Biwa in Japan. It took over
1 month from the appearance of the first male to a saturation in number of males. In resource-defense-polygyny, males are
expected to try to defend as large an area as possible in the optimal habitat. In fact, a small number of the earliest settling
males divided up the breeding ground almost completely as territories among themselves and these were later reduced in size
with the addition of later settlers. The reduction was not due to a seasonal decline of aggressiveness on the part of the
owners but to a higher level of intrusion pressure by later arriving males. The neighbor-neighbor relationship, once established,
was rather stable. Home range overlap was small and territorial contacts were few between neighbors. Territorial boundaries
seem not to shift despite the addition of new males as long as the neighbors were the same. The stable relationships between
neighbors are expected to help the earlier resident to maintain his large and good territories throughout the breeding season

A mutation assay in cultured mammalian cells based on the direct analysis of minisatellite DNA was developed. Band pattern variations reflect DNA alterations ranging from single base changes to complex rearrangements. By DNA fingerprinting a large number of autosomal loci throughout the human genome can be simultaneously checked, therefore minimizing the size of the samples of cell colonies to be scored in the absence of phenotypic selection. For the mutation assay chinese hamster cells (V79) were treated with Nitrosoguanidine and 14 independent colonies were isolated and expanded. DNA fingerprints were obtained after digestion of the DNA extracted from each clone with bothHinfI andHae III, and hybridisation with both 33.15 and 33.6 probes. Twelve colonies from untreated cells were also analysed. Several differences in the band pattern of treated colonies were observed when compared with untreated cells; digestion withHae III and hybridisation with 33.15 probe allowed the detection of the highest frequency of induced variants. The results suggest that minisatellite sequences are hypermutable sites that can be used to monitor the mutagenic potential of chemical agents directly at the DNA level, without phenotypic selection. Moreover, with the method herein decribed, it is possible to distinguish between true mutations and epimutations, such as those caused by changes in DNA methylation. 相似文献

The metabolism of [3H]formate has been examined in etiolated and greening leaves of barley (Hordeum vulgare), dwarf bean (Phaseolus vulgarls), broad bean (Vicia faba) and corn (Zea mays). Tritium was extensively incorporated by primary leaves incubated for 20-min periods in light or dark. The organic acids and free amino acids were the principal products of formate metabolism but these and other products were more heavily labelled in green tissues. Time course experiments with barley leaves revealed a rapid labelling of serine, accompanied by increasing amounts of 3H in glycine and aspartate as the feeding period was extended. These amino acid products were formed throughout a 4-day greening period with an approximate doubling in total incorporation being due to large accumulations of tritiated glycine and aspartate. The involvement of tetrahydrofolate-dependent reactions in formate metabolism was indicated by inhibition of [14C] and [3H]formate incorporation by the folate antagonist, aminopterin. Labelling of glycine and serine was also strongly inhibited (up to 90%) when the leaves were incubated with increasing concentrations of isonicotinylhydrazide. 相似文献

Emphasis has increased on accuracy in predicting the effect that anthropogenic stress has on natural ecosystems. Although toxicity tests low in environmental realism, such as standardized single species procedures, have been useful in providing a certain degree of protection to human health and the environment, the accuracy of such tests for predicting the effects of anthropogenic activities on complex ecosystems is questionable. The use of indigenous communities of microorganisms to assess the hazard of toxicants in aquatic ecosystems has many advantages. Theoretical and practical aspects of microbial community tests are discussed, particularly in related to widely cited problems in the use of multispecies test systems for predicting hazard. Further standardization of testing protocols using microbial colonization dynamics is advocated on the basis of previous studies, which have shown these parameters to be useful in assessing risk and impact of hazardous substances in aquatic ecosystems. 相似文献

Leukocytes incubated with Cu(II) showed a decrease in both glutathione reductase activity and reduced glutathione content.
The glucose 6-phosphate dehydrogenase activity under the same conditions was not affected. Serum albumin added to mixtures
prevented the loss of enzyme activity, whiled-penicillamine andl-histidine had little effect. Prior oxidation of the cell-reduced glutathione did not diminish the enzyme inhibitory action
of Cu(II). The amount of regeneration of reduced glutathione in leukocytes previously treated with diamide to oxidize their
reduced glutathione was a function of Cu(II) concentration in the media. No evidence was obtained that elevated serum ceruloplasmin
levels in rabbits, nor incubation of leukocytes in vitro with ceruloplasmin, affect leukocyte glutathione reductase activity.
It was proposed that the major mechanism by which copper affects glutathione metabolism in leukocytes is by inhibition of

Mixing was studied in free water surface wetland receiving pumped river water, by measurement of the non-interacting tracer lithium. The flow pattern was found to be intermediate between plug flow and well-mixed. The nominal detention time, calculated from volume aand flow, was 50% larger than the mean tracer detention time. The peak time was found to be one-half the tracer detention time. Three models were constructed: plug flow with dispersion, tanks in series, and a series-parallel network of tanks. All proved capable of fitting the exit tracer concentration curves but the network model provided a better fit to internal measurements. Pumping frequency was high enough to allow use of an average flowrate. The degree of mixing, as characterized by the variance of the exit tracer response curve, was comparable to that found by other researchers for wetlands, ponds and rivers. 相似文献

Flow chamber observations of the filamentous pollen of Zostera marina L. (Potamogetonales) revealed that pollen rotated and moved toward inflorescences where they were captured by stigmas. The mechanics of this abiotic pollination process were examined and found to be related to the flow environment around emergent flowers. The translational movement of pollen was imparted by the advection of the fluid (e.g., pollen kinetic energy, K, ranged from 0.8 x 10-14 to 2.4 x 10-14 J, and the average K of the fluid was _ 0.7 x 10-14 J), while the rotational motion was imparted by the fluid shear stress (tau) within the velocity gradient (e.g., pollen shear stress, sigmat = omegamu where omega is the rotational velocity and mu is the dynamic viscosity, ranged from 3.4 x 10-4 to 26 x 10-4 Pa, and the average fluid shear stress was tau _ 10 x 10-4 Pa; Ackerman, 1997, American Journal of Botany 84: 1099-1109). These results indicate that there is a greater potential for pollination by filamentous pollen relative to spherical pollen. Functionally, while spherical pollen needs to be directly upstream from stigmas to be captured, filamentous pollen need only be in the vicinity of inflorescences and flowers to be captured by stigmas. Thus, in addition to direct interception on stigmas, filamentous pollen can be captured while they rotate past flowers or when they are redirected through the velocity gradient towards flowers.
