Herein, we present the solution‐state NMR studies on dextromethorphan ( 1 ) under both isotropic and anisotropic conditions. From the measurement of 22 residual dipolar couplings using a stretched polystyrene gel (PS), we show that accurate and detailed structural information is readily determined including the relative stereochemical assignments of chiral centers, validation of diastereomer configuration, and the stereospecific assignment of the seven pairs of prochiral protons. This utility of PS gels is thus showcased to obtain rapid, accurate conformational, and relative configuration information in this important class of compounds without recourse to X‐ray analysis. Plant species delimitation within tropical ecosystems is often difficult because of the lack of diagnostic morphological characters that are clearly visible. The development of an integrated approach, which utilizes several different types of markers (both morphological and molecular), would be extremely useful in this context. Here we have addressed species delimitation of sympatric tropical tree species that belong to Carapa spp. (Meliaceae) in Central Africa. We adopted a population genetics approach, sampling numerous individuals from three locations where sympatric Carapa species are known to exist. Comparisons between morphological markers (the presence or absence of characters, leaf-shape traits) and molecular markers (chloroplast sequences, ribosomal internal transcribed spacer region (ITS) sequences, and nuclear microsatellites) demonstrated the following: (i) a strong correlation between morphological and nuclear markers; (ii) despite substantial polymorphism, the inability of chloroplast DNA to discriminate between species, suggesting that cytoplasmic markers represent ineffective DNA barcodes; (iii) lineage sorting effects when using ITS sequences; and (iv) a complex evolutionary history within the genus Carapa, which includes frequent inter-specific gene flow. Our results support the use of a population genetics approach, based on ultra-polymorphic markers, to address species delimitation within complex taxonomic groups. 相似文献

Linking ecology with parasite diversity in Neotropical fishes 总被引:1,自引:0,他引:1
A comparative analysis was performed to seek large-scale patterns in the relationships between a set of fish species traits (body size, type of environment, trophic level, schooling behaviour, depth range, mean habitat temperature, geographical range, ability to enter brackish waters and capability of migration) and the diversity of their metazoan parasite assemblages among 651 Neotropical fish species. Two measurements of parasite diversity are used: the species richness and the taxonomic distinctness of a fish's parasite assemblage, including all metazoan parasites, ectoparasites only, or endoparasites only. The results showed that, on this scale, the average taxonomic distinctness of parasite assemblages was clearly more sensitive to the influence of host traits than parasite species richness. Differences in the taxonomic diversification of the parasite assemblages of different fish species were mainly related to the fish's environment (higher values in benthic–demersal species), trophic level (positive correlation with increasing level), temperature (positive correlation with temperature in marine ectoparasites, negative in endoparasites; positive for all groups of parasites in freshwater fishes) and oceanic distribution (higher values in fish species from the Pacific Ocean than those of the Atlantic). The results suggest that, among Neotropical fish species, only certain key host traits have influenced the processes causing the taxonomic diversification of parasite assemblages. 相似文献

Legumin, which amounts to approximately 55% of the seed protein in field beans (Vicia faba L. var. minor), is a representative of the 12S storage globulin family. The 12S storage globulins are hexameric holoprotein molecules composed of different types of polymorphic subunits encoded by a multigene family. Type-A legumin subunits contain methionine whereas type-B are methionine-free subunits. Sequencing of two different type A-specific cDNAs, as well as an FPLC/HPLC-based improvement of subunit fractionation and peptide mapping with subsequent partial amino-acid sequencing, permit the assignment of some of the polymorphic legumin subunits to members of the multigene family. Two different type A subunits (A1 and A2) correspond to the two different cDNA clones pVfLa129 (A2) and 165 (A1), but microheterogeneity in the amino-acid sequences indicates that polymorphic variants of both representatives of this type may exist. Two groups of published type B-specific gene sequences (LeB7, and LeB2, LeB4, LeB6, respectively) are represented by two polymorphic subunit fractions (B3I, B3II, and B4I, B4II). A seventh clone, LeB3, encodes one of the large legumin subunits that is only a minor component of the legumin seed protein complex. 相似文献

A quantitative comparison of the classifications of 24 species of male Diaspididae by principal component and principal coordinate analysis was extended to allow the comparison of classifications based on selected subsets of the 101 available characters. It was shown that subsets of 25–28 characters could be chosen numerically and used to construct classifications that resembled very closely the classification based on the reference set of characters. The conclusions are discussed in relation to other views on character-weighting and in relation to classifications of the Diaspididae based on traditional taxonomic characters provided by the females. 相似文献

Landscape genetics is a rapidly growing discipline that examines how heterogeneous landscapes and other environmental factors influence population genetic variation. We conducted a systematic review of the landscape genetic literature which demonstrates that birds are severely under‐represented relative to their species diversity and general publication prevalence. Most avian studies were on species that have relatively low dispersal ability, and we suggest that this reflects an assumed high vagility of birds that precludes spatial genetic variation relatable to landscape heterogeneity. However, spatial genetic variation exists in several bird species with very high dispersal ability, but this has not been considered in the context of landscape features. Genetic patterns may also relate to landscape due to breeding habitat selection and territorial behaviour, despite the fact that species may be able to move throughout different landscape elements with minimal movement costs. Habitat loss and fragmentation are continuing globally and are strongly related to declines in bird populations. Landscape genetic studies provide a means to understand, predict and mitigate the effects of anthropogenic landscape change on birds. This review promotes the need for landscape genetic studies of birds, such that a greater understanding of the drivers of their genetic structuring can be developed and generalizations can be made from landscape genetic studies that apply more broadly across taxa. 相似文献

Chinese Hamster Ovary [CHO] cells are the workhorse for production of modern biopharmaceuticals. They are however immortalized cells with a high propensity for genetic change. Judging from published culture records, CHO cell populations have undergone hundreds of population doublings since their origin in the late 1950s. Different cell populations were established and named from 1 to 3 decades after their generation, such as CHO-Pro–, CHO-K1, CHO-DG44, CHO-S, CHO-DUK, CHO-DXB-11 to indicate origin and certain phenotypic features. These names are commonly used in scientific publications still today. This article discusses the relevance of such names. We argue that they provide a false sense of identity. To substantiate this, we provide the long (and poorly recorded) history of CHO cells as well as their highly complex genetics. Finally, we suggest an alternative naming system for CHO cells which provides more relevant information. While the implementation of a new naming convention will require substantial discussions among members of the relevant community, it should improve interpretation and comparability between laboratories. This, in turn will help scientific communities and industrial users to attain and further the full potential of CHO cells. 相似文献

Reproductive allocation (RA) plays a vital role in the development of ecological strategies during the life cycle of plant species. Invasive alien plants (IAP) may exist at various invasion degrees across a gradient of the colonization process with several grades of relative abundances in the occupied environments. The progressive variation in the invasion degree of IAP has the potential to modify their RA strategy. This study purposes of estimating the RA strategy of the IAP Solidago canadensis L. and the correlations among RA of S. canadensis, the invasion intensity of S. canadensis, the invasiveness of S. canadensis, and the community invasibility across a gradient of invasion degrees by using the field sampling experiment. The height and relative abundance of S. canadensis did not remarkably affect its RA. The RA of S. canadensis was positively related to its reproductive biomass and total biomass. The key reason may be that plant individuals with higher total biomass can allocate more resources into sexual reproduction. The RA of S. canadensis was positively related to its invasiveness. Thus, the RA of S. canadensis may be crucial to its invasiveness. 相似文献

For endangered populations with low genetic diversity, low levels of immigration could lead to genetic rescue, reducing the risk of inbreeding depression and enhancing chances of long‐term species survival. Our genetic monitoring of Maui's dolphins revealed the first contemporary dispersal of their sister subspecies, Hector's dolphin, from New Zealand's South Island into the Maui's dolphin distribution along ~300 km of the North Island's northwest coast. From 2010 to 2012, 44 individuals were sampled within the Maui's dolphin distribution, four of which were genetically identified as Hector's dolphins (two living females, one dead female, one dead male). We also report two Hector's dolphins (one dead female neonate, one living male) sampled along the North Island's southwest coast, outside the presumed range of either subspecies. Together, these records demonstrate long‐distance dispersal by Hector's dolphins (≥400 km) and the possibility of an unsampled Hector's dolphin population along the southwest coast of the North Island. Although two living Hector's dolphins were found in association with Maui's dolphins, there is currently no evidence of interbreeding between the subspecies. These results highlight the value of genetic monitoring for subspecies lacking distinctive physical appearances as such discoveries are not detected by other means, but have important conservation implications. 相似文献
