The aim of the study was to assess the reliability and variability of stretch reflex magnitude (SRmag) in sub-acute stroke patients. For testing, rapid dorsiflexion stretches were induced 24?h apart in 22 patients and 34 controls. SRmag between sessions in patients and controls was not different and the SRmag on the more-affected side was significantly larger than the less-affected, dominant, and non-dominant sides. The SRmag was consistent between sessions. Therefore, patients were not as variable between sessions as we had hypothesized. 相似文献

Tail lesions are important pig welfare indicators that could be recorded during meat inspection as they are more visible on the carcass than on the live animal. Tail biting is associated with reduced performance in the bitten pig, but it is not clear whether problems with tail biting are reflected in general farm performance figures. Farm advisory services aim to improve farm productivity which could be associated with improvements in pig welfare. Record keeping forms an integral part of such advisory services. The aim of this study was to examine the influence of record keeping in the Teagasc eProfit Monitor (ePM herds) on the prevalence of tail lesion severity scores in Irish slaughter pigs. In addition, we investigated associations between the prevalence of tail lesion scores and production parameters at farm level in ePM herds. Pigs were observed after scalding/dehairing and tail lesion score (0 to 4), sex and farm identification were recorded. Tail lesion scores were collapsed into none/mild lesions (score ⩽1), moderate lesions (score 2) and severe lesions (score ⩾3). The effect of record keeping (ePM herd) on the different tail lesion outcomes was analysed at batch level using the events/trials structure in generalized linear mixed models (PROC GLIMMIX). Spearman’s rank correlations were calculated between average tail lesion score of a batch and production parameters. A total of 13 133 pigs were assessed from 73 batches coming from 61 farms. In all, 23 farms were identified as ePM herds. The average prevalence of moderate tail lesions was 26.8% and of severe tail lesions was 3.4% in a batch. Batches coming from ePM herds had a lower prevalence of moderate tail lesions than non-ePM herds (P<0.001). Average tail lesion score was negatively associated with age (P<0.05) and weight (P<0.05) at sale/transfer of weaners, and tended to be positively associated with the number of finishing days (P=0.06). In addition, the prevalence of severe tail lesions was negatively associated with average daily gain in weaners (P<0.05) and tended to do so with average daily gain in finishers (P=0.08). This study provides the first indication that record keeping through an advisory service may help to lower the risk of tail biting, which is associated with improved farm performance. 相似文献

Nonlethal injury is pervasive in metazoans, but surprisingly little is known about its impact upon reproductive allocation. The impact of injury on reproduction has been explored in some detail in lizards and salamanders, which have tails that are adapted for fat storage but are also injured or lost during predatory and social encounters. We synthesize diverse insights from these studies and propose new hypotheses using graphical models which highlight three distinct, hierarchical effects of injury on reproduction: reproductive inhibition, reduction in propagule number and diminished per‐propagule investment (PPI), a maternal effect. Previous studies, which involved experimentally amputating the whole tail, have provided evidence of the first two effects, although there is little evidence of reduced PPI. We assayed these effects in free‐ranging Desmognathus salamanders exhibiting naturally occurring injury. Whereas earlier studies found that tail injury prohibits reproduction (precipitating functional conflict), we found that females missing 80% of their tails, including the smallest mature individuals, still reproduce. We also detected a negative correlation between magnitude of injury and PPI, a continuous maternal effect. Continuous (graded) effects of injury on PPI have not been reported previously; neither discrete nor continuous maternal effects due to injury have been previously demonstrated in free‐ranging vertebrates. The dearth of evidence for such effects may be due to the design of experimental manipulations that use all‐or‐nothing treatments. Future studies employing quantitative field data, or more realistic experimental treatments that mimic the continuous distribution of injury are likely to detect maternal effects arising from nonlethal injury. Although our comparative and empirical findings derive from studies of lizards and salamanders, we discuss how they apply in principle to all metazoans. The high mobility group box (HMGB) 1 protein is a very abundant and conserved protein that is implicated in many key cellular events but its functions within the nucleus remain elusive. The role of this protein in replication of closed circular DNA containing a eukaryotic origin of replication has been studied in vitro by using native and recombinant HMGB1 as well as various modified HMGB1 preparations such as truncated protein, lacking its C-terminal tail, in vivo acetylated protein, and recombinant HMGB1 phosphorylated in vitro by protein kinase C (PKC). Native HMGB1 extracted from tumour cells inhibits replication and this effect is reduced upon acetylation and completely abolished upon removal of the acidic C-terminal tail. Recombinant HMGB1, however, fails to inhibit replication but it acquires such a property following in vitro phosphorylation by PKC. 相似文献

CD1 proteins are a family of major histocompatibility complex (MHC) class I-like antigen-presenting molecules that present lipids to T cells. The cytoplasmic tails (CTs) of all human CD1 isoforms, with the exception of CD1a, contain tyrosine-based sorting motifs, responsible for the internalization of proteins by the clathrin-mediated pathway. The role of the CD1a CT, which does not possess any sorting motifs, as well as its mode of internalization are not known. We investigated the internalization and recycling pathways followed by CD1a and the role of its CT. We found that CD1a can be internalized by a clathrin- and dynamin-independent pathway and that it follows a Rab22a- and ADP ribosylation factor (ARF)6-dependent recycling pathway, similar to other cargo internalized independent of clathrin. We also found that the CD1a CT is S-acylated. However, this posttranslational modification does not determine the rate of internalization or recycling of the protein or its localization to detergent-resistant membrane microdomains (DRMs) where we found CD1a to be enriched. We also show that plasma membrane DRMs are essential for efficient CD1a-mediated antigen presentation. These findings place CD1a closer to MHC class I in its trafficking and potential antigen-loading compartments among CD1 isoforms. Furthermore, we identify CD1a as a new marker for the clathrin- and dynamin-independent and DRM-dependent pathway of internalization as well as the Rab22a- and ARF6-dependent recycling pathway. 相似文献

A common objective in protein engineering is the enhancement of the thermodynamic properties of recombinant proteins for possible applications in nanobiotechnology. The performance of proteins can be improved by the rational design of chimeras that contain structural elements with the desired properties, thus resulting in a more effective exploitation of protein folds designed by nature. In this paper, we report the design and characterization of an ultra-stable self-refolding protein fiber, which rapidly reassembles in solution after denaturation induced by harsh chemical treatment or high temperature. This engineered protein fiber was constructed on the molecular framework of bacteriophage P22 tail needle gp26, by fusing its helical core to the foldon domain of phage T4 fibritin. Using protein engineering, we rationally permuted the foldon upstream and downstream from the gp26 helical core and characterized gp26-foldon chimeras by biophysical analysis. Our data demonstrate that one specific protein chimera containing the foldon immediately downstream from the gp26 helical core, gp26(1-140)-F, displays the highest thermodynamic and structural stability and refolds spontaneously in solution following denaturation. The gp26-foldon chimeric fiber remains stable in 6.0 M guanidine hydrochloride, or at 80 degrees C, rapidly refolds after denaturation, and has both N and C termini accessible for chemical/biological modification, thereby representing an ideal platform for the design of self-assembling nanoblocks. 相似文献

本文旨在观察模拟失重28 d大鼠解除尾部悬吊前、后(2 h内),清醒自由活动状态下动脉收缩压(systolic bloodpressure,SBP)、舒张压(diastolic blood pressure,DBP)和心率(heart rate,HR)的变化.采用自回归模型法对不同时间点的收缩压变异性(SBP variability,SBPV)和心率变异性(HR variability,HRV)进行自谱与互谱分析,并比较自回归法与周期图法的自谱分析结果:由传递函数分析得到反映压力感受器-心率反射敏感性(baroreceptor-heart rate reflex sensitivity,BRS)变化的有关数据.结果显示,用自回归模型法对清醒大鼠血压信号进行短时程谱分析可得到较为平滑、谱峰清楚的谱估计曲线.28 d模拟失重大鼠解除尾部悬吊前、后,SBP、DBP和HR及其主要谱指标,以及高、低频段传递函数的平均增益均无显著性变化,不同时间点的谱指标也无显著差别;但模拟失重组的SBP、DBP和HR却显著高于对照组.上述结果提示,中期模拟失重大鼠恢复正常体位后,其清醒状态的BPV与HR均处于升高状态,但其短时程BPV与HRV谱及BRS均无显著变化,与最近报道的航天员HRV与BRS无显著改变一致. 相似文献

In lizards and Sphenodon , often the fourth toes of individuals with intact tails have more subdigital lamellae on the right than on the left side, and the opposite situation frequently occurs in individuals with injured tails. The difference between intact and injured individuals in morphological directional asymmetry was statistically significant ( P <0.05) in 11.4% among 193 species from various lizard families. Lizard families varied in extent and direction of association, but no phylogenetic constraints were detected within genera. Statistical significance was greater in samples from homogenous geographic origin than from heterogeneous ones. Among gekkonid species, the difference was stronger in those with cursorial (terrestrial) habits, than in those with scansorial (rupestral or arboreal) habits. In Scincidae, loss seems more often lethal in left-footed than in right-footed individuals. Statistically significant associations between morphological left-side dominance and tail injury exist also in three independent lineages with reduced limbs (Anguidae, Scincidae and Teidae). Hence such association is probably not a result of limb function. Rather, left-side dominance seems to be the symptom of an unknown, perhaps organism-wide, detrimental trait. Polymorphism in morphological dominance existed in all species, suggesting advantages and disadvantages in different situations to both phenotypes. We propose the hypothesis that an inversion of side dominance may occur in a single trait without systematic inversion of side dominance in all traits of the body. Inversion in a single trait causes incompatibility in multiple-trait functions. Such a mechanism, rather than cultural conventions, could increase accident proneness also in left-handed Homo sapiens , and could explain increased proneness to accident and warfare mortality in left-handed men, beyond the possible involvement of cultural factors. 相似文献

Previous studies have shown no significant effect of experimentaltail length manipulation in female barn swallows (Hirundo rustica)at the beginning of a breeding season on reproductive successor behavior during that breeding season. In the present study,we investigate if tail length manipulation had any effect onreproductive performance the following year, the so-called long-termeffect, in contrast to the short-term effects already studied.We found that females with experimentally elongated externaltail feathers at the beginning of a breeding season producedless offspring during the breeding season the following yearthan did females with shortened or unmanipulated tails. Theseresults suggest that tail elongation caused flight deficienciesthat deteriorated the condition of females and eventually reducedreproductive success. The finding of long-term effects but nosignificant short-term effects for female tail elongation suggeststhat female barn swallows have the ability to adjust immediateparental investment. Detrimental effects of long tails in femalesin terms of decreased reproductive success might explain whyfemale tails are not as long as those of males. Finally, femalesmated to long-tailed (sexually attractive) males decreased theirreproductive success the following year more than did femalesmated to short-tailed males, possibly owing to differentialparental effort causing a deterioration of their condition. 相似文献
