Biologics produced from CHO cell lines with endogenous virus DNA can produce retrovirus-like particles in cell culture at high titers, and other adventitious viruses can find their way through raw materials into the process to make a product. Therefore, it is the industry standard to have controls to avoid introduction of viruses into the production process, to test for the presence of viral particles in unclarified cell culture, and to develop purification procedures to ensure that manufacturing processes are robust for viral clearance. Data have been accumulated over the past four decades on unit operations that can inactivate and clear adventitious virus and provide a high degree of assurance for patient safety. During clinical development, biological products are traditionally tested at process set points for viral clearance. However, the widespread implementation of platform production processes to produce highly similar IgG antibodies for many indications makes it possible to leverage historical data and knowledge from representative molecules to allow for better understanding and control of virus safety. More recently, individualized viral clearance studies are becoming the rate-limiting step in getting new antibody molecules to clinic, particularly in Phase 0 and eIND situations. Here, we explore considerations for application of a generic platform virus clearance strategy that can be applied for relevant investigational antibodies within defined operational parameters in order to increase speed to the clinic and reduce validation costs while providing a better understanding and assurance of process virus safety. 相似文献

Effect of different sources of dietary protein on muscle hypertrophy in functionally overloaded mice

Dietary protein intake is important for skeletal muscle protein synthesis. In this study, we investigated the differential effect of protein sources on hypertrophy of plantaris muscle induced by surgical ablation of gastrocnemius and soleus muscles. Six-week old mice were fed diets containing caseinate, whey, or soy as protein sources for 2 weeks. Plantaris muscle hypertrophy was induced by a unilateral ablation of synergistic muscles after a week. Food intake of soy protein-fed mice was higher than that of caseinate and whey-fed mice, resulting in higher body and fat weights. Plantaris muscle weight in sham-operated mice was not different across the groups. Overload-operated plantaris muscle weight and increased ratio of overloaded muscle to sham-operated muscle weights were higher in caseinate-fed mice than in whey- and soy protein-fed mice, suggesting caseinate as a promising protein source for muscle hypertrophy. 相似文献

In this study, a model synthetic azo dye (Basic red 46) bioremoval by Carpinus betulus sawdust as inexpensive, eco-friendly, and sustainable biosorbent from aqueous solution was examined in a batch biosorption system. The effective environmental parameters on the biosorption process, such as the value of pH, amount of biosorbent, initial dye concentration and contact time were optimized using classical test design. The possible dye-biosorbent interaction was determined by Fourier Transform Infrared Spectroscopy (FTIR) and Scanning Electron Microscopy (SEM). The equilibrium, thermodynamic, and kinetic studies for the biosorption of Basic red 46 onto the sawdust biomass were performed. In addition, a single-stage batch dye biosorption system was also designed. The dye biosorption yield of biosorbent was significantly influenced by the change of operating variables. The experimental data were best described by the Freundlich isotherm model and both the pseudo-first-order kinetic and the pseudo-second-order kinetic models. Thermodynamic research indicated that the biosorption of dye was feasible and spontaneous. Based on the Langmuir isotherm model, the biosorbent was found to have a maximum biosorption potential higher than many other biosorbents in the literature (264.915?mg g?1). An ultrasound‐responsive carrier for protein drugs is promising for site‐specific release of proteins at disease sites in a designated time course because ultrasound readily penetrates deep into the interior of the body in a non‐invasive way. However, the guideline for designing ultrasound‐responsive carriers that are applicable to any protein remains to be established. Here, the aim is to develop an ultrasound‐responsive material for the controlled release of a variety of proteins regardless of their charge and structure. The supramolecular polymeric hydrogel crosslinked with a host–guest interaction of β‐cyclodextrin and adamantane can enclose two kinds of model proteins and site‐specifically and stepwisely release them in an ultrasound‐guided manner without losing their activities. Furthermore, ultrasound‐guided protein delivery to living cells is achieved on model tissue consisting of cells and extracellular matrix. The results of this study provide the proof of principle that the supramolecular polymeric hydrogel is applicable as the core carrier material in an ultrasound‐guided protein delivery system. 相似文献

We appraised mating disruption (MD) to control pea moth, Cydia nigricana (Fabricius) (Lepidoptera: Tortricidae), by assessing male attraction to monitor traps, larval pod infestation, and larval age structure in pheromone‐treated and untreated grain pea fields [Pisum sativum L. (Fabaceae)], over a 5‐year period. Cellulose pheromone dispensers were manually attached to the top shoots of pea plants and released 540 mg ha?1 day?1 synthetic pheromone E8,E10‐dodecadien‐1‐yl acetate in a first test series (2000–2001) and ca. 4 200 mg pheromone ha?1 day?1 in a second series (2004–2006). The dispensers had a half‐life of about 30 days. Although male attraction to pheromone monitoring traps was largely suppressed at the edges and within MD fields in both test series, MD treatments did not reduce pod infestation in the open field in 2000 and 2001. In the 2004–2006 series, larval damage reduction was achieved in the majority of the trials but overall MD efficacy in the open field was only 61% and not significant. In contrast, in field cages placed within the experimental sites and supplied with unmated pea moths, MD control was consistently high and significant. There were no obvious differences in the larval age distribution in all MD and control treatments, suggesting that infestations started and developed further similarly. As a univoltine species, C. nigricana larvae stay in the soil of pea fields for hibernation and pupate. The following year, emerging adults disperse and fly to the closest pea crop. Combined emergence site and pea crop treatments were conducted over 2 years to include this early migration phase of C. nigricana adults. However, the emergence site treatments did not enhance MD‐control efficacy. We conclude that mating activity was only prevented in cage tests, whereas substantial mating occurred during the transit phase outside the pheromone‐treated fields either within non‐crop vegetation and/or at the edges of pheromone‐treated pea fields orientated upwind. Thus, resulting gravid female entry can be regarded as the major constraint to reliable MD control. 相似文献
