微生物多样性对土壤氮磷钾转化、酶活性及油菜生长的影响 总被引:8,自引:0,他引:8
采用灭菌土壤分别接种不同稀释倍数(1、10~(-2)、10~(-4)和10~(-6))未灭菌土壤悬浊液的方法,研究了土壤微生物多样性降低对油菜生长和养分吸收、土壤养分有效性和酶活性的影响。结果表明:(1)随着接种土壤悬浊液稀释倍数增加,油菜生物量逐渐降低,10-4的油菜生物量显著低于1和10~(-2),10~(-6)仅为1的26%;(2)油菜氮、磷和钾的吸收量与油菜生物量呈现相同的变化规律;(3)土壤铵态氮浓度随接种土壤悬浊液稀释倍数增加而降低;而土壤硝态氮则以10~(-4)为最高,其它处理间没有显著差异;土壤有效磷未发生显著变化;有效钾反而有上升趋势;(4)土壤多酚氧化酶(PhO X)活性随接种土壤悬浊液稀释倍数增加逐渐升高;β-1,4-葡萄糖苷酶(βG)活性以10~(-6)为最高,而其它处理差异不显著;土壤亮氨酸酶氨肽酶(LAP)活性和酸性磷酸酶(AP)活性变化不显著;(5)相关分析表明,油菜生物量与土壤铵态氮浓度的对数显著正相关;与多酚氧化酶、葡萄糖苷酶和亮氨酸氨肽酶活性显著负相关。研究表明,微生物多样性降低主要通过抑制土壤氮素释放影响植物生长。 相似文献

The hydraulic conductivity of a paddy field (Anthraquic Dystrustept), a silty clay soil containing more than 29% (w/w) of gravel, in Nagoya University Farm was reduced by in situ treatment of subsurface soil using bentonite and biocalcification (microbial calcium carbonate precipitation) through the addition of CaCl2, urea, and corn steep liquor (CSL). The treatment decreased the hydraulic conductivity of the field from an average of 10?3 cm/s to a range of 10?5 to 10?7 cm/s during 69 days, with reducing the proportion of pores of subsurface soil larger than 75 µm in diameter. The biocalcification effect was observed at 10-cm thickness from the treated subsurface. Laboratory soil core experiments demonstrated that the decrease in the hydraulic conductivity was not attributed to the effect of bentonite but mainly to the effect of biocalcification. The addition of CSL enhanced the urease activity of soil required for biocalcification, even at 4°C, as indicated by a decrease in urease activation energy temperature sensitivity. These experimental results agreed with the gradual decrease in hydraulic conductivity observed in the field when the average daily temperature was 7°C (days 24–69). We have used the chemically synthesized sequence of pre-pro-parathyroid hormone and several of its analogues to test the notion that the capacity of amphipathic peptides to aggregate in membranes and form ion-permeable channels correlates with their ability to function as signal sequences for secreted proteins. We found that pre-pro-parathyroid hormone (the signal sequence and pro-region of parathyroid hormone (M)), as well as some of its analogues, forms aggregates of monomers which are ion-permeable. The ion-permeable aggregates (2–3 monomers) formed by (M) are voltage-dependent and are more permeable for cations than for anions. The compounds which formed ion channels in bilayers also acted as potential signal sequences. We conclude that the ability of peptides to form ion-permeable pathways in bilayers may be correlated to their ability to function as signal peptides. 相似文献

The antifungal action of four essential oils of Foeniculum vulgare (fennel), Thymus vulgaris (thyme), Eugenia caryophyllata (Clove) and Salvia officinalis (sage) was tested in vitro against Penicillium digitatum Sacc. Direct contact and vapour phase were used to test the antifungal activity of these essential oils against P. digitatum that is responsible for green mould rot of citrus fruits. The vapour phase and direct contact of clove and thyme essential
oils exhibited the strongest toxicity and totally inhibited the mycelial growth of the test fungus. Thyme and clove essential
oils completely inhibited P. digitatum growth either when added into the medium 600 μl l−1 or by their volatiles with 24 μl per 8 cm diameter Petri dish. In in vitro mycelial growth assay showed fungistatic and fungicidal
activity by clove and thyme essential oils. Sage and fennel oils did not show any inhibitory activity on this fungus. Scanning

Peculiarities of the rat behavior were studied in a series of experimental stress models after a systemic administration of new N-uronoyl derivatives of amino acids. The psychotropic effect was shown to be determined by the nature of the amino acid fragment. N-(1,2:3,4-Di-O-isopropylidene-α-D-galactopyraneuronoyl)-glycylglycine exhibited an anxiolytic effect more pronounced than that of pyracetam, whereas N-(1,2:3,4-di-O-isopropilidene-α-D-galactopyranuronoyl)-glycylglutamic acid has antidepressant action stronger than that of amitriptyline. 根据GenBank中公布的粟酒裂殖酵母(Schizosaccharomyces pombe)N-糖酰胺酶(Png1p)cDNA序列, 设计并合成一对特异性引物, 利用RT-PCR技术从粟酒裂殖酵母中克隆出糖酰胺酶cDNA。将得到的基因克隆到表达载体pET-15b中。重组质粒转入大肠杆菌BL21(DE3)中, 经诱导表达和纯化提取后, 进行酶活测定。实验结果表明, 该酶的分子量约为39 kD, 纯化后的重组N-糖酰胺酶可以对变性处理的糖蛋白进行糖链的切除, 且这种作用需要还原剂DTT的辅助作用; N-糖酰胺酶只对错误折叠的糖蛋白有作用, 对天然的糖蛋白没有作用。等量粟酒裂殖酵母Png1p在不同温度、pH、DTT浓度和底物变性温度下对等量核糖核酸酶B(RNase B)的脱糖基化检测发现, 重组酶的最适反应温度30°C, 最适反应pH为7.0, 需要的最适DTT浓度为10 mmol/L, 底物在100°C处理10 min时酶的脱糖基化率最高。 相似文献

The fish gill is a multifunctional organ responsible for gas exchange and ionic regulation. It is hypothesized that both morphological
and functional differentiation can be found in the gills of the aquatic air-breathing fish, Trichogaster leeri. To test this, we used the air-breathing fish, Trichogaster leeri, to investigate various morphological/functional parameters. First, we evaluated the importance of performing the aquatic
surface respiration behavior in T. leeri. A reduced survival rate was observed when fish were kept in the restrained cages in hypoxic conditions. On the gross anatomy
of gills, we found evidence of both morphological and functional modification in the first and the second gills and are responsible
for ionic regulation. There were large-bore arterioarterial shunts in the fourth gill arch. It is specialized for the transport
of oxygenated blood and is less responsive to environmental stress. In addition, the anterior and the posterior gills differed
in the Na+, K+-ATPase activity upon ionic stresses. That is, only the Na+, K+-ATPase activity of the anterior two gills was up-regulated significantly in the deionized water. Lastly, we found that the
number of mitochondria-rich cells in the first and the second gills increased following ionic stress and no difference was
found in the third and the fourth gills following such an exposure. These results supported the hypothesis that there are
morphological and functional differences between anterior and posterior gill arches within the air-breathing Trichogaster leeri. In contrast, no significant difference was found among gills in gross anatomy, filament density and Na+, K+-ATPase activity in the non-air-breather, Barbodes schwanenfeldi. 相似文献

Solanum elaeagnifolium Cav. fruits contain high concentrations of steroidal saponins. Treatment of 3-day-old clover seedlings with aqueous fruit extracts modified Ca2+ uptake without significantly altering K+ and H2 PO4 − uptake. The extracts increased Ca2+ uptake in the concentration range of 0.2 to 20 m M Ca2+ . Uptake curves could be represented by two phases. In the lower phase (0.2-1.0 m M Ca2+ ), this change could be related to an increase in Vmax . Pretreatment of seedlings with saponin extracts significantly reduced ATP-dependent Ca2+ uptake and Ca2+ -dependent ATPase activity in a fraction isolated from root homogenates by centrifugation at 1500 g for 15 min. Saponins purified from S. eleagnifolium extracts by thin-layer chromatography modified in vitro the Ca2+ -ATPase activity of this fraction, indicating that the steroid may act directly on Ca2+ transport across membranes. 相似文献

Valeriana jatamansi Jones, an important medicinal herb of the Himalayan region, is an essential source of many therapeutic compounds and is traded/consumed in very high volume. The hypothesis of this study was that different seasons and light conditions may affect the content of medicinally valuable components with changes in the morpho-physiological attributes of the plant. Growing plants under suitable light conditions and harvesting of appropriate plant parts in optimum season is crucial for harnessing the full potential of the crop. Thus, the study was carried out to determine the seasonal response of V. jatamansi plants (genetically identical plants of same age) in terms of growth and phytochemical content under two different light conditions (full sunlight and 50% shade). During all seasons, growth parameters (plant height, leaf number, leaf area, relative water content, plant biomass) and the principle bioactive compounds (valerenic acid) were higher under shade conditions, while total flavonoids, tannins, phenolic compounds and antioxidant activities were higher under full sunlight conditions. HPLC analysis revealed that valerenic acid and most of the phenolic content were higher during summer season, especially in leaf part of the plant. The study suggested harvesting of V. jatamansi plants (especially leaf), during summer season to harness high quality raw material and to prevent loss of belowground parts.
