Many monitoring programs for white-tailed deer (Odocoileus virginianus) on both private and public lands across the United States have long relied on the use of road-based spotlight surveys for monitoring population size and trends. Research has suggested spotlight surveys are ineffective and that road-based surveys for deer are biased because of highly variable detection rates. To evaluate variability in detection rates relative to the assumption that repeated surveys along roads will provide reliable trend data for use in calculating deer density estimates, we collected 5 years of thermal-imager and spotlight survey data using a multiple-observer, closed-capture approach. Using a Huggin's closed capture model, data bootstrapping, and variance components analyses, our results suggest that density estimates for white-tailed deer generated from data collected during road-based spotlight surveys are likely not reflective of the standing deer population. Detection probabilities during individual spotlight surveys ranged from 0.00 to 0.80 (median = 0.45) across all surveys, and differed by observer, survey, management unit, and survey transect replicate. Mean spotlight detection probability (0.41) and process standard deviation (0.12) estimates indicated considerable variability across surveys, observers, transects, and years, which precludes the generation of a correction factor or use of spotlight data to evaluate long-term trends at any scale. Although recommended by many state, federal, and non-governmental agencies, our results suggest that the benefit of spotlight survey data for monitoring deer populations is limited and likely represents a waste of resources with no appreciable management information gained. © 2012 The Wildlife Society. 相似文献

浮游植物作为水生态系统中最重要的生物组成部分之一,对水环境敏感,在水环境监测中得到了广泛的关注。然而水生环境复杂多样,准确高效地识别浮游植物是监测工作中的一大挑战。当前浮游植物识别方法可分为经典形态学分类、分子标记和人工智能图像识别三类。前两种方法已被广泛采用,但费时费力,不利于监测机构的大规模应用和推广。同样,利用图像进行自动化分类难以在高准确率与高效率上达到平衡。深度学习技术的发展为此提供了新思路。本文提出一种新的深度卷积神经网络RAN-11。该网络以残差注意力网络Attention-56和Attention-92为基础,凭借通道对齐融合主干上的底层特征与顶层特征,通过调整注意力模块和残差快个数以精简结构,并引入了Leaky ReLU激活函数代替ReLU。以太湖11个优势属共计1036张图像为数据来源进行对比验证。除星杆藻外,RAN-11对单一优势属的的查准率都在90%以上,并且有5个优势属达到100%的查准率。RAN-11的识别准确率为95.67%,推理速率为41.5帧/s,不仅比Attention-92(95.19%的准确率,23.6帧/s)更准确,而且比Attention-56(94.71%的准确率,41.2帧/s)更快,真正兼顾了准确率与效率。研究结果表明:(1)RAN-11在查准率、准确率和推理速率上优于原始残差注意力网络,更优于以词包模型为代表的传统图像识别方法;(2)融合多尺度特征、精简网络结构和优化激活函数是提高卷积神经网络性能的有力手段。建立在经典分类基础之上,本文提出新的残差注意力网络来提升浮游植物鉴定技术,并构建出浮游植物自动化识别系统,识别准确率高、易于推广,对于实现水体中浮游植物的自动化监测具有重要意义。 相似文献

The detection, attribution and prediction of global and large scale regional change are goals for the Global Observing Systems
of the United Nations. Coastal areas are particularly sensitive to global change, but there is a variety of limitations to
universal coverage of observations. The coastal module of the Global Terrestrial Observing System (C-GTOS) considers sentinel
ecosystems to address these goals for the terrestrial, wetland and freshwater ecosystems of the coast. Sentinel ecosystems
for observing systems are a limited number of well understood systems that have substantial datasets and are observed in a
sustained fashion, forming an early warning and core system for broader regional and global change. A necessary step in the
development of C-GTOS is the examination of current definitions of coastal areas by anticipated users and information providers,
and identification of potential coastal networks and sites. We applied the sentinel system framework to the selection of C-GTOS
observation sites from several international programs using various global delineations of coastal areas. Delineations were
based on the most common definitions of the coast adopted by potential C-GTOS users and information providers, and included
mapped areas of various distance from the coastline, coastal areas of low elevation, and a seaward boundary matching the Economic
Exclusive Zone (EEZ). Decreases in the number of sites within each international program occurred with each definition marking
area closer to the coastline. The Ramsar Convention on Wetlands demonstrates the greatest percentage of coastal sites by any
definition. The process of choosing specific sentinel sites for C-GTOS continues from this initial screening, and is the next
step towards the development of an in situ site network supporting the observation of global and large scale change.

An intervening sequence (IVS) can be present or absent in the 23S rRNA of Campylobacter jejuni and Campylobacter coli. As part of a survey, we used a polymerase chain reaction (PCR) assay to detect the presence of the IVS in 43 isolates of C. coli and 82 isolates of C. jejuni. An IVS was present in 40 (93.0%) of the C. coli and only 34 (41.5%) of the C. jejuni isolates. Twelve (27.9%) of the C. coli isolates and seven (8.5%) of the C. jejuni isolates resulted in two polymerase chain reaction products, indicating heterogeneity in the presence of the 23S rRNA IVS. Fourteen of the isolates with two products were evaluated by pulse-field gel electrophoresis; 13 different patterns were observed. The total band size of one isolate was substantially greater than the expected 1.7 Mb, possibly indicating a mixed culture. Southern blot analyses demonstrated the expected three rRNA operons in all tested isolates. Nested PCR reactions with operon-specific primers followed by primers for the IVS confirmed that the strains of interest contained either one or two operons carrying the IVS and the remaining operon(s) did not. Sequence analysis of the IVS and flanking regions of the 23S rRNA genes did not discriminate C. jejuni and C. coli as distinct populations. These results indicate horizontal transfer of 23S rRNA genes or portions of the genes between C. jejuni and C. coli. Also, data showing sequence polymorphisms between the three 23S rRNA loci outside of the IVS region suggest that the isolates with intra-genomic heterogeneity appear to be members of clones that have an ancient defect in gene conversion mechanisms needed for concerted evolution of the ribosomal operons. 相似文献

The National Trust is of paramount importance in the conservation of butterflies in the UK on account of the scale of its land holding and its ability to manage habitats beneficially. Fifty two of the fifty four species currently regarded as resident in Britain occur on Trust land. The Trust has major responsibilities for the conservation of all bar one of the British rarities, and is of special significance in the conservation of the high brown fritillary and heath fritillary, two protected species. The Trust owns some 35 areas (many of which are large) of national importance for butterflies, plus much other property where the butterfly fauna is of regional importance. The Trust is implementing many dynamic projects aimed at conserving rare species, maintains a butterfly site data base and is developing a butterfly population monitoring programme. The concept of a competitive enzyme immunoassay that utilizes simultaneously the bound and the free analyte-enzyme conjugate (heterobifunctional conjugate) for signal generation in response to varying analyte concentrations in samples has been investigated. Two antigenic sites of the heterobifunctional conjugate are used in the assay for binding to immunoglobulins: the analyte derivative binds to an immobilized antibody, Ab(1), and the enzyme component binds to a spatially separated immobilized antibody, Ab(2). The analytical system is set up such that in the absence of analyte, the conjugate is predominantly bound in the compartment that contains Ab(1). With increasing concentration of native analyte in samples, an increasing concentration of native analyte in samples, an increasing amount of conjugate migrates to the second compartment that contains Ab(2). The enzyme bound in each compartment is used for signal generation. Mathematical models have been developed to determine the optimal conditions and to predict the performance of such dual-antibody systems. The theoretical predictions are supported by experimental results. The dual-antibody system has been compared with a conventional competitive enzyme immunoassay using the same reagents. 相似文献

Given their important role in neuronal function, there has been an increasing focus on altered lipid levels in brain disorders. The effect of a high-fat (HF) diet on the lipid profiles of the cortex, hippocampus, hypothalamus, and olfactory bulb of the mouse brain was investigated using nanoflow ultrahigh pressure liquid chromatography-electrospray ionization-tandem mass spectrometry in the current study. For 8?weeks, two groups of 5-week-old mice were fed either an HF or normal diet (6 mice from each group analyzed as the F and N groups, respectively). The remaining mice in both groups then received a 4-week normal diet. Each group was then subdivided into two groups for another 4-week HF or normal diet. Quantitative analysis of 270 of the 359 lipids identified from brain tissue revealed that an HF diet significantly affected the brain lipidome in all brain regions that were analyzed. The HF diet significantly increased diacylglycerols, which play a role in insulin resistance in all regions that were analyzed. Although the HF diet increased most lipid species, the majority of phosphatidylserine species were decreased, while lysophosphatidylserine species, with the same acyl chain, were substantially increased. This result can be attributed to increased oxidative stress due to the HF diet. Further, weight-cycling (yo-yo effect) was found more critical for the perturbation of brain lipid profiles than weight gain without a preliminary experience of an HF diet. The Upwey Corridor Biodiversity Project has seen substantial investment by agencies and in‐kind voluntary work by the community over a ten year period. 2008年11月至2009年11月,2010年8月至2016年8月期间,采用红外相机调查浙江凤阳山–百山祖国家级自然保护区内凤阳山片区的兽类和鸟类多样性。调查期内共布设98个不同相机位点(45个公里网格),累计28 256相机日,共拍摄到8 208张有效独立照片,鉴定为18种野生兽类和38种野生鸟类物种,分别隶属5目12科和7目16科;家畜及家禽共5种。红外相机拍摄率最高的前5种野生动物依次为小麂(Muntiacus reevesi)(拍摄率CR=11.15)、白鹇(Lophura nycthemera)(CR=2.63)、黑麂(M. crinifrons)(CR=1.03)、野猪(Sus scrofa)(CR=0.96)、猕猴(Macaca mulatta)(CR=0.59);国家I级重点保护野生动物2种,国家II级重点保护野生动物有6种;被中国脊椎动物红色名录评估为濒危(EN)、易危(VN)、近危(TN)的物种分别有2、4、9种。调查中记录到倭花鼠(Tamiops maritimus)、凤头鹰(Accipiter trivirgatus)和丘鹬(Scolopax rusticola)等15个物种为凤阳山保护区新记录种,其中斑尾鹃鸠(Macropygia unchall)为浙江省鸟类新记录物种。此外红外相机还拍摄到大量的人类活动照片,表明当地人类活动较为严重,应加强管理。调查结果提供了较为全面的凤阳山保护区兽类和鸟类的本底信息,为后续的保护管理和长期监测提供了数据支持和指导。 相似文献
