The subunit heterogeneity of the globulin fraction of sunflower seeds was investigated by two dimensional electrophoresis, using isoelectric focusing in the first dimension and sodium dodecyl sulphate polyacrylamide gel electrophoresis in the second dimension. Under non reducing conditions, intermediary subunits B, C and D (molecular weight 54 000, 48 000 and 40 000, respectively) were focused within a pI range 5.4-6.0 but intermediary subunits A (molecular weight 60 000) focused within a pI range 6.3-6.8. Under reducing conditions the electrophoretic patterns show that intermediary subunits consist in large "acidic" and small "basic" subunits linked by disulphide bonds. The large subunits of B species are more acidic and less heterogeneous than the corresponding subunits of the A species. These results confirm that helianthinin had a "legumin-type" structure.  相似文献

Sunflower ( Helianthus annuus L.) seedlings were grown in aeroponic chambers which allowed for easy access to and easy harvesting of undamaged roots. In different portions of these roots we followed the rate of ethylene production, levels of 1-aminocyclopropane-1-carboxylic acid (ACC), N-malonyl-ACC and ACC oxidase mRNA and activity of ACC oxidase. ACC oxidase was measured with an in vitro assay, ACC and N-malonyl-ACC by selected ion monitoring gas chromatography-mass spectrometry. Ethylene production was highest in the tip of the root and tower in the middle and basal (part nearest the hypocotyl) portions of the root. The levels of ACC and ACC oxidase mRNA mirrored the levels of ethylene production. The lowest quantities of N-malonyl-ACC were found in the root tips. Upon gentle transfer of seedlings from an aeroponic system to treatment tubes, ACC content transiently increased; the greatest increase occurred in the tips. This brief rise in ACC content was not correlated with an increase in ethylene production. ACC oxidase activity was lowest in the tip and higher in the middle and base; the opposite of the pattern of ethylene production. Treating the seedlings with ACC produced a rapid rise in ACC content and ethylene production and inhibited root elongation. ACC oxidase activity was not induced by ACC treatment.  相似文献

The control of ion concentration in the cytosol and the accumulation of ions in vacuoles are thought to be key factors in salt tolerance. These processes depend on the establishment in vacuolar membranes of an electrochemical H+ gradient generated by two distinct H+-translocating enzymes: a H+-PPase and a H+-ATPase. H+-lrans locating activities were characterized in tonoplast-enriched membrane fractions isolated by sucrose gradient centrifugation from sunflower ( Helianthus annuus L.) roots exposed for 3 days to different NaCl regimes. The 15/32% sucrose interface was enriched in membrane vesicles possessing a vacuolar-type H+-ATPase and a H+-PPase, as indicated by inhibitor sensitivity, pH optimum, substrate specificity, ion effects kinetic data and immunolabelling with specific antibodies. Mild and severe stress did not alter the pH profile, ion dependence, apparent Km nor the amount of antigenic protein of either enzyme. Saline treatments slightly increased K+-stimulaied PPase activity with no change in ATPase activity, while both PPi-dependent and NO3-sensitive ATP-dependent H+ transport activities were strongly stimulated. These results are discussed in terms of an adaptative mechanism of the moderately tolerant sunflower plants to salt stress.  相似文献

We have isolated two sunflower genes, Ha hsp 18.6 G2 and Ha hsp 17.7 G4, that encode small heat shock proteins (sHSPs). RNAse A protection experiments, carried out with RNA probes transcribed from each gene and hybridized to sunflower total RNA, allowed us to distinguish their mRNA accumulation patterns. In sunflower, Ha hsp 17.7 G4 mRNAs accumulated during zygotic embryogenesis at 25°C. In vegetative tissues, these mRNAs accumulated in response to either heat shock (42°C), abscisic acid (ABA), or mild water stress treatments. In all cases, the mRNAs were transcribed from the same initiation site. In contrast, Ha hsp 18.6 G2 mRNAs accumulated only in response to heat-shock. This result demonstrates differential regulation of these two sHSP genes. The complex regulation depicted by the Ha hsp 17.7 G4 promoter has been further analyzed in transgenic tobacco, using G4::GUS translational fusions. Developmental induction of Ha hsp 17.7 G4 during zygotic embryogenesis was faithfully reproduced in the transgenic plants. 5-distal sequences (between -1132 and -395) were required to confer a preferential spatial expression of GUS activity in the cotyledons. More proximal sequences (from -83 to +163) conferred to the chimeric genes most of the developmental regulation, and the responses to ABA and heat shock characteristic of the Ha hsp 17.7 G4 promoter. The water stress response of this gene was not reproduced in transgenic tobacco and, thus, could be uncoupled from its regulation during embryogenesis.  相似文献

In August 1994 verticilliosis-like symptoms were noticed in a maturing crop of sunflower at IACR-Rothamsted. Wilting plants with chlorotic areas on some leaves, and dark areas of microsclerotia at the base of stems, were first observed on 11 August. As the plants matured, dark stripes were also observed on the stems. Many stems became brittle and later collapsed. Infected xylem showed a brown discolouration, later turning to black, and microsclerotia developed. Verticillium dahliae was isolated from all parts of the plant.
In general, once symptoms were present, the incidence of verticilliosis-affected plants increased, and the disease developed, more rapidly in early maturing varieties than in the later maturing types. At harvest, incidence ranged between 24.5% and 89.8%, depending on variety. Early maturing varieties appeared to be more susceptible than later maturing types.
Isolates of V. dahliae were cultured in the laboratory. Sunflower plants were artificially inoculated with these isolates and developed verticilliosis symptoms when grown under controlled environmental conditions.  相似文献

With the ultimate purpose of clarifying the mechanism for aluminium (Al) toxicity and for Al tolerance, we tried to isolate cDNAs whose expression is induced by Al treatment and phosphate (Pi) starvation. We performed Pi starvation and Al treatment (two-step treatment) on suspension-cultured cells of Nicotiana tabacum L. cv. Samsun and then constructed a cDNA library using poly(A)+-RNA derived from the treated cells. Four independent cDNA clones (pAL 111, 139, 141 and 142) were isolated from the library by differential screening. Northern blot hybridization analysis indicated that the expression of these clones was induced by Pi starvation. Furthermore, we found that pAL 111 and pAL 142 are also induced by Al treatment. The complete cDNA sequencing of these 4 clones was determined. The results indicated that pAL111 is identical to the parA gene of N. tabacum, which is described as an auxin-regulated gene and that pAL142 is highly homologous to the parB gene of N. tabacum whose product has glutathione S-transferase (GST, EC 2.5.1.18) activity. Furthermore, we found a cysteine-rich domain in the amino acid sequence of pAL139. No DNA and deduced amino acid sequences homologous to the pAL141 were found.  相似文献

Shoot activity has been reported to affect rates of ion uptake by plant roots in other ways than merely through supply of assimilates. To elucidate the mechanisms by which a signal from the upper part of the plant controls the rate of K+ and NO3 uptake by roots, both uptake of K+ and NO3 and secretion into the xylem of young sunflower plants ( Helianthus annuus L.) were measured after changes in light intensity.
No close correlation was observed between the uptake of NO3 and that of K+; an increase in light intensity produced a much greater stimulation of NO3 uptake than of K+ uptake. On the other hand, secretion of NO3 into the xylem was tightly coupled to that of K+, and this coupling was strongly disturbed by excision of the root. The results suggest the involvement of the K2-malate shuttle on the regulation by the shoot of K+ and NO3 secretion in the xylem, which is linked to NO3 uptake, while K+ uptake is independent of this regulation mechanism.  相似文献
