We have tested the hypothesis that wall-bound calcium is redistributed from the lower to the upper sides of horizontal stems and coleoptiles during the reaction phase of gravicurvature. We used atomic absorption spectrometry to measure wall-bound calcium in the epidermal and internal layers of sunflower (Helianthus annum L.) hypocotyls and pea (Pisum sativum L.) epicotyls, and total calcium in maize (Zea mays L.) coleoptiles at the time of maximal gravireaction. In every case, we found that there was no measurable redistribution of wall-bound calcium either from the lower to the upper sides, or between epidermal and inner tissues in response to the gravistimulus. These results indicate that a redistribution of wall-bound calcium does not play a role during the gravireaction of stems and coleoptiles.  相似文献

Sweet sorghum [Sorghum bicolor (L.) Moench] is a promising non‐food energy crop. The objective of this study was to determine the economic costs and input sensitivity of sweet sorghum compared to cotton, maize, and sunflower, at two saline‐alkali sites in Shandong (Wudi County) and Inner Mongolia (Wuyuan County) provinces of China. The data were collected quantitatively based on a face‐to‐face interview with 100 and 67 sweet sorghum growers at the two sites, respectively. The sweet sorghum grown at Wudi had lower external input (5469 CNY ha?1), higher net return (7305 CNY ha?1) and benefit‐cost ratio (2.36) than its reference crop, cotton (18 454 CNY ha?1; 3615 CNY ha?1; 1.24). At Wuyuan, the sweet sorghum showed contrasting economic performance (19 541 CNY ha?1; ?3438 CNY ha?1; 0.91), which was largely because of the higher labor costs compared to sunflower (10 896 CNY ha?1; 15 133 CNY ha?1; 2.49); and maize (9108 CNY ha?1; 14 760 CNY ha?1; 2.76). The productivity of sweet sorghum per unit of external input costs (kg CNY?1) was 13.12 for Wudi and only 3.26 for Wuyuan. Based on the Cobb‐Douglas production function, the external inputs of diesel and seed had a significantly positive impact on the profitability of sweet sorghum at Wudi but not at Wuyuan. However, the costs of irrigation and film cover were significantly negative. The energy crop, sweet sorghum, showed a better return to scale on investment than cotton and sunflower.  相似文献

2,3‐Butanediol (2,3‐BDO) is a promising bulk chemical owing to its high potential in industrial applications. Here, we engineered Klebsiella oxytoca for the economic production of 2,3‐BDO using mixed sugars from renewable biomass. First, to improve xylose consumption, the xylose transporter gene (xylE) was integrated into the methylglyoxal synthase A (mgsA)‐coding gene loci, and the engineered CHA004 strain showed much faster consumption of xylose than wild‐type (WT) strain with 1.4‐fold increase of overall sugar consumption rate. To further improve sugar utilization, we performed adaptive laboratory evolution for 90 days. The evolved strain (CHA006) was evaluated by cultivating it in the media containing single‐ or mixed‐sugars, and it was clearly observed that CHA006 has improved sugar consumption and 2,3‐BDO production than those of the parental strain. Finally, we demonstrated the superiority of CHA006 by culturing in two lignocellulosic hydrolysates derived from sunflower or pine tree. Particularly, in the pine tree hydrolysate containing xylose, glucose, galactose, and mannose, the CHA006 strain showed much improved consumption rates for all sugars, and 2,3‐BDO productivity (0.73 g L?1 hr?1) increased by 3.2‐fold compared to WT strain. We believe that the engineered CHA006 strain can be a potential host in the development of economic bioprocess for 2,3‐BDO through efficient utilization of mixed sugars derived from lignocellulosic biomass.  相似文献

The objective of this work was to develop an economically relevant enzymatic process of butyl ester production using crude high-oleic sunflower oil. Novozym 435, a non-regiospecific biocatalyst, provided the best compromise between activity and butyl-ester yield. The inhibition caused by the presence of phopholipids in crude oil was eliminated by using tert-butanol. It demonstrates the key role of the medium polarity in order to insure the stability of a process. Initial substrate concentrations and their molar ratio were optimized in a continuous packed-bed reactor to maximize product yield and productivity. The best compromise was obtained for an initial oil concentration of 500 mM and a molar ratio of 5. It enabled a high productivity of 13.8 tons year−1 kg Novozym 435−1 with a butyl-ester purity of 96.5% to be obtained. Experiments with the continuous reactor were performed over 50 days without any loss of enzyme activity.  相似文献

A novel phytotoxic compound, in addition to the known phytotoxin deoxyradicinin, has been isolated from Alternaria helianthi and its structure elucidated as 3-epideoxyradicinol. * Here, we examined phenotypic differences between locally adapted serpentine and riparian populations of the serpentine sunflower Helianthus exilis from northern California, USA. * Within a common environment, plants from serpentine and riparian sites were grown in regular potting soil or serpentine soil. Physiology, morphology, phenology and fitness-related traits were measured. * Overall, riparian plants grew more rapidly, attained a larger final size, produced larger leaves, and smaller flowering heads. Riparian plants also invested less in root biomass and were more water-use-efficient than the serpentine plants. Serpentine and riparian plants also differed in leaf concentrations of boron, magnesium, sodium and molybdenum. * These ecotypic differences suggest contrasting adaptive strategies to cope with either edaphic stress in serpentine sites or intense above-ground competition at riparian sites. There was a significant population origin x soil type crossing interaction in one fitness trait (average dry weight) that mirrored local adaptation previously documented for these riparian and serpentine ecotypes. However, because all other fitness traits did not exhibit this crossing interaction in our common garden study, it is possible that phenotypic differences underlying local adaptation may be amplified in the field as a result of biotic and abiotic interactions.  相似文献

Sunflower is a major oil seed crop worldwide, and it is also an important crop in Mediterranean areas where salinity is an increasing problem. In this paper, the effect of saline irrigation water on seed yield and quality of sunflower was evaluated. A pot experiment was carried out over two crop seasons on two hybrids – a standard one (Carlos) and a high oleic one (Tenor) – submitted to five salinity levels of irrigation water (0.6, 3, 6, 9 and 12 dS m?1). Soil salinity was monitored over the entire crop cycle, and leaf ion content was determined at maturity. Tenor showed higher Na+ and Mg2+ content but lower K+ values. No difference between the two hybrids was observed for Cl? content. A progressive increase in leaf Na+, K+ and Cl? contents and Na+/K+ ratio with increasing salinity level was observed. Seed weight per head, 1000 achene weight, number of seeds per plant and oil yield significantly decreased under salt stress in both hybrids. The percent seed yield decrease was higher per unit increase in electrical conductivity of irrigation water, ECw (8%), than per unit increase in electrical conductivity of saturated‐soil extracts, ECe (5%). Concerning oil fatty acid composition, the main significant difference as result of salt stress was a progressive increase in oleic acid content, from 82.2% to 86.7% for Tenor and from 21.8% to 27.3% for Carlos, which was consistent with a decrease in linoleic acid content, from 5.9% to 3% for Tenor and from 66% to 61.3% for Carlos. These results confirm the possible inhibition of oleate desaturase under salt stress.  相似文献

Lumbrokinase (LK) is a group of serine proteases with strong fibrinolytic and thrombolytic activities and is useful for treating diseases caused by thrombus. Cholera toxin B subunit (CTB) has been widely used to facilitate antigen delivery by serving as an effective mucosal carrier molecule for the induction of oral tolerance. We investigate here the application of CTB as a transmucosal carrier in enhancing its fusion protein‐LKs effect to protect rats against thrombosis. Thus, in this study, CTB‐LK fusion gene separated by a furin cleavage site was expressed in seeds of Helianthus annuus L. The activity of recombinant protein in seeds of transgenic sunflower was confirmed by Western blot analysis, fibrin plate assays and GM1‐ganglioside ELISA. The thrombosis model of rats and mice revealed that the oral administration of peeled seeds of sunflower expressing CTB‐LK had a more significant anti‐thrombotic effect on animals compared with that administration of peeled seeds of sunflower expressing LK. It is possible to conclude that CTB can successfully enhance its fusion protein to be absorbed in rats or mice thrombosis model. The use of CTB as a transmucosal carrier in the delivery of transgenic plant‐derived oral therapeutic proteins was supported. In addition, for the purpose of that recombinant CTB‐LK was designed for oral administration, thus the expression of CTB‐LK in edible sunflower seeds eliminated the need for downstream processing of proteins. © 2014 American Institute of Chemical Engineers Biotechnol. We investigated the effects on physiological response, trace elements and nutrients accumulation of sunflower plants grown in soil contaminated with: 5 mg kg(-1) of Cd; 5 and 300 mg kg(-1) of Cd and Zn, respectively; 5, 300, and 400 mg kg(-1) of Cd, Zn, and Cu, respectively. Contaminants applied did not produce large effects on growth, except in Cd-Zn-Cu treatment in which leaf area and total dry matter were reduced, by 15%. The contamination with Cd alone did not affect neither growth nor physiological parameters, despite considerable amounts of Cd accumulated in roots and older leaves, with a high bioconcentration factor from soil to plant. By adding Zn and then Cu to Cd in soil, significant were the toxic effects on chlorophyll content and water relations due to greater accumulation of trace elements in tissues, with imbalances in nutrients uptake. Highly significant was the interaction between shoot elements concentration (Cd, Zn, Cu, Fe, Mg, K, Ca) and treatments. Heavy metals concentrations in roots always exceeded those in stem and leaves, with a lower translocation from roots to shoots, suggesting a strategy of sunflower to compartmentalise the potentially toxic elements in physiologically less active parts in order to preserve younger tissues.  相似文献
