A multi‐test screening system identified 63 fungal isolates with high in vitro biocontrol activity against Sclerotinia sclerotiorum. A bioassay method was developed, using sunflower seedlings growing in an unsterilized loam mixture. Twenty‐six isolates were tested in a series of five bioassay tests and a significant correlation (P < 0.01) was found between sclerotial infection in vitro and the number of healthy plants in vivo. Conversely, activity in an in vitro mycelial overgrowth test was not significantly correlated with activity in vivo. However, some isolates showing only mycelial activity still exerted significant disease control in both the bioassays at Littlehampton and in three additional bioassays at Sittingbourne. Only one isolate not previously reported showed significant activity in both sets of bioassays and the lack of consistency in disease control activity by all other isolates, and biocontrol agents in general, was deemed a major barrier to their use.  相似文献

The ontogenetic changes in stomatal size, frequency and conductance (gs) on abaxial and adaxial leaf surfaces of sunflower plants (Helianthus annuus L. Russian Mammoth) were examined under controlled environmental conditions. The stomatal frequency on the adaxial and abaxial leaf surfaces decreased with leaf ontogeny and insertion level. The ratio of adaxial to abaxial stomatal frequency did not change with leaf ontogeny and insertion level, and 42–44% of total stomata was apportioned to the adaxial surface. Ontogenetic changes in stomatal pore length were detected and increased with ontogenesis. The stomatal length of both leaf surfaces had linear relationships with leaf area. Ontogenetic changes in gs were similar between the two surfaces. However the adaxial gs was lower than abaxial gs in leaves of higher insertion levels. Conductance had a linear relationship with width x frequency but not with pore area.  相似文献

Mesophyll cells were isolated from sunflower leaves by an enzymic procedure. The cell suspensions possessed high photosynthesis rates. The products of cell photosynthesis were similar to the products of leaf disc photosynthesis. The relatively high radioactivity incorporated into malate after 14CO2 feeding suggests that PEP carboxylase might participate in CO2 fixation. Sunflower leaf extracts possessed a PEP carboxylase activity slightly higher than that of other C3 species. Inhibition of PEP carboxylase by maleate decreased cell photosynthesis by only 15% and the first products of cell photosynthesis were phosphorylated compounds. The extractability of chlorogenic acid from defatted sunflower seed flour in water and salt solutions at different pH values and also in aqueous organic solvents was determined. It increased with increase in pH and at pH 8 in water nearly 70% chlorogenic acid was removed in a single extraction, while NaCl did not increase the extraction, and, MgCl2 and CaCl2 increased it, especially at higher concentrations. Methanol, ethanol, isopropanol and acetone, at 20% concentration in water, caused the maximum extraction of polyphenol. These organic solvents without added water were poor solvents for the extraction of polyphenol from the flour.  相似文献

Sunflower seedlings ( Helianthus annuus hybrid Select) were grown in a complete nutrient solution in the absence or presence of Cd2+ (10 and 20 μM). Analyses were performed to establish whether there was a differential effect of Cd2+ on mature and young leaves. After 7 d the growth parameters as well as the leaf area had decreased in both mature and young leaves. Accumulation of Cd2+ in the roots exceeded that in the shoots. Seedlings treated with Cd2+ exhibited reduced contents of chlorophyll and CO2 assimilation rate, with a greater decrease in young leaves. The photochemical efficiency of photosystem II (PSII) was not altered by Cd2+ treatment in either mature or young leaves, although during steady-state photosynthesis in young leaves there was a significant alteration in the following parameters: quantum yield of electron transport by PSII (ΦPSII), photochemical quenching ( q P), non-photochemical quenching ( q NP), and excitation capture efficiency of PSII (Φexc).  相似文献

Phytotoxicity of nickel (Ni) varies within plant species and cultivars as well as with the concentration of Ni in the rooting medium. Moreover, it is known that several nutrients can modify the plant response to excess Ni. Nitrogen can be absorbed by plants as different N forms and because N metabolism and Ni are closely related, a hydroponic experiment was conducted to study the effect of Ni toxicity on the growth, nutrient status of the different plant parts and leaf chlorophyll concentrations in sunflower plants (Helianthus annuus L.) cv Quipu grown with different forms of N supply. The plants were grown under controlled conditions for 35 days. Depending on the N source supplied, there were significant differences in the sensitivity of sunflower plants to excess Ni. Tolerance was lowest when grown with NO3 alone. A high Ni and NO3 as the only N source resulted in reduced dry weight and significant decreases in nutrient concentration. Plants supplied with a mixture of NO3 and NH4 + absorbed in the presence of Ni in solution about three times less Ni than those supplied with NO3 alone. Consequently, there were great differences in Ni concentrations between treatments. With a N nutrition of 100% NO3 -N, Ni supply led to severe growth inhibition. Just contrary, simultaneous supply of NO3 and NH4 + not only reduced Ni toxicity, but growth was even stimulated by Ni if supplied to plants fed with NO3 and NH4 +. This indicates the significant role of the N form supplied in the behaviour of Ni toxicity in sunflower plants. Abstract.— Although there are many studies of the evolution and ecology of hybrid zones, few extend over long enough time periods to track evolutionary changes in the zones or assess the ultimate outcome of hybridization. Here we describe the current genetic and morphological composition of a hybrid sunflower population relative to its initial makeup 50 years ago. It appears that few genetically pure parental plants remain in the hybrid population and the average phenotype has shifted from an initial bias toward Helianthus bolanderi to a predominance of H. annuus -like plants. The similarity to H. annuus is more pronounced for morphology than for neutral genetic markers. In contrast to the shift in morphology that occurred primarily in the past 40 years, overall pollen viability increased to its current level during the first 10 to 15 years of hybridization, indicating the presence of strong fertility selection. Dramatic differences are seen in morphology, genetics, and pollen viability between the eastern and western halves of the population, thus confirming observations by previous authors and suggesting that selection pressures in the two halves differ. The trends seen in this hybrid population over the past 50 years suggest that H. bolanderi is undergoing genetic assimilation, and this trend may be representative of its fate throughout its range.  相似文献

Enhanced phytoextraction proposes the use of soil amendments to increase the heavy-metal content of above-ground harvestable plant tissues. This study compares the effect of synthetic aminopolycarboxylic acids [ethylenediamine tetraacetatic acid (EDTA), nitriloacetic acid (NTA), and diethylenetriamine pentaacetic acid (DTPA)] with a number of biodegradable, low-molecular weight, organic acids (citric acid, ascorbic acid, oxalic acid, salicylic acid, and NH4 acetate) as potential soil amendments for enhancing phytoextraction of heavy metals (Cu, Zn, Cd, Pb, and Ni) by Zea mays. The treatments in this study were applied at a dose of 2 mmol/kg(-1) 1 d before sowing. To compare possible effects between presow and postgermination treatments, a second smaller experiment was conducted in which EDTA, citric acid, and NH4 acetate were added 10 d after germination as opposed to 1 d before sowing. The soil used in this screening was a moderately contaminated topsoil derived from a dredged sediment disposal site. This site has been in an oxidized state for more than 8 years before being used in this research. The high carbonate, high organic matter, and high clay content characteristic to this type of sediment are thought to suppress heavy-metal phytoavailability. Both EDTA and DTPA resulted in increased levels of heavy metals in the above-ground biomass. However, the observed increases in uptake were not as large as reported in the literature. Neither the NTA nor organic acid treatments had any significant effect on uptake when applied prior to sowing. This was attributed to the rapid mineralization of these substances and the relatively low doses applied. The generally low extraction observed in this experiment restricts the use of phytoextraction as an effective remediation alternative under the current conditions, with regard to amendments used, applied dose (2 mmol/kg(-1) soil), application time (presow), plant species (Zea mays), and sediment (calcareous clayey soil) under study.  相似文献

Summary We describe a protocol, and several experiments that helped lead to its development, for sunflower regeneration. Important factors for sunflower regeneration were explant age, cytokinin type and concentration, basal medium, and explant source. We could not induce shoot regeneration from the explants derived from mature tissues including leaf, petiole, and stem. However, use of juvenile explants such as embryo meristem and primordial leaf tissues allowed routine regeneration of 17 different sunflower genotypes. High frequency of shoot regeneration was achieved with these explants taken from seedlings up to 5 d after germination. Explant age was less critical for embryo meristem explants than for primordial leaf tissues. Of the four basal media tested, MS and B5 media produced higher shoot-regeneration frequencies than did Anderson and woody plant media. The highest shoot-regeneration frequency was obtained with MS medium supplemented with 2 μM BA and without auxin. Addition of 1 μM naphthalene-acetic acid to the medium significantly reduced both the percentage of explants producing shoots and average number of shoots per explant. Regenerated shoots were grown to maturity in a greenhouse.  相似文献

Ophraella communa (Coleoptera: Chrysomelidae), originally from North America, has been used for biological control of common ragweed, Ambrosia artemisiifolia, in China since 2007. However, there is still a debate on whether O. communa can attack sunflowers under field conditions. To re-evaluate the biosecurity of O. communa against sunflower (Helianthus annuus), we investigated the population density of O. communa on three sunflower varieties that were intercropped with or planted in circumambience of A. artemisiifolia under field conditions. Our results showed that only very few O. communa eggs (<0.5 eggs/plant) were found on sunflower plants at the last two surveys when sunflowers were planted in circumambience of common ragweed. O. communa eggs were not found on sunflower plants at each survey when sunflowers were intercropped with common ragweed. The first–second instar larvae, third instar larvae, pupae and adults of O. communa were occasionally found on sunflower plants, but their densities were very low under either case of planting patterns. Based on these results, we conclude that sunflower is not a potential host plant for O. communa and the beetle is an effective host-specific biological control agent of common ragweed.  相似文献
