Much of eastern Australia's coastal lowlands are underlain by Holocene sulfidic sediments. Large areas have been drained for agriculture. Drained, sulfidic sediments oxidize and produce highly acidic discharge (pH<4) with significant impacts on estuarine ecosystems. The rate of production of acid from drained floodplains is between 100 to 300 kg H2SO4 /ha/y and hundreds of tonnes of H2SO4 can be discharged in a single flood from the floodplain. Generation and export of acidity is controlled by the water balance of the floodplain, the characteristics of the drainage system and the distribution of sulfides. Evapotranspiration by native plants and crops plays a dominant role in the oxidation of sediments in dry periods. In wet periods, upland discharges to floodplains dominate the water balance. Drain spacing and drain depth are critical factors in the export of acidity into coastal streams. Amelioration of acidic outflows requires an understanding of the interaction between chemical and hydrological processes in sulfidic landscapes. Redesign of drainage systems to manage surface waters and reduce drain density with the treatment of drains with lime offer promise for treating acidic discharge and reducing impacts. Reflooding of drained, partially oxidized floodplains with freshwater may not be a panacea because of the large volumes of acid stored in the soil, a lack of labile organic matter in the sediments needed to reduce sulfate and irreversible changes to the soil due to oxidation. Tidal brackish water reflooding of unproductive acidified lowlands offers promise for rehabilitating wetlands. Sulfidic wetlands which are still undrained should remain so unless all acidic discharge can be treated.  相似文献

The utilization of inorganic carbon by three species of marine diatom, Skeletonema costatum (Grev.) Cleve. Ditylum brightwellii (West) Grun., and Chaetoceros calcitrans Paulsen was investigated using an inorganic carbon isotopic disequilibnum technique and inorganic carbon dose-response curves. Stable carbon isotope data of the diatoms are also presented. Observed rates of photosynthetic oxygen evolution were greater than could be accounted for by the theoretical rate of CO2 supply from the uncatalyzed dehydration of HCO3? in the external medium, suggesting use of HCO3? as an inorganic carbon source. Data from the isotopic disequilibrium experiment demonstrate the use of both HCO3? and CO2 for photosynthesis. Carbon isotope discrimination values support the use of HCO3? by the diatoms.  相似文献

A freshwater sediment from a ditch of a peat grassland near Zegveld (Province of Utrecht, The Netherlands) was investigated for its potential methanogenic and syntrophic activity and the influence of sulfate and nitrate on these potential activities. Methanogenesis started after a 10 days lagphase. After 35–40 days aceticlastic methanogens were sufficiently enriched to cause a net decrease of acetate. In the presence of sulfate methane formation was only slightly affected. The addition of nitrate led to an outcompetion of aceticlastic methanogens by nitrate reducers. When inorganic electron acceptors were absent, substrates like propionate and butyrate were converted by syntrophic methanogenic consortia. Addition of inorganic electron acceptors resulted in an outcompetition of the syntrophic propionate and butyrate degrading consortia by the sulfate and nitrate reducers.  相似文献

Sulfur is essential for the production of certain amino acids in plants. As amino acid sulfur is the major form of sulfur in trees, there is a strong relationship between organic S and organic N in tree tissue. Sulfur deficiencies occur in parts of southeastern Australia and northwestern North America, remote from pollutant inputs. Since bilogical S requirements of forests are modest (< 5 kg · ha–1 yr–1 for net vegetative increment), however, atmospheric S inputs in polluted regions (10–80 kg · ha–1 yr–1 ) often exceed not only the forest ecosystem S requirement but also its ability to biologically accumulate S. There is some increase in the SO2– 4–S content of forest vegetation in response to elevated atmospheric S inputs, but this capacity is apparently easily saturated. 4,4′-Diisothiocyano-1,2-diphenylethane-2,2′-disulfonic acid (H2DIDS) known as an irreversible inhibitor of the anion transport in red blood cells (Cabantchik, Z.I. and Rothstein, A. (1972) J. Membrane Biol. 10, 311–330) blocks also the uptake of bile acids and of some foreign substrates in isolated hepatocytes (Petzinger, E. and Frimmer, M. (1980) Arch. Toxicol. 44, 127–135). [3H]H2DIDS was used for labeling of membrane proteins probably involved in anion transport of rat liver cells. The membrane proteins modified in vitro by [3H]H2DIDS were compared with those labeled by brominated taurodehydrocholic acid. The latter is one of a series of suitable taurocholate derivatives, all able to bind to defined membrane proteins of hepatocytes and also known to block the uptake of bile acids as well as of phallotoxins and of cholecystographic agents (Ziegler, K., Frimmer, M., Möller, W. and Fasold, H. (1982) Naunyn-Schmiedeberg's Arch. Pharmacol. 319, 254–261). The radiolabeled proteins were compared after SDS-electrophoresis with and without reducing agent present, solubilization by detergents, two-dimensional electrophoresis and after separation of integral and peripheral proteins. Our results suggest that the anion transport system of liver cells cannot distinguish between bile acids and the anionic stilbene derivative (DIDS). The labeling pattern for both kinds of affinity labels was very similar. Various combinations of separation techniques gave evidence that the radiolabeled membrane proteins are not subunits of a single native channel protein.  相似文献

A Photosystem-II (PS-II)-enriched chloroplast submembrane fraction has been subjected to non-denaturing gel-electrophoresis. Two chlorophyll a (Chl a)-binding proteins associated with the core complex were isolated and spectrally characterized. The Chl protein with apparent apoprotein mass of 47 kDa (CP47) displayed a 695 nm fluorescence emission maximum (77 K) and light-induced absorption characteristics indicating the presence of the reaction center Chl, P-680, and its primary electron acceptor, pheophytin. A Chl protein of apparent apoprotein mass of 43 kDa (CP43) displayed a fluorescence emission maximum at 685 nm. We conclude that CP43 serves as an antenna Chl protein and the PS II reaction center is located in CP47.  相似文献

Type IX collagen, a recently discovered, unusual protein of cartilage, has a segmented triple-helical structure containing interchain disulfides. Its polymeric form and function are unknown. When prepared by pepsin from bovine articular cartilage, type IX collagen was found to contain a high concentration of hydroxypyridinium cross-links, similar to that in type II collagen. Fluorescence spectroscopy located the hydroxylysyl pyridinoline and lysyl pyridinoline cross-linking residues exclusively in the high-molecular-weight collagen fraction, from which they were recovered predominantly in a single CNBr-derived peptide. The results point to a structural role for type IX collagen in cartilage matrix, possibly as an adhesion material to type II collagen fibrils.  相似文献

The putative hexose transport component of Band 4.5 protein of the human erythrocyte membrane was covalently photolabelled with [3H]cytochalasin B. Its transmembrane topology was investigated by electrophoretically monitoring the effect of proteinases applied to intact erythrocytes, unsealed ghosts, and a reconstituted system. Band 4.5 was resistant to proteolytic digestion at the extracellular face of the membrane in intact cells at both high and low ionic strengths. Proteolysis at the cytoplasmic face of the membrane in ghosts or reconstituted vesicles resulted in cleave of the transporter into two membrane-bound fragments, a peptide of about 30 kDa that contained its carbohydrate moiety, and a 20 000 kDa nonglycosylated peptide that bore the cytochalasin B label. Because it is produced by a cleavage at the cytoplasmic face and because the carbohydrate moiety is known to be exposed to the outside, the larger fragment must cross the bilayer. It has been reported that the Band 4.5 sugar transporter may be derived from Band 3 peptides by endogenous proteolysis, but the cleavage pattern found in the present study differs markedly from that previously reported for Band 3. Minimization of endogenous proteolysis by use of fresh cells, proteinase inhibitors, immediate use of ghosts and omission of the alkaline wash resulted in no change in the incorporation of [3H]cytochalasin B into Band 4.5, and no labelling of Band 3 polypeptides. These results suggest that the cytochalasin B binding component of Band 4.5 is not the product of proteolytic degradation of a Band 3 component.  相似文献

We have studied the effect of R5020, a synthetic progestin, on the biosynthesis of cellular proteins extracted from the MCF7 and T47D human breast cancer cells, using gel electrophoresis. R5020 stimulates the synthesis, as measured after [35S]-methionine labelling, and the accumulation, as shown by silver staining, of a protein of molecular weight approximately equal to 250,000. The increase of the labelled 250-kilodalton protein was rapid (3 hours) and after 3 days this protein represented approximately equal to 6% of the total cellular proteins (approximately equal to 1 microgram/150,000 cells). The induction of the 250-kilodalton protein was obtained by physiologically active concentrations of several progestins and high concentrations of 5 alpha-dihydrotestosterone but not by estradiol or dexamethasone. It was inhibited by R486 , a progestin antagonist, but not by flutamide, an androgen antagonist. These results indicate a mediation by the progesterone receptor. The 250-kilodalton protein appears to be an excellent probe to study in cell culture the mechanism of action of progestin on human cells.  相似文献
