目的:目前HBV感染动物模型各有局限,无法全面研究HBV。拟建立人HBV血清感染Babl/c乳鼠的动物模型,以便于研究HBV感染与乳鼠免疫力低下的相关性。方法:将20只Babl/c乳鼠随机分为实验组、对照组、PBS组及空白组,通过高压水动力尾静脉注射法将人HBV血清、正常人血清、PBS注入各组乳鼠体内,记录接种后乳鼠体温及体质量。于接种后第7、15、30 d采集血清标本,应用ELISA检测HBs Ag、HBe Ag的表达情况,实时荧光定量PCR检测HBV DNA浓度。结果:接种后各组小鼠体温及体质量均无明显变化。实验组中共4只乳鼠可检测到HBs Ag为阳性且维持时间长达30 d,但HBe Ag均为阴性,HBV DNA浓度均未达500 IU/ml;余下各组检测HBs Ag、HBe Ag均为阴性。结论:通过高压水动力尾静脉注射法接种人HBV血清,可成功使Babl/c乳鼠感染HBV。本实验证实乳鼠免疫力低下,HBV血清进入体内未能有效清除,对HBV存在易感性,为建立HBV感染动物模型提供实验依据,可用于研究HBV对免疫状态的影响。  相似文献

Suckling behavior of primiparae in free-ranging Japanese macaques (Macaca fuscata) was compared with that of multiparae at Jigokudani Monkey Park, the Shiga Heights, Nagano Prefecture, Japan, from April 1984 to June 1994. The estimated rates of milk secretion in a single preferred nipple among primiparae were lower than those among multiparae from birth to 5 months of age. Milk secretion capabilities of primiparous mothers, hence, appear to be inferior to those of multiparous mothers. In nutritive sucking, although Japanese macaque infants preferred one single nipple, nipple preferences in primiparae were weaker than those in multiparae. With supplementary two-nipple use during sucking, however, the infants of primiparae appear to overcome a suckling flaw of their primiparous mothers. After infants were 5 months of age and after a drop (from approximately 10% to approximately 5%) in the rates of milk secretion, however, milk secretion rates in a single preferred nipple among primiparae were similar to those in multiparae and the supplementary two-nipple use in primiparous mother-infant dyads disappeared. The nursing behaviors of two beluga whale calves were observed for the first 55 days postpartum. For both calves, the total time spent nursing in a given period peaked around 7–10 days postpartum and then declined over time. One calf, however, was found to have an unusual decline in nursing at day 35 postpartum. Ten days later, this calf was diagnosed as having a bacterial infection and given antibiotics. This suggests that early detection of health problems may be possible by observing calves' nursing behavior. For both calves, their general nursing patterns were similar, although one calf consistently nursed more than the other. Both calves nursed roughly every half hour, neither showed any signs of a circadian pattern, and carryover effects for both calves were found, indicating that there was a relationship in the overall nursing pattern from one interval to the next. Hooded seal pups are suckled for a total of four days; chimpanzees and elephants sometimes are suckled for as long as eight years. This enormous range suggests considerable diversity in reproductive strategies. Lactation represents early maternal effort, whereas weaning is the infant's transition to nutritional independence. Weaning behavior is thus important from two interconnected perspectives: that of the infant during this transition and that of the mother with the cessation of lactation and the resumption of her reproductive capacity. What controls such diversity in the duration of lactation? What are the consequences of this diversity for mothers in terms of reproduction and for infants in terms of their growth and survival? How are differences in lactation reflected in behavioral interactions? The answers to these questions involve consideration of weaning and growth, and have implications for life histories.  相似文献

The aim of this study was to investigate the effects of body condition (BC), BC changes, and plasma leptin concentrations on the reproductive performance of Lusitano broodmares on extensive systems. Data from 119 mares (ranging from 4 to 22 years of age) were collected over a period of four consecutive breeding seasons. Each case was considered as one foaled mare bred in 1 year. Body condition changes at conception (ΔBCScon) showed a strong effect on fertility at the first two postpartum estrous cycles and a significant interaction with body condition score at conception (BCScon) was observed (P < 0.01). The best fertility results were obtained with positive and greater ΔBCScon. The best predictive value of fertility (91%) was achieved when the BCScon was 3.0 and ΔBCScon was 0.375. Global foaling rate for the 4-year period was 74.5%. Mean foaling interval and gestation length were, respectively, 368.0 ± 2.8 and 340.3 ± 1.0 days. Gestation length was influenced by the month of foaling (P < 0.05) and decreased as mares got older (P < 0.01). The number of estimated estrous cycles per live foal was 1.78 ± 0.12, including foal heat ovulation. Leptin was correlated with BCS (0.41; P < 0.001), but in the present study, plasma leptin concentrations on late gestation and early lactation did not influence fertility results. At 90 days of age, lower growth performances were obtained in foals which dams presented negative BCS changes on the first 3 months of lactation (P < 0.05). In conclusion, BC changes at early postpartum period influence the reproductive efficiency of broodmares and the growth of their suckling foals. These findings are important to help breeders on management options concerning feeding strategies in the Lusitano production systems and others under similar conditions.  相似文献

The aim of this experiment was to evaluate the effect of calf management during lactation on dam performance and nursing behaviour. Thirty-six multiparous beef cows (aged 7.5 ± 0.52 years) of Parda de Montaña breed (mean live-weight (LW) at calving 566 ± 9.3 kg) with a moderate body condition (mean 2.6 ± 0.02 on a 1 to 5 scale), were assigned to three nursing frequencies from the day after parturition: Once-daily restricted nursing during a 30 min period at 0800 h (RESTR1), twice-daily restricted nursing during two 30 min periods at 0800 and 1530 h (RESTR2) and ad libitum nursing (ADLIB). Cows were fed throughout the experiment 80 MJ/day of metabolizable energy. Cow–calf behaviour recordings were performed on days 30 and 66 of lactation in each treatment. Blood samples were collected fortnightly to analyse non-esterified fatty acids (NEFA) by an enzymatic colorimetric method, and twice weekly to determine progesterone concentrations by radioimmunoassay. Cow LW losses (P < 0.05), fat-corrected milk yield (P < 0.05) and calf average daily gain (P < 0.01) during lactation were higher in ADLIB. Nursing frequency affected the mean serum concentrations of NEFA, being lower in RESTR1 (0.18 mmol/l; P < 0.05) than in RESTR2 and ADLIB cows (0.29 and 0.25 mmol/l, respectively). The highest occurrence of nursing bouts was observed in ADLIB, leading to greater nursing duration per day (not statistically compared) in this treatment (58.8 ± 5.84 min) than in their restricted nursing counterparts (22.2 ± 1.42 and 34.3 ± 1.16 min, in RESTR1 and RESTR2, respectively). The nursing system did not affect the proportion of cows cycling within 3 months post partum (58%, 46% and 55% in RESTR1, RESTR2 and ADLIB, respectively; P > 0.05). The majority of cows in all treatments showed a short oestrus cycle after first ovulation (75%, 100% and 83%; P > 0.05). In conclusion, the post-partum luteal function in cows managed under continuous access to calves was similar to restricted nursing frequencies, despite the greater suckling intensity of the formers. Restricting calf presence does not enhance the resumption of ovarian activity when suckler cows are managed in moderate nutritional conditions.  相似文献

The effect of the intensity of odour signals has rarely been investigated in the regulation of odour-guided behaviour in young mammals. This series of experiments used the mammary pheromone (MP) of the female rabbit to assess the influence of stimulus concentration on neonatal pup responsiveness. The MP is a single compound isolated from rabbit milk that releases in pups the typical head searching and oral seizing behaviour. The pups (n = 621) were exposed to graded concentrations of the MP in bioassays varying in stimulus delivery conditions. Experiment 1 demonstrated that in aqueous dilutions the MP efficiently elicits behavioural responses only within a limited range of concentrations (from 2.5 x 10(-9) to 2.5 x 10(-5) g/ml). Experiment 2 yielded the same outcome with highly purified MP delivered in dynamic conditions with a gas chromatograph. Finally, Experiment 3 used deodorized milk as the solvent of the MP; despite this change in the physico-chemical context of stimulation, similar results were reached.  相似文献

In this paper we present data from a long-term study on earlygrowth and related variables in fallow deer fawns living inlarge enclosures. Pre-winter body mass was constantly higherand more strongly correlated to subadult body mass in malesthan in females. To find out the mechanism for this higher pre-wintermass in males, we analyzed the variation in pre-winter massin relation to sex, year, mother's body mass, age and parityof mother, birth date, birth mass, growth rate, suckling behavior,and other behaviors. Birth mass was higher for male fawns, andgestation length, birth date, and weaning date did not differbetween the sexes. Consequently, both pre- and postnatal growthwere faster in males than in females. No behavioral differenceswere found between the sexes that could explain the differencein postnatal growth. Pre-winter mass was positively relatedto mother's body mass. Heavy mothers gave birth earlier andto larger offspring who grew at a higher rate, independent ofoffspring sex. However, male fawns born to primiparous mothershad relatively lower growth than male fawns born to multiparousmothers. This was not the case for female fawns. Suckling timeafter the first 2 weeks was positively related to mother's bodymass and growth of offspring. However, no measurements of sucklingbehavior differed between male and female fawns. Our results,except for the effect of parity on male and female growth, indicatethat selection has not acted on mothers to promote faster earlygrowth in males.  相似文献

The transport of vanadium across placenta and milk rats was studied by iv injection of low doses of 48V-labeled pentavanadate (0.1 micrograms V/rat) to pregnant and nursing rats. Significant concentrations of vanadium were found in the liver, intestine, and kidneys of the fetuses, showing that vanadium is capable to pass the placental barrier and, thus, being metabolized in the fetuses. Two days after injection of 0.1 micrograms V/rat as 48V-labeled vanadate to nursing rats, 34 ng V/g milk were found, decreasing to 4 ng V/g at the d 12 after dosing. The corresponding suckling rats showed a significant absorption of vanadium taken up by the milk, as suggested by the presence in their intestine, where 48V is easily absorbed in form of low molecular weight components. Vanadium in milk may be transported in the form of a biocomplex with lactoferrin, since at 12 d after injection, the 48V in the rat milk was mainly found in fractions corresponding to proteins. In weanling rats, 7 d postlactation and 18 d after administration of vanadium to the mother, only very small amounts of 48V were still present in the organs. Excretion studies on weanling rats for 7 d showed that vanadium is rapidly released mainly via feces when earlier taken up by the milk of iv injected mothers and having the same elimination pattern as the milk, which lost about 80% of its vanadium concentration after 12 d.  相似文献

Transfer of milk is the fundamental common characteristic of mammalian reproduction, but species differ considerably with respect to nursing strategies. The consequences of teat orders and allonursing have been studied intensively in domestic pigs. However, whether similar nursing strategies also exist in wild boar, the ancestor of domestic pigs is so far not known. The occurrence of allonursing in wild boar is only described anecdotally, and the question whether a teat order is established has not yet been investigated. Studying suckling behaviour of 29 piglets from six primiparous wild boar females in a semi‐natural environment we found a surprisingly fast development of a rigid teat order among littermates, which established within 2.2 hr after birth. This suggests strong fitness benefits of this phenomenon that may ultimately explain the evolution and maintenance of this trait. We further found a strong synchrony of oestrus and delivery among the females of a group, as well as the occurrence of allonursing. By determining the amount of allonursing for the first time quantitatively, we demonstrate the importance of this trait and its presumably pivotal role for juvenile survival under truly natural conditions.  相似文献
