Summary The effects of abscisic acid (ABA) on the orientation and cold stability of cortical microtubules (MTs) in epidermal cells of epicotyls of the dwarf pea,Pisum sativum L. cv. Little Marvel, were examined by immunofluorescence microscopy. The effect of ABA on the elongation of epicotyls and on the orientation of cortical MTs was opposite to that of gibberellin A3 (GA3). Treatment with ABA, which reduced the promotion of epicotyl elongation by GA3, eliminated the GA3-induced predominance of transverse MTs and resulted in a predominance of longitudinal MTs. The effect of ABA on the cold stability of cortical MTs was also opposite to that of GA3. ABA increased the cold stability of MTs, while GA3 decreased it. The predominance of longitudinal MTs brought about by ABA may have some relationship to ABA-induced inhibition of the elongation of the epicotyl. Reflexed saltmarsh-grass suspension cultures produced by seed callus were frozen to the liquid nitrogen temperature. Cooling rates, cryoprotectants and holding times were taken as a function of transfer temperatures. The highest survival of cells (45%) was found at a freezing rate of 1°C min-1, without cryoprotectant treatments. The cryoprotectants (proline, dimethyl sulphoxide, glycerol), used at different concentrations and transfer temperatures, increased the survival rate. The maximum value was 78% at 12.5% (w/v) of proline with –30°C transfer temperature. Considerable improvement of viability (from 0% to 95%) among the 12.5 and 15.0% (v/v) dimethyl sulphoxide cryopreserved cells was achieved by holding them at – 20°C for 10–30 min before plunging into the liquid nitrogen. A 20 min holding time at 15.0% (v/v) glycerol level and – 30°C transfer temperature significantly enhanced the viability of the explants from 42% to 92%. Plants were successfully regenerated from cells cryopreserved with proline (w/v) and dimethyl sulfoxide (v/v) levels of 12.5 and 15.0%, respectively.  相似文献

A model of boreal forest dynamics was adapted to examine the factors controlling carbon and nitrogen cycling in the boreal forests of interior Alaska. Empirical relationships were used to simulate decomposition and nitrogen availability as a function of either substrate quality, the soil thermal regime, or their interactive effects. Test comparisons included black spruce forests growing on permafrost soils and black spruce, birch, and white spruce forests growing on permafrost-free soils. For each forest, simulated above-ground tree biomass, basal area, density, litterfall, moss biomass, and forest floor mass, turnover, thickness, and nitrogen concentration were compared to observed data. No one decay equation simulated forests entirely consistent with observed data, but over the range of upland forest types in interior Alaska, the equation that combined the effects of litter quality and the soil thermal regime simulated forests that were most consistent with observed data. For black spruce growing on permafrost soils, long-term simulated forest dynamics in the absence of fire resulted in unproductive forests with a thick forest floor and low nitrogen mineralization. Fires were an important means to interrupt this sequence and to restart forest succession.  相似文献

The effects of light-induced non-photochemical quenching on the minimal Fo, and variable Fv, fluorescence emissions at 690 and 730 nm in leaves were determined. Non-photochemical quenching of Fo, but not Fv, was found to be dependent upon the wavelength of emission, and was greater at 690 nm than at 730 nm. For emission at 730, compared to at 690 nm, approx. 30% of Fo was not affected by non-photochemical quenching processes in leaves of C3 plants; in maize leaves this was found to be approx. 50%. The data indicate that a substantial proportion of the pigments contributing to Fo emission at 730 nm are not quenched by light-induced, non-photochemical quenching processes and that there are large differences in the pigment matrices contributing to Fo and Fv emissions at 730 nm, compared to those at 690 nm. These findings have important implications for the accurate estimation and interpretation of non-photochemical quenching of fluorescence parameters and their use in the calculation of photochemical efficiencies in leaves. Measurements of fluorescence emissions at wavelengths above 700 nm are likely to give rise to significant errors when used for determinations of photochemical and non-photochemical quenching parameters.  相似文献

Membrane penetration depth is an important parameter in relation to membrane structure and organization. A methodology has been developed to analyze the membrane penetration depths of fluorescent molecules or groups utilizing differential fluorescence quenching caused by membrane embedded spin-label probes located at different depths. The method involves determination of the parallax in the apparent location of fluorophores, detected when quenching by phospholipids spin-labelled at two different depths is compared. By use of relatively simple algebraic expressions, the method allows calculation of depth in å. This method has been used to determine the location of fluorophores in NBD-labelled lipids and anthroyloxy-labelled fatty acids in model membranes and of the membrane embedded tryptophan residues in the reconstituted nicotinic acetylcholine receptor.  相似文献

本文研究了大蟾蜍心肌和腓肠肌肌球蛋白钙激活ATP酶的活性。实验结果显示,大蟾蜍心肌和腓肠肌肌球蛋白钙激话ATP酶活性具有不同的季节变化规律。并且这种酶活性的季节变化不受人工低温环境因素的影响。此外,大蜍蟾心肌中ATP酶的活性始终高于腓肠肌的。上述酶活性的变化均具有一定的生理意义。  相似文献

 用硫酸铵分段盐析、超滤膜分级分离及DEAE-纤维素、Sephadex A-25和Sephadex G-50三种柱层析方法从双胸蚓组织的粗提取液中分离纯化出一种纤溶酶,分子量为29kD,由一条肽链组成。此晦具有强烈的溶解纤维蛋白的作用,对家兎实验性血凝块也具有明显的溶解作用。此酶的最适pH为8.0,在pH7.6~8.4之间活力相差不到2%;酶在PH4.7—11.0范围内稳定;酶作用的最适温度为57℃;此酶热稳定性较好,于25~50℃保温3小时,酶活力基本不变,60℃时,活力保留65%。金属离子Na~(+)、K~(+)、Mg~(2+)等可提高此酶的活力,而Hg~(2+)、Ca~(2+)等金属离子对此酶有不同程度的抑制作用。  相似文献

在低温吸胀阶段,经PVA(聚乙烯醇)和PEG(聚乙二醇6000)预处理的大豆胚轴蛋白质合成和ATP含量均比对照高。在萌发阶段,胚轴生长增快,蛋白质合成明显加快,ATP迅速被消耗,而对照胚轴则相反。试验结果表明,预处理大豆种子萌发和生长与其蛋白质合成、ATP水平和消耗能力有密切关系。  相似文献

The 1st feature revealed typical for diapause phenomena inTrichogramma species is the interaction of environmental conditions in both parental and filial generations in induction of diapause in the latter. Lowered temperature during larval development is the ultimate factor evoking diapause in pronymphs but the norm of this thermal reaction is not fixed but varies depending on photoperiod and temperature in the previous generation due to maternal influence. Short day and in some cases high temperature in parental generation enhance tendency to diapause in the progeny. Unlike maternal influence the development to filial generation itself is not (or almost is not) governed by its own photoperiodic reaction. The 2nd typical feature revealed is the occurrence of endogenous process running in the sequence of generations and causing changes in the diapause tendency and underlying reaction norms even under constant rearing conditions.   相似文献

前文由柑桔枝条在不同低温下、不同冷冻时间的电解质外渗测定,提出胁强(stress)、作用时间与胁变(strain)之间关系的数学模型。在这个模型中共有3个参数:屈服点温度(yield point temperature),胁强敏感度(stress sensitivity)和作用时间敏感度(sensitivity to duration),用以描述植物的抗性。抗性强的植物应表现为屈服点温度较低,胁强敏感度或者时间敏感度较低。为验证此数学模型,本工作以经冷锻炼与未经冷锻炼的盆栽柑桔枝条为材料,作不同温度与时间处理的电解质外渗率的测定,研究了冷锻炼对于上述3个参数的影响。发现胁强敏感度和屈服点温度受冷锻炼影响而下降,时间敏感度未表现明显变化。对于田间柑桔、油桐与毛竹的定期测定,在固定冷冻时间下,得到了类似于盆栽柑桔的结果。入冬时,植物抗冻性提高,3种植物都表现出下列两种变化:1.胁强敏感度的明显下降;2.屈服点温度和/或时间敏感度亦下降。开春时的变化则相反。胁强敏感度的变化与后一种变化有各自的规律,且因植物种类而不同。拐点胁强(stress at inflection point)具有与半致死温度(50%killing point temperature)不同的意义,它的变化是上述两种变化的综合结果。本试验结果表明,冷锻炼对于植物胁强敏感度有明显影响,用本数学模型的3个抗性指标描述  相似文献
