The involvement of ethylene in shoot formation in vitro was studied in one year old lavandin (Lavandula officinalis Chaix x Lavandula latifoliaVillars) callus. A peak in ethylene evolution characterized thenon-regenerating leaf callus line, as compared to the shoot-forming calyxcallus line, on the growth medium provided with 2,4-dichlorophenoxyacetic acid (1 mg dm-3) and kinetin (0.5 mg dm-3). After one year in culture, calyxcallus attained the capacity to grow on auxin-reduced media, showing decreased ethylene production and faster shoot bud emergence, when transferred onto the regeneration medium, supplemented with 10 mg dm-3 benzyladenine. Shoot formation was also inhibited by addition of the ethylene precursor 1-aminocyclopropane-1-carboxylic acid, indicating an involvement of ethylene in the failure of regeneration. Abhibition of the accumulation of anthocyanin but not of growth in wild carrot cultures was achieved using 2-amino-oxyphenylpropionic acid or phenylpropiolic acid. The concentration of inhibitor required to achieve this differential effect increased with increased culture density.By several additions of inhibitor at differential times, anthocyanin accumulation in the parental culture and in high-and low-acculmulating subclones could be completely inhibited. Under these conditions the level of anthocyanin in the cultures remained unchanged. These data show that the accumulated anthocyanin is not metabolized and that differences in the rate of degradation of anthocyanin does not account for the differences in anthocyanin accumulation by the subclones examined.  相似文献

The effects of a 50-Hz extremely low frequency magnetic field on cultured K562 cells growing in suspension were studied by means of scanning electron microscopy and electron paramagnetic resonance spectroscopy. Exposure of K562 cells at 2.5 mT for periods to 96 hours induced significant changes in cell-surface structure and physiology without modification of proliferative capability as indicated by quantitative analysis. Thus extremely low frequency fields seem able to induce injurious, sublethal cell alterations, and the plasma membrane seems to play an important role in this effect. © 1993 Wiley-Liss, Inc.  相似文献

No cardenolide was detected in cultured cells and roots of root forming callus of Digitalis purpurea under such conditions as addition of auxins, varied sugar concentration of the medium and illumination. Cardenolides such as digitoxin and purpurea glycoside A were found in leaves from the regenerated shoot from callus cultures. The manifestation of cardenolide-synthesizing potentiality seems to be closely related to some process of leaf development.  相似文献

1. While phosphorus (P) is often considered the most important growth limiting factor for plants in lakes, recent studies of shallow lakes indicate that nitrogen (N) may be of greater importance than realized hitherto and that submerged macrophytes may be lost when the N concentration exceeds a certain threshold, as long as the concentration of P is sufficiently high. 2. We studied the effects of different loadings of NH4‐N and NO3‐N on chlorophyll a and on a macrophyte tolerant of eutrophication, Vallisneria spinulosa (Hydrocharitaceae). In outdoor mesocosms we used water from a pond as control and created four levels of NH4‐N and NO3‐N (approximately 2.5, 5, 7.5 and 10 mg L−1) by dosing with NH4Cl and NaNO3, respectively. After the experiment, the plants were transferred back to a holding pond to study their recovery. In contrast to previous research, we used a low background concentration of phosphorus (TP 0.024 ± 0.003 mg L−1) so we could judge whether any effects of N were apparent when P is in short supply. 3. Chlorophyll a increased significantly with N dosing for both forms of N, but the increase was highest in the NH4‐N dosed mesocosms (maximum 58 μg L−1 versus 42 μg L−1 in the NO3‐N mesocosms), probably due to a higher total inorganic N concentration (part of the added ammonia was converted to nitrate during the experiment). 4. Although the number of ramets of V. spinulosa was not affected by the N treatment, the biomass increased up to concentrations of 7.5 mg L−1, while biomass at 10 mg L−1 remained at the control level for both N ions treatments. A similar pattern was apparent for the content of N and soluble sugar of the plant, while there were no differences in the plant P content among treatments. Five months after transplantation back to the pond no difference was found in the number of ramets or in biomass, except that the biomass of plants grown at 10 mg N L−1 during the experiment was greater than that in the control, while the N and P contents of plants were similar to those of the controls. 5. Nitrogen concentration had little influence on the growth of the eutrophication tolerant submerged macrophyte at moderately low concentrations of phosphorus. Moreover, the two N ions showed no toxic effects, suggesting that loss of macrophytes observed in other studies, run at higher phosphorus concentrations, was probably related to enhanced shading by periphyton and/or phytoplankton rather than to any toxic effects of N.  相似文献

Marine benthic diatoms growing in biofilms on sediment surfaces generally occur associated with heterotrophic bacteria, whereas modern molecular techniques and analyses of species‐specific physiology create a demand for axenic cultures. Numerous benthic diatoms were isolated from surface sediments during a monitoring of the Solthörn tidal flat (southern North Sea, Germany) from May 2008 to May 2009. Of these, around 50% could be purified from the accompanying heterotrophic bacteria using different antibiotics combined with physical separation methods (vortexing, ultrasound). Overall, seven different antibiotics were tested at different concentrations, and a best working protocol was developed. The axenic strains were stable on average for only around 15 months, indicating a symbiotic interaction between the benthic diatoms and the associated bacteria. While most short‐term effects during the purification process were restricted to differences in growth rates among xenic and axenic diatom strains, long‐term cultivation led to distinct changes in cell volumes and growth characteristics of the axenic strains.  相似文献

Stem cells are promising cell sources for many biomedical applications including cell therapy, regenerative medicine, and drug discovery. However, the commonly used static tissue culture vessels can only generate a low number of cells. To provide an adequate number of stem cells for clinical applications, a scalable process based on bioreactors is needed. Stem cells can be either cultured as free cells/aggregates in suspension or as adherent cells on the solid substrates. Based on the cell property, different bioreactor configurations are developed to better expand stem cells while maintaining their differentiation capacity. In this review, several major types of bioreactor systems and their applications in stem cell engineering are discussed. Continued advancements in bioprocess and bioreactor research and development are important to engineer stem cells for their use in biomedical applications.  相似文献

Bortezomib (BTZ) is the first proteasome inhibitor entered in clinical practice. Peripheral neuropathy is likely to be a class side effect of these drugs, although its severity is largely variable, and it deserves to be further investigated, since the mechanisms of BTZ-induced peripheral neurotoxicity (BiPN) are still unknown.

In our study, we investigated in vivo and in vitro possible pathogenic events relevant to BiPN using a well-established rat model, with particular reference to the extent of proteasome inhibition and the effects on α-tubulin polymerization in sciatic nerves and dorsal root ganglia specimens obtained from animals treated with chronic regimens at a dose of 0.2 mg/kg intravenously. The same assessments were also performed after a single injection. Moreover, these studies were replicated in vitro using embryonic DRG neurons exposed to 100 nM BTZ and adult DRG neurons exposed to 10–50 nM BTZ for 24 h and 48 h.

A significant increase in the polymerized fraction of α-tubulin and prolonged proteasome inhibition were observed after the chronic BTZ treatment in vivo. Recovery to physiological levels was observed after a 4-week follow-up post-treatment period. Proteasome inhibition and increased α-tubulin polymerization were also observed following BTZ treatment of both embryonic and adult DRG neurons in vitro.

Our in vivo results suggest that proteasome inhibition and alteration of tubulin dynamics contribute to BiPN. The in vitro systems here described reliably replicate the in vivo results, and might therefore be used for further mechanistic studies on the effects of proteasome inhibitors on neurons.  相似文献
