In vitro propagation of AM fungi using transformed root cultures (TRC) is commonly used to obtain pure AM fungal propagules for use in research and industry. Early observations indicate that such an artificial environment can alter traits and function of AM fungi over time. We hypothesized that increased in vitro cultivation may promote ruderal strategies in fungi by enhancing propagule production and reducing mutualistic quality. To examine the effect of in vitro cultivation on the trait and function of AM fungi, we inoculated plants with 11 Rhizoglomus irregulare isolates which fell along a cultivation gradient spanning 80 generations. We harvested plants at 10, 20 and 30 d post inoculation to observe differences in fungal and plant traits post infection. In vitro cultivation led to increased spore production but reduced plant shoot phosphorus. Our results indicate that in vitro propagation may indirectly select for traits that affect symbiotic quality.  相似文献

Bone marrow stroma contains mesenchymal stem cells (MSC) which are progenitor cells, at least for tissues arising from mesechyma. The study of MSC biology yields controversial data. Therefore further experiments are needed to characterize these cells. The aim of our research was to compare primary cultures and subcultures of stromal precursor cells isolated from rat bone marrow. Long-term cultures of these cells isolated from 5 animals have been obtained. Morphological, immunophenotypic, and functional (capacity to osteogenic differentiation) characteristics of the cells have been investigated. We show that the cell morphology in the cultures is highly heterogenic. Morphological cell types are described. Heterogeneity of stromal cells declines on late passages. Cell cultures isolated from different animals have the same immunophenotypic markers (CD90, CD44, CD54, CD106, CD45, CD11b) but different morphological characteristics and a different capacity to osteogenic differentiation during long-term cultivation. The data show that more specific markers and functional tests should be applied to identify MSC.  相似文献

A 3-year study was done to quantify the biomass of submerged aquatic vegetation (SAV) and its relationship with environmental attributes in Lake Okeechobee, the largest lake in the southeastern United States. Plants were sampled on 21 occasions at sites located along 15 fixed transects around the shoreline, giving rise to 721 observations of SAV species (Chara spp., Vallisneria americana, Hydrilla verticillata, Potamogeton illinoinensis) dry weight biomass. Environmental sampling focused on factors that attenuate light, including phytoplankton chlorophyll a (chl a), total suspended solids (TSS), non-volatile suspended solids (NVSS) and color. Depth and Secchi transparency also were measured. Based on regression analysis, NVSS was considerably more important in attenuating light than chl a or color. Total biomass of SAV varied from 0 to 271 g dw m–2, with a mean of 4.7 g dw m–2, and strong dominance by Chara. The SAV biomass was lower than average for Florida lakes, and may reflect the influence of suspended solids on underwater irradiance, as well as high water level in the late 1990s. Dense SAV was found only where depth was < 2 m and TSS < 20–30 mg l–1. At locations where high biomass of SAV occurred, the plants may have influenced water quality, because concentrations of TSS, NVSS, and chl a were 2–3 fold lower than at sites with no plants. The potential effects of SAV also were apparent at a regional scale. The shoreline region of the lake displayed a pattern of rising and falling chl a and NVSS with water depth. This occurred both at sites with and without plants, suggesting that it may be driven by physical processes, such as water circulation patterns, which are influenced by depth. However, the pattern was dampened at sites with SAV, indicating a potential to influence these attributes of water quality.  相似文献

In axenic Chlorella pyrenoidosa Chick cultures, extracellular release was linear with time, but plateau-type curves were obtained in cultures with added bacteria. Initial rates of excretion were identical in both, systems. Kinetics of extracellular release in axenic Anabaena flos-aquae (Lyng.) Bréb. cultures were more complex than in Chlorella but the initial excretion rates were identical in axenic and mixed algal-bacterial cultures. In lakewater, extracellular release kinetics resemble the pattern in mixed Chlorella-bacteria cultures. An explanation is an initial lag in bacterial utilization of algal extracellular products. As a result, both in situ and in the laboratory, consecutive short, experiments give higher excretion rates than single long incubations. It is suggested that the former are close to total or gross extracellular release rates whereas the latter give net values, detecting only substances not, removed by heterotrophs.  相似文献

The quantitative effects of pH, temperature, time of fermentation, sugar concentration, nitrogen concentration and potassium ferrocyanide on citric acid production were investigated using a statistical experimental design. It was found that palmyra jaggery (sugar syrup from the palmyra palm) is a suitable substrate for increasing the yield of citric acid using Aspergillus niger MTCC 281 by submerged fermentation. Regression equations were used to model the fermentation in order to determine optimum fermentation conditions. Higher yields were obtained after optimizing media components and conditions of fermentation. Maximum citric acid production was obtained at pH 5.35, 29.76 °C, 5.7 days of fermentation with 221.66 g of substrate/l, 0.479 g of ammonium nitrate/l and 2.33 g of potassium ferrocyanide/l.  相似文献

Fourteen monosporal isolates of Hirsutella thompsonii grew vegetatively in various liquid media producing typical phialidic-like conidiophores. Hirsutella thompsonii var. synnematosa from Ivory Coast (HtIC) was the only pathotype which produced true conidia in submerged culture. HtIC began producing conidia after 3 days incubation reaching a peak ranging from 6.8 × 105 to 9.7 × 107 conidia/ml between 6 and 11 days. A concentration of 10 g/liter of corn steep liquor and 0.2% Tween 80 were essential for maximum conidiation. Submerged conidia had a smooth but somewhat rugose conidial walls, whereas aerially formed conidia were distinctly verrucose. Germination of submerged conidia ranged from 5.2 to 12.9% and were virulent causing 32.5% infection to adult citrus rust mites when sprayed on citrus foliage at 1.2 × 109 conidia/ml.  相似文献

Abstract Spontaneous nitrogenase-negative (Nif mutants of Rhodopseudomonas capsulata were observed to accumulate with time in ammonium- or glutamate-limited chemostat cultures.
Nif mutants were characterized by their inability to grow under N2 and to reduce acetylene or produce hydrogen gas when grown on glutamate. They lacked the nitrogenase structural proteins as evidenced by immunological techniques. On the other hand, no significant differences were found in the pathways of ammonia assimilation between the Nif mutants and the wild-type strain. The Nif mutants seem to result from a mutation in a regulatory gene.  相似文献

以外来种伊乐藻与土著种苦草为实验生物,通过室外受控实验研究了基质类型对2种水生植物种间关系的影响.结果表明:伊乐藻在2种不同营养水平的基质中均具有明显竞争优势.植物竞争(混栽伊乐藻)对低密度苦草的生长影响不明显,对密度高苦草抑制显著,然而基质类型对低密度苦草的生长影响显著,对高密度苦草无明显影响.同样,植物竞争(混栽苦草)对低密度伊乐藻的生长无明显影响,对密度高伊乐藻抑制显著,然而,无论种植密度高低,基质类型对伊乐藻的生长均有极大影响.本文还对伊乐藻与苦草的种间竞争关系机理进行了探讨,其研究结果对分析外来种入侵后水生植被的演变规律以及湖泊水生植被的管理有一定参考价值.  相似文献
