Intracanopy variation in net leaf nitrogen (N) resorption and N cycling through leaves in mature walnut (Juglans regia L. cv Hartley) trees were monitored in 3 different years. Differential irradiance among the spurs sampled was inferred from differences among leaves in dry weight per unit area (LW/LA) which varied from 4.0 mg · cm–2 to 7.0 mg · cm–2 in shaded (S) and exposed (E) canopy positions, respectively. Our results, using 15N-depleted (NH4)2SO4 validated the concept that N influx and efflux through fully expanded leaves occurred concurrently during the period of embryo growth. Additionally, it also suggested that N influx into leaves was substantially greater in exposed as compared with shaded canopy positions. Because of its well documented phloem immobility, leaf Ca accumulation was used to better estimate the relative influx of N into exposed and shaded leaves. N cycling varied locally within the tree canopy, i. e. Ca (and presumably N) influx was 100% greater in exposed than shaded tree canopy positions, but influx was not influenced significantly by the proximity of developing fruit. In contrast, both the amount and percentage N efflux was significantly greater during embryo growth in fruit-bearing than defruited spurs. Net leaf N resorption averaged 2–4 times greater (25–30%) in fruit-bearing spurs than the 5–10% decrease in the leaf N content in defruited spurs. Since about 90% of leaf N content reportedly occurs as protein, fruit N demand apparently influenced protein turnover and catalysis in associated spur leaves. The amount of leaf N resorption was greater in exposed than shaded positions in the tree canopy in 2 of the 3 years of data collection. Our data show that like leaf N content, N influx, N efflux and net leaf N resorption vary throughout mature walnut tree canopies under the combined local influences of fruiting and irradiance. 相似文献

Stability of wild-type and mutant RTEM-1 beta-lactamases: effect of the disulfide bond 总被引:6,自引:0,他引:6
Uniquely among class A beta-lactamases, the RTEM-1 and RTEM-2 enzymes contain a single disulfide bond between Cys 77 and Cys 123. To study the possible role of this naturally occurring disulfide in stabilizing RTEM-1 beta-lactamase and its mutants at residue 71, this bond was removed by introducing a Cys 77----Ser mutation. Both the wild-type enzyme and the single mutant Cys 77----Ser confer the same high levels of resistance to ampicillin in vivo to Escherichia coli; at 30 degrees C the specific activity of purified Cys 77----Ser mutant is also the same as that of the wild-type enzyme. Also, neither wild-type enzyme nor the Cys 77----Ser mutant is inactivated by brief exposure to p-hydroxymercuribenzoate. However, above 40 degrees C the mutant enzyme is less stable than wild-type enzyme. After introduction of the Cys 77----Ser mutation, none of the double mutants (containing the second mutations at residue 71) confer resistance to ampicillin in vivo at 37 degrees C; proteins with Ala, Val, Leu, Ile, Met, Pro, His, Cys, and Ser at residue 71 confer low levels of resistance to ampicillin in vivo at 30 degrees C. The use of electrophoretic blots stained with antibodies against beta-lactamase to analyze the relative quantities of mutant proteins in whole-cell extracts of E. coli suggests that all 19 of the doubly mutant enzymes are proteolyzed much more readily than their singly mutant analogues (at Thr 71) that contain a disulfide bond.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献

1. For north temperate lakes, the well‐studied empirical relationship between phosphorus (as measured by total phosphorus, TP), the most commonly limiting nutrient and algal biomass (as measured by chlorophyll a, CHL) has been found to vary across a wide range of landscape settings. Variation in the parameters of these TP–CHL regressions has been attributed to such lake variables as nitrogen/phosphorus ratios, organic carbon and alkalinity, all of which are strongly related to catchment characteristics (e.g. natural land cover and human land use). Although this suggests that landscape setting can help to explain much of the variation in ecoregional TP–CHL regression parameters, few studies have attempted to quantify relationships at an ecoregional spatial scale. 2. We tested the hypothesis that lake algal biomass and its predicted response to changes in phosphorus are related to both local‐scale features (e.g. lake and catchment) and ecoregional‐scale features, all of which affect the availability and transport of covarying solutes such as nitrogen, organic carbon and alkalinity. Specifically, we expected that land use and cover, acting at both local and ecoregional scales, would partially explain the spatial pattern in parameters of the TP–CHL regression. 3. We used a multilevel modelling framework and data from 2105 inland lakes spanning 35 ecoregions in six US states to test our hypothesis and identify specific local and ecoregional features that explain spatial heterogeneity in TP–CHL relationships. We include variables such as lake depth, natural land cover (for instance, wetland cover in the catchment of lakes and in the ecoregions) and human land use (for instance, agricultural land use in the catchment of lakes and in the ecoregions). 4. There was substantial heterogeneity in TP–CHL relationships across the 35 ecoregions. At the local scale, CHL was negatively and positively related to lake mean depth and percentage of wooded wetlands in the catchment, respectively. At the ecoregional scale, the slope parameter was positively related to the percentage of pasture in an ecoregion, indicating that CHL tends to respond more rapidly to changes in TP where there are high levels of agricultural pasture than where there is little. The intercept (i.e. the ecoregion‐average CHL) was negatively related to the percentage of wooded wetlands in the ecoregion. 5. By explicitly accounting for the hierarchical nature of lake–landscape interactions, we quantified the effects of landscape characteristics on the response of CHL to TP at two spatial scales. We provide new insight into ecoregional drivers of the rate at which algal biomass responds to changes in nutrient concentrations. Our results also indicate that the direction and magnitude of the effects of certain land use and cover characteristics on lake nutrient dynamics may be scale dependent and thus likely to represent different underlying mechanisms regulating lake productivity. 相似文献

Cestode larvae spend one phase of their two-phase life cycle in the viscera of rodents, but cases of cestodes infecting subterranean rodents have only been rarely observed. To experimentally gain some insight into this phenomenon, we captured approximately 300 plateau zokors(Eospalax baileyi), a typical subterranean rodent inhabiting the Qinghai-Tibet Plateau, and examined their livers for the presence of cysts. Totally, we collected five cysts, and using a mitochondrial gene(cox1) and two nuclear genes(pepck and pold) as genetic markers, we were able to analyze the taxonomy of the cysts. Both the maximum likelihood and Bayesian methods showed that the cysts share a monophyly with Taenia mustelae, while Kimura 2-parameter distances and number of different sites between our sequences and T. mustelae were far less than those found between the examined sequences and other Taeniidae species. These results, alongside supporting paraffin section histology, imply that the cysts found in plateau zokors can be regarded as larvae of T. mustelae, illustrating that zokors are a newly discovered intermediate host record of this parasite. 相似文献

The suitability as a host of Diadegma armillata (Gravenhorst) (Hymenoptera, Ichneumonidae) was determined for eight West-European representatives of the genus Yponomeuta Latreille (Lepidoptera, Yponomeutidae) by dissecting and rearing parasitized larvae under controlled conditions.The suitability of the various species differed considerably, and dissection showed that this was mainly due to the ability of the host species to encapsulate the parasitoid eggs.Encapsulation by Yponomeuta-species corresponded fairly well to the differences between the parasitoid complexes concerning D. armillata.Classifying the Yponomeuta-species into three categories according to their suitability, showed a remarkable parallelism with the phylogenetic relationships as determined by allozyme comparison: species showing high encapsulation rates are those that have diverged early in the evolution of the genus, whereas the more recently evolved species showed an intermediate percentage or were not able to encapsulate eggs of D. armillata.
Résumé L'étude a porté sur la valeur de 8 représentants européens du genre Yponomeuta (Lep., Yponomeutidae) comme hôtes pour Diadegma armillata (Hym., Ichneumonidae). La présence, l'état du parasitoïde, le nombre de chenilles contenant un hyménoptère et la mortalité des chenilles on été examinés par dissection et par élevage des chenilles attaquées en conditions contrôlées. Pour examiner si la mortalité des chenilles saines et parasitées diffèrent, des chenilles non contaminées par l'ichneumonide ont été utilisées comme témoins. De plus, des chenilles provenant de différentes parties des Pays-Bas et d'Allemagne ont été disséquées pour connaître leur taux de parasitisme par D. armillata.La valeur des diverses espèces diffère considérablement. Les dissections montrent que cela provient de différentes aptitudes à l'encapsulation des ufs du parasite. La comparaison des taux observés par la dissection des chenilles des différentes parties des Pays-Bas et d'Allemagne montre une surprenante uniformité pour les populations d'origines géographiques différentes.Les observations sur l'encapsulation par les différentes espèces d'yponomeutes correspondent assez bien aux différences observées parmi les divers complexes parasitaires dans lesquels D. armillata intervient. Certains éléments montrent cependant que le taux d'attaques victorieuses (sans tenir compte d'une éventuelle encapsulation ultérieure) contribue aussi aux différences observées entre les complexes parasitaires.Le classement des espèces d'yponomeutes en 3 catégories, d'après leur valeur comme hôte, montre un remarquable parallélisme avec les relations phylogéniques mises en évidence par analyse des allozymes: les espèces présentant une forte proportion d'encapsulation sont celles qui ont divergé précocement au cours de l'évolution du genre, tandis que les espèces ayant divergé le plus récemment présentent un pourcentage intermédiaire ou sont incapables d'encapsuler les ufs de D. armillata.相似文献

When males in captive tufted capuchin monkey (Cebus apella) groups separate and come together within their group, they have been observed to embrace and vocalize upon reunion. This display has not been observed in any other age/sex class. To investigate this, we deliberately separated six animals, including the only two adult males, from each of two social groups. We hypothesized that only the adult males would embrace upon reintroduction. When two males were consecutively reintroduced to their group, they typically ran to each other and came together in a frontal embrace, emitting stereotyped vocalizations. No other combination of animals showed this “reunion display.” Though dramatic, this behavior was in no way associated with incidents of aggression. (R)-(+)- and (S)-(?)-dihydrozeatins [(R)-(+)- and (S)-(?)-6-(4-hydroxy-3-methylbutylamino)purines, 1a and 1b] and their ribosides {(?)-6-[(R)-4-hydroxy-3-methylbutylamino]- and (?)-6-[(S)-4-hydroxy-3-methyl-butylamino]-9-β-D-ribofuranosylpurines, 3a and 3b} were synthesized and tested for their cytokinin activity by four bioassay systems, the growth of tobacco callus, the seed germination of lettuce, the fr. wt increase of excised radish cotyledons and the retardation of chlorophyll degradation in radish cotyledons. In tobacco callus bioassay, 1a was more active than 1b. The ribosides 3a and 3b were not less active than their corresponding aglycones 1a and 1b. In other bioassays used the activity followed the order: 1a >3a >1b >3b. The relationships among fifteen isolates of whitefly-transmitted geminiviruses (WTGs) from North, Central and South America and six from other continents were assessed (a) in nucleic acid hybridisation tests with sulphonated DNA probes for eight of the viruses, and/or (b) in triple-antibody-sandwich ELISA with panels of monoclonal antibodies (MAbs) to particles of African cassava mosaic virus (ACMV) and Indian cassava mosaic virus (ICMV). Probes specific for DNA-A of four American viruses, abutilon mosaic (AbMV), bean golden mosaic (BGMV), squash leaf curl (SLCV) and tomato golden mosaic (TGMV), detected virtually all the American viruses but reacted weakly if at all with ICMV, ACMV or tomato yellow leaf curl virus from Thailand (TYLCV-T). Conversely, the probe for ACMV DNA-A did not detect any of the American viruses, and that for TYLCV-T DNA-A reacted weakly with SLCV and TGMV0020but did not detect the others. In contrast, probes specific for DNA-B of the four American viruses or ACMV detected only the homologous virus, except for slight reactions between the AbMV DNA-B probe and both chino del tomate virus (CdTV)-DNA and SLCV-DNA. However, a probe for DNA-B of bean calico mosaic virus (BCMoV) reacted weakly with BGMV-PR DNA, and a probe for DNA-B of CdTV from Mexico detected several American viruses. Six out of 17 MAbs specific for ACMV and six out of 10 MAbs specific for ICMV reacted with one or other of the 14 American virus isolates tested. Two and-ACMV MAbs reacted with all, and one anti-ACMV MAb and two anti-ICMV MAbs reacted with nearly all the American viruses, one anti-ACMV MAb reacted with about half the American viruses and six other MAbs reacted with only one or two of them. Of the American viruses, CdTV and AbMV were the least closely related to the others. The epitope profiles of BCMoV, BGMV, cotton leaf crumple virus, serrano golden mosaic virus and SLCV were virtually indistinguishable. TGMV, potato yellow mosaic virus (PYMV) and an euphorbia virus had profiles intermediate between those of the BGMV cluster and AbMV-CdTV. In general, the epitope profiles and the results of hybridisation tests with DNA-A probes show that the similarities among the American viruses are greater than those between the American viruses and the viruses from other continents; the hybridisation tests with DNA-B probes show that substantial differences exist between individual American viruses. In America, geminivirus evolution seems to have proceeded convergently from different progenitor viruses, or divergently from one ancestral form, with DNA-B diverging to a greater extent than DNA-A and its particle-protein gene. 相似文献
