Batch production of xylitol from the hydrolysate of wheat straw hemicellulose using Candida guilliermondii was carried out in a stirred tank reactor (agitation speed of 300 rpm, aeration rate of 0.6 vvm and initial cell concentration of 0.5 g l–1). After 54 h, xylitol production from 30.5 g xylose l–1 reached 27.5 g l–1, resulting in a xylose-to-xylitol bioconversion yield of 0.9 g g–1 and a productivity of 0.5 g l–1 h–1.  相似文献

Studies were conducted to compare viability of immature and mature porcine oocytes vitrified in ethylene glycol (EG) using open-pulled straws (OPS). Oocytes that had been allowed to mature for 12 h (germinal vesicle group; GV) and 40 h (metaphase II group; MII) were divided into three treatments: (1) control; (2) treated with cytochalasin B and exposed to EG; and (3) treated with cytochalasin B and vitrified by stepwise exposure to EG in OPS. After warming, a sample of oocytes was fixed and evaluated by specific fluorescent probes before visualization using confocal microscopy. The remaining oocytes were fertilized and cleavage rate was recorded. Exposure of GV oocytes to EG or vitrification had a dramatic effect on spindle and chromosome configurations and no cleavage was obtained after in vitro fertilization. When MII oocytes were exposed to EG or were vitrified, 18 and 11% of oocytes, respectively, maintained the spindle structure and either EG exposure or vitrification resulted in substantial disruption in microfilament organization. The cleavage rates of mature oocytes after being exposed to EG or after vitrification were similar (14 and 13%, respectively) but were significantly less than that of control oocytes (69%). These results indicate that porcine oocytes at different meiotic stages respond differently to cryopreservation and MII porcine oocytes had better resistance to cryopreservation than GV stage oocytes.  相似文献

Abstract:  Carabid beetles were sampled by pitfall traps in May–September 2002 and 2003 in an experimental organic strawberry field in seven different mulching row treatments and in fescue-growing row spaces to determine the effect of mulches on carabid activity–density and species composition. There were no differences between treatments or row spaces in total numbers of all carabid species in either season. Of the 67 identified species and 4374 individuals, the most trapped ones were Clivina fossor (11.7% of total catch), Dyschirius globosus (10.2%), Patrobus atrorufus (9.3%), Pterostichus niger (8.8%), Bembidion properans (7.4%), Pterostichus melanarius (6.0%), Bembidion lampros (5.7%), Pterostichus crenatus (5.7%), Amara communis (4.1%), Trechus secalis (3.9%), Pterostichus strenuus (3.9%), Loricera pilicornis (3.1%), Harpalus rufipes (2.8%) and Carabus nemoralis (2.6%). Large carabid species P. niger and P. melanarius were trapped more in all mulched rows than in row spaces, and C. nemoralis was trapped more in the mixture of pine woodchip/buckwheat husk mulch than in row spaces, barley straw and birch woodchip mulch. The small but abundant species P. atrorufus was trapped more in buckwheat husk mulch than in row spaces and B. properans in row spaces than in buckwheat husk mulch. H. rufipes , being also a pest in strawberry, was trapped more in cut grass mulch than in row spaces. The Shannon H' diversity index was significantly higher in the mixture of pine woodchip/buckwheat husk mulch than in black plastic, buckwheat husk mulch or in row spaces, and in birch woodchip and cut grass mulches than in row spaces. The results of trapping studies show the species preference to the specific mulches, and reflect positive or negative effect of mulches on carabid diversity with consequences to plant protection.  相似文献

应用~(13)N标记稻草饲喂3只山羊,以探明羊对稻草N、C化合物的消化、吸收、排泄和转化规律。结果表明,已宰杀的2只羊消化、吸收、转化为羊机体的~(15)N占试验日粮中~(15)N富集总量的38.54和23.78%,平均为31.16%。3只羊从粪尿中排泄的~(15)N各占饲料中~(15)N的34.78、33.88和33.18%,平均为33.95±0.80%,已屠宰的2只羊对饲料~(15)N总回收率为73.32和56.96%,损失率为26.68和43.04%。饲料~(15)N的回收利用率与饲料中氨基酸的消化率(%)相吻合。1、2、3号羊对饲料碳水化合物的消化率分别为76.40、68.66和65.19%。其中饲喂2、3号羊的饲料中都含稻草50%左右,羊对碳水化合物的平均消化率为66.93%。  相似文献

Two field experiments examined the effect of straw spread on the soil surface on the incidence of bean yellow mosaic potyvirus (BYMV) in plots of narrow-leafed lupin (Lupinus angustifolius) sown at narrow (17.5 cm) vs wide (35 cm) row spacing and low (25–30 kg/ha) vs medium (50–60 kg/ha) seeding rates. Virus ingress was by vector aphids flying from adjacent pastures dominated by subterranean clover. In Expt 1, in which BYMV infection was extensive, straw greatly decreased the rate and amount of virus spread regardless of row spacing or plant density, decreasing infection more than 70% by the final assessment date. This effect of straw was attributed to decreased landing rates of incoming vector alates. In the plots without added straw, narrow row spacing decreased BYMV % infection by 38% by the last assessment date. Sowing at the medium seeding rate also decreased infection. The effect of wide row spacing seemed due to delayed canopy closure between rows which is likely to have increased the landing of aphids while the effect of medium seeding rate was attributed partly to the dilution effect of greater plant numbers and partly to the effects of partial canopy development in decreasing landing rates. In Expt 2, in which the incidence of BYMV infection was low, added straw again decreased BYMV spread, but by only 25–27% at final assessment; there were no effects of row spacing or seeding rate. In both experiments, an additional “reference” treatment was included which had a high (90–100 kg/ha) seeding rate, narrow rows and no straw. The dense canopy it developed also decreased BYMV incidence but less than in the plots with added straw in Expt 1. In Expt 1, adding straw and the resulting decrease in plants killed by BYMV, were associated with an overall increase in lupin grain yield of 20%. The greater plant densities resulting from the medium seeding rate also increased grain yield but row spacing did not affect it significantly. These results indicate that retaining stubble on the soil surface at seeding will assist in management of BYMV infection in lupin crops but that wide row spacing in the absence of retained stubble is undesirable.  相似文献

The thermal decomposition characteristics of four types (wheat, barley, oats and rye) of cereal straws were studied. Two varieties from each type of straw were used. The thermal degradation behaviours and kinetic parameters (order of reaction, activation energy and preexponential factor) of the straws were compared. Two distinct reaction zones were observed for all types and varieties of straws. Thermal degradation rates in the first reaction zone were significantly higher than those in the second reaction zone. The activation energy was in the ranges of 80–102 kJ/mol and 34–75 kJ/mol, whereas the order of reaction was in the ranges of 1·3–2·3 and 0·1–0·7 for the first and second reaction zones, respectively. The Shaw variety of oats straw had the highest activation energies (102 and 75 kJ/mol) and reaction orders (2·3 and 0·7) in both the first and second reaction zones, respectively. The lowest activation energy (80 kJ/mol) and order of reaction (1·3), in the first reaction zone, corresponded to Absolvant and Monopol wheat straws. The activation energies and reaction orders of barley and rye straws were in the ranges of 85–94 kJ/mol and 1·9–2·3, respectively. There was not any significant difference between the rate constants of the straw varieties, in the first reaction zone. However, oats straws had significantly higher rate constants in the second reaction zone as compared to the rate constants of wheat, barley and rye straws.  相似文献

Decomposing barley straw has previously been shown to inhibit the growth of a limited number of algae under both laboratory and field conditions. Bioassays were conducted on a range of algae to evaluate their relative sensitivities to straw-derived inhibitor(s). A range of sensitivities was found, including some species that were resistant to the straw-derived inhibitor(s). A microcystin-producing strain of Microcystis aeruginosa was very susceptible to decomposing barley straw. Bioassays using Euglena gracilis suggest that the inhibitory compounds are not derived from the phototransformation of straw decomposition products and do not act primarily by inhibiting photosynthesis. Susceptibility to barley straw appears not to be related to general taxonomic or structural features. Possible implications for algal populations in natural freshwaters are briefly discussed. Fourteen assays were conducted to study the influence of different variables, namely xylose concentration, inoculum level, agitation speed and nutrient supplementation, on xylitol biosynthesis by Candida guilliermondii FTI 20037. The maximum predicted values for xylitol yield (0.65 g g–1) and xylitol productivity (0.66 g l–1 h–1) can be attained with rice straw hydrolysate containing 60 g xylose l–1 without supplementation of ammonium sulfate, calcium chloride and rice bran extract, using 5 g inoculum l–1, at 250 rpm. Xylose concentration and inoculum level were selected for further optimization studies.  相似文献

Previous studies suggested that allelochemicals from sugarcane straw may suppress the growth of arrowleaf sida (Sida rhombifolia L.). A study was conducted to establish: (1) the direct or indirect role of the organic molecules from sugarcane straw leachate on the growth suppression of arrowleaf sida and (2) if leachate phytotoxins induce proline accumulation in arrowleaf sida tissues as an adaptative response to a water or an oxidative stress. Inhibition of root elongation was the primary effect of sugarcane straw leachate on arrowleaf sida grown in unsterile soil. Addition of activated charcoal to unsterile soil before incorporation of straw leachate reduced the inhibition in root growth suggesting a direct participation of organic molecules in leachate phytotoxicity on arrowleaf sida. Inorganic straw constituents did not inhibit root growth while microbial activity increased leachate phytotoxicity. Soil chemical analysis suggested a direct action of organic molecules in leachate phytotoxicity rather than variations in macro and micronutrients or nutrient microbial immobilization. Straw leachate induced proline accumulation in roots and cotyledons of arrowleaf sida. Proline increase was related with oxidative stress in the roots but not in the cotyledons. Our results indicate a direct action of organic compounds from sugarcane straw and/or their microbial transformation products on root growth of arrowleaf sida. These substances induced proline accumulation in roots mainly as consequence of an oxidative stress while water stress may be the main cause of high proline content in the cotyledons. Although the observed responses could be due to phenolic compounds, the involvement of organic molecules with other chemical nature could not be excluded.  相似文献
