Aims: The microbial quality of farm bulk‐tank raw milk produced in Estonia during years 2004–2007 was investigated. Methods and Results: Bulk‐tank milk samples were analysed for lactic acid bacteria count (LABC), psychrotrophic bacteria count (PBC), aerobic spore‐forming bacteria count (ASFBC), total bacterial counts using BactoScan and somatic cell count (SCC) using Fossomatic. Randomly selected psychrotrophic isolates were subjected to 16S–23S PCR‐ribotyping. LABC remained below 104 CFU ml?1 in most samples, while psychrotrophic micro‐organisms dominated in 60% of farms. PBC ranged from 4·2 × 102 to 6·4 × 104 CFU ml?1, and ASFBC varied from 5 to 836 CFU ml?1. Conclusions: In general, the microbiological quality of the farm bulk‐tank milk was good – more than 91% of samples contained <50 000 CFU ml?1, and SCC in the majority of samples did not exceed the internationally recommended limits. Genus Pseudomonas spp. was the dominating spoilage flora with Pseudomonas fluorescens as the prevailing species. Significance and Impact of the Study: Specific bacterial groups (LABC, PBC and ASFBC), not analysed routinely by dairies, were determined in bulk‐tank raw milk of numerous dairy farms during 4‐year period. Based on the survey, dairy plants can better control their supply chains and select farms (milk) for the production of specific products, i.e. milk with low PBC and high LABC for cheesemaking. 相似文献

Batch production of xylitol from the hydrolysate of wheat straw hemicellulose using Candida guilliermondii was carried out in a stirred tank reactor (agitation speed of 300 rpm, aeration rate of 0.6 vvm and initial cell concentration of 0.5 g l–1). After 54 h, xylitol production from 30.5 g xylose l–1 reached 27.5 g l–1, resulting in a xylose-to-xylitol bioconversion yield of 0.9 g g–1 and a productivity of 0.5 g l–1 h–1. 相似文献

The growth and photosynthetic responses ofPterocladiella capillaceato NH4, PO4, CO2-enrichment, pH, irradiance and temperature were evaluated for winter or summer plants cultivated under laboratory and outdoor settings. In the laboratory, using a gradient table, optimal growth temperature and irradiance for winter plants occurred at 10–20 °C and 100 mol photon m–2s–1, averaging 24.3% per week. The optimal growth conditions found for summer plants were 10–20 °C and 20 mol photon m–2s–1, averaging 29.0% per week. In a pH-stat cultivation system photosynthetic rates and growth rates were largely unaffected by pH in the range 6.5–8.5, however, they both decreased significantly above 8.5. In outdoor settings, using 40 L tanks,P. capillaceawas more responsive to the frequency the algae were fed with NH4and PO4rather than the relative concentrations of these nutrients. The average growth rates during winter were 28.3% and 12.5% per week when NH4and PO4were included once and twice a week for 24-h periods, respectively, while summer plants grew 15.0% and 25.3% per week at these nutrient regimes. Algae grown in seawater (containing 13.8 ± 1.8 M CO2) or CO2-enriched seawater (averaging 33.7 ± 13.2 M CO2) had similar growth rates or even reduced productivity under CO2-enrichment during winter. Concentrations of chlorophyllawere in average significantly higher in winter as compared to summer especially when nutrients were included twice a week. Phycoerythrin levels were also higher for plants fed with nutrients twice a week particularly during summer time. Although agar yields were limited and not seasonally dependent, this study shows high growth capacity forP. capillaceaas compared to previous investigations. Future mariculture prospective using current tank cultivation techniques for this species will likely depend on market demands for high quality agar. 相似文献

Diel changes in mean cell volume, cellular carbon (carbon content per cell), cellular Chl a, C/N ratio, Chl a/carbon ratio and pigment composition were determined for an axenic clonal culture of Pyramimonas parkeae Norris et Pearson through three 12:12 h LD cycles in a laboratory culture tank of 1 m3. Mean cell volume and cellular C, N and most pigments increased during the light period as a result of photosynthesis and decreased with an increase in cell density by phased cell division during the dark period. Chi a and Chi b increased in a parallel manner during the light period. Increases in the diel synthesis pattern of carotenoids varied. Violaxanthin and lutein content increased for a few hours at the beginning of the light period and preceeded that of neoxanthin. The diel synthesis pattern of neoxanthin was similar to that of Chi a. Increases of loroxanthin and its ester form were slower than that of Chi a at the beginning of the light period. A net increase of α-carotene was observed during the dark period. Mass spectroscopy of carotenoid structure showed a new xanthophyll, loroxanthin dodecenoate, in this species. 相似文献

Abstract The complex-forming compound oxalic acid can effectively solubilise metals such as aluminium, iron, lithium, and manganese. In order to produce high amounts of oxalic acid for biohydrometallurgical processes, it was the aim of this work to optimise oxalic acid production by Aspergillus niger , a fungus well known for its ability to produce oxalic acid. A. niger excreted 427 mmol oxalic acid 1−1 if it was cultivated in a pH-controlled (pH 6.0) fed-batch run in a 2-1 stirred tank reactor. Sucrose and lactose permeate were suitable carbon sources for oxalic acid production. In sucrose medium, A. niger produced high amounts of gluconic and oxalic acids, whereas in lactose permeate medium only oxalic acid was produced. This study's objective was to clarify both experimentally and theoretically whether a vertical multistage tower aeration tank system is advantageous as compared with a completely mixed system, particularly with respect to purification efficiency, sludge settleability, and excess sludge production. In comparing the two systems: (1) purification efficiency in the multistage tower aeration system with partial fluid mixing with a large Peclet number was higher than in a corresponding completely mixed system for all applied organic loadings; (2) the multistage tower aeration system had some definite advantages with respect to sludge settleability and excess sludge production; (3) the activated sludge system's higher performance with partial fluid mixing was shown quantitatively with the axial dispersion model in conjunction with growth kinetics which involved rapid uptake such as biosorption and subsequent oxidative biodegradation processes of organic substances. Apical callus formation in Solieria filiformis (Gigartinales,Rhodophyta) cultured in tanks 总被引:1,自引:1,他引:0
Loose-lying wild plants of the carragenophyte Solieria filiformis (Kützing) Gabrielson were cultivated under greenhouse conditions in 600 l tanks in stationary and turbulent cultures, produced either by air bubbling or water jets at the bottom of the tanks. One week after inoculation 90.3% of the apices of the plants grown in air turbulent cultures initiated the formation of callus. The apices were not broken and apparently non-wounded. No callus formation were observed from a few accidentaly broken apices in any culture. Only 4% of the apices in water turbulent cultures induced callus. Reorganization of branches from the calli took place after three weeks. Organogenetic calli detached from the mother plant after four weeks and formed spherical masses of 3 cm in diameter growing as unattached balls. Cellular disorganization (i.e. callus formation) in S. filiformis seems to be a consequence of intermittent abrasion or contact stimuli against tank walls produced by turbulence. 相似文献

The mycorrhizal status of water-impounding tank bromeliad epiphytes from three locales differing in altitude and moisture regime within Venezuelan cloud forest was examined. Species of vesicular-arbuscular mycorrhizal (VAM) fungi found in arboreal soils were compared to VAM fungi found in terrestrial soils. Sixteen of the 19 epiphytes examined for the presence of VAM fungi had roots with infection stages; 14 of these specimens showed growth of the fine endophyte Glomus tenue. Fine endophyte was the only VAM fungus found associated with epiphytes in the driest locale studied, while coarse VAM fungi (Gigaspora and Scutellospora spp.) were found at sampling locales receiving more moisture. Root infection was usually composed of intercellular hyphae and peletons; few arbuscules were observed. However, abundant extracellular hyphae were often observed tangled about roots in arboreal soil. It is concluded that epiphytic bromeliads probably benefit, at least periodically, from VAM fungi scavenging for sporadically available nutrients in arboreal soils. Glomus tenue may be particularly important as a colonizing VAM fungus in drier sites of Venezuelan cloud forest. The species composition of VAM fungi in arboreal soils was different to that of terrestrial soils sampled directly under epiphytic bromeliad perches, suggesting that VAM fungi species associated with bromeliads are dispersed to their hosts by vagile animal vectors. 相似文献

The scale of operation of freely suspended animal cell culture has been increasing and in order to meet the demand for recombinant
therapeutic products, this increase is likely to continue. The most common reactor types used are stirred tanks. Air lift
fermenters are also used, albeit less commonly. No specific guidelines have been published for large scale (≥10 000 L) animal
cell culture and reactor designs are often based on those used for microbial systems. However, due to the large difference
in energy inputs used for microbial and animal cell systems such designs may be far from optimal. In this review the importance
of achieving a balance between mixing, mass transfer and shear effects is emphasised. The implications that meeting this balance
has on design of vessels and operation, particularly in terms of strategies to ensure adequate mixing to achieve homogeneity
in pH and dissolved gas concentrations are discussed.

In Thermus thermophilus HB27 cultures the localisation of lipolytic activity is extracellular, intracellular and membrane bound, with low percentage for the former. Therefore, the extracellular secretion must be increased in order to simplify the downstream process and to reduce the economic cost. This study focuses on the design of an innovative operational strategy to increase extracellular lipolytic enzyme production by T. thermophilus HB27 at bioreactor scale. In order to favour its secretion, the effect of several operational variables was evaluated. Among them, the presence of oils in the culture medium leads to improvements in growth and lipolytic enzyme activity. Sunflower oil is the most efficient inducer showing better results when added after 10 h of growth. On the other hand, although surfactants lead to an almost complete inhibition of growth and lipolytic enzyme production, their addition along the culture could affect the location of the enzyme. Thus, by addition of surfactants at the stationary phase, a release of intracellular and membrane enzyme which increases the extracellular enzyme proportion is detected. Based on these results, strategies with successive addition of oil and surfactant in several culture phases in shake flask are developed and verified in a laboratory scale stirred tank bioreactor. 相似文献
