Assessment of a model for intron RNA secondary structure relevant to RNA self-splicing--a review 总被引:36,自引:0,他引:36
A widespread class of introns is characterized by a particular RNA secondary structure, based upon four conserved nucleotide sequences. Among such "class I" introns are found the majority of introns in fungal mitochondrial genes and the self-splicing intron of the large ribosomal RNA of several species of Tetrahymena. A model of the RNA secondary structure, which must underlie the self-splicing activity, is here evaluated in the light of data on 16 further introns. The main body or "core structure" of the intron always consists of the base-paired regions P3 to P9 with the associated single-stranded loops, with P2 present also in most cases. Two minority sub-classes of core structure occur, one of which is typical of introns in fungal ribosomal RNA. Introns in which the core structure is close to the 5' splice site all have an internal guide sequence (IGS) which can pair with exon sequences adjacent to the 5' and 3' splice sites to align them precisely, as proposed by Davies et al. [Nature 300 (1982) 719-724]. In these cases, the internal guide model allows us to predict correctly the exact location of splice sites. All other introns probably use other mechanisms of alignment. Summary Herein is described an attempts to establish chromosome pairing-interchange relationships in Drosophila melanogaster female. For this purpose, the formation of half-translocations was studied in XXY and XX females bearing compounds of the second pair of autosomes. With respect to XXY females, it was expected that the free Y chromosome would pair with these compounds and that half-translocations involving 2L would arise. In as much as compound chromosomes in XX females had no partner for pairing, the formation of half-translocations involving 2L was not expected.Half-translocations were registered in the F1 from crosses of XX and XXY females to b j pr cn/T(Y;2)C males. The cross was designed to permit the detection of very rarely occurring non-homologue interchanges.Offspring number was 335 in XX females and 550 in XXY females. The majority of offspring consisted of individuals arisen from the spontaneous restitution of compounds and the formation of 2n egg cells. Based on phenotype, the offspring of XX females contained 4 individuals with half-translocations involving 2L; there were 48 such flies among the offspring of XXY females. As confirmed by progeny analysis, 38 half-translocations occurred in XXY females and none in XX females. Of the 31 spontaneous interchanges in XXY females 28 were recorded between the Y and the left compound, one between the Y and the right compound, and one between the X and the left compound. Non-homologue interchanges were of oogonial origin judging by the fact that individuals with half-translocations arose in clusters. The nucleotide sequence from the 5′ terminus inward of one third of mouse α- and βmaj-globin messenger RNAs has been established. In addition, using 5′ 32P end-labeled mRNAs as substrates and S1 and T1 nucleases as probes for single-stranded regions, the secondary structures of mouse and rabbit α- and β-globin mRNAs have been analyzed. Our results indicate that the AUG initiator codon in both mouse and rabbit β-globin mRNA is quite susceptible to cleavage with S1 and T1 nucleases, suggesting that it resides in a single-stranded exposed region. In contrast, the initiator AUG in the α-globin mRNA of both species is inaccessible to cleavage, indicating that it is either buried by tertiary structure or is base-paired. Since the rate of initiation of protein synthesis with β-globin mRNA in rabbit reticulocyte is 30–40% faster than for α-globin mRNA, these results imply a possible correlation between the differential rates of initiation with these two mRNAs and the accessibility of the respective AUG initiator codons. 相似文献

The active site metal in horse liver alcohol dehydrogenase has been studied by metal-directed affinity labeling of the native zinc(II) enzyme and that substituted with cobalt(II) or cadmium(II). Reversible binding of bromoimidazolyl propionic acid to the cobalt enzyme blueshifts the visible absorption band originating from the catalytic cobalt atom at 655 to 630 nm. Binding of imidazole to the cobalt(II) enzyme redshifts the 655 nm band to 667 nm. Addition of bromoimidazolyl propionic acid blueshifts this 667 nm band back to 630 nm. This proves direct binding of the label to the active site metal in competition with imidazole. The affinity of the label for the reversible binding site in the three enzymes follows the order Zn ? Cd ? Co. After reversible complex formation, bromoimidazolyl propionic acid alkylates cysteine-46, one of the protein ligands to the active site metal. The nucleophilic reactivity of this metal-mercaptide bond in each reversible complex follows the order Co ? Zn ? Cd. 相似文献

Flower formation was induced in the shortday plant Kalanchoë blossfeldiana Poellnitz under long-day conditions by grafting with flowering shoots of the short-long-day plant Echeveria harmsii (Rose) MacBr. and of the long-day plant Echeveria pulvoliver (E. pulvinata Rose x E. harmsii). Vegetative shoots from induced Echeveria plants failed to cause a flowering response in Kalanchoë. The presence of flowering and vegetative shoots side by side on induced Echeveria plants provides evidence for physiological chimeras in this genus.Abbreviations LD

- 1 Eggs of the British woodland mosquito, Aedes punctor (Kirby), were subjected to a variety of media in an attempt to determine the hatching stimulus.
- 2 It was found that continuous immersion in distilled water evoked almost no hatching (0.2%); intermittent removal from the water gave low hatching (8.9%).
- 3 Slow deoxygenation of the medium by either bacterial growth or the introduction of nitrogen produced the highest level of hatching (26.0–90.0%). Eggs of A.punctor were shown to hatch within a 4 h period of the oxygen concentration in the medium reaching zero.
