We explored the potential of the cox1 gene in the species resolution of soil fungi and compared it with the nuclear internal transcribed spacer (ITS) and small subunit (SSU)-rDNA. Conserved primers allowing the amplification of the fungal cox1 gene were designed, and a total of 47 isolates of Zygomycota and Ascomycota were investigated. The analysis revealed a lack of introns in >90% of the isolates. Comparison of the species of each of the six studied genera showed high interspecific sequence polymorphisms. Indeed, the average of nucleotide variations (4.2–11%) according to the genus, due mainly to the nucleotide substitutions, led to the taxonomic resolution of all the species studied regarding both ITS and SSU-rDNA, in which <88% were discriminated. The phylogenetic analysis performed after alignment of the cox1 gene across distant fungal species was in accordance with the well-known taxonomic position of the species studied and no overlap was observed between intra- and interspecific variations. These results clearly demonstrated that the cox1 sequences could provide good molecular markers for the determination of the species composition of environmental samples and constitute an important advance to study soil fungal biodiversity.  相似文献

Abstract

Recent work has demonstrated indirect effects between mycorrhizal fungi and insect herbivores and pollinators. The existence of indirect effects between mycorrhizal fungi and protection-for-food mutualists, such as extrafloral nectar-foraging ‘bodyguard ants’, is unknown. In this study, we examined the potential for indirect effects of arbuscular mycorrhizal fungi on aggressive ant bodyguards, mediated by changes in the expression of extrafloral nectaries of a shared host plant. We found that mycorrhizal plants grew larger and produced more extrafloral nectaries compared to their non-mycorrhizal counterparts. The difference in the number of nectaries between mycorrhizal and non-mycorrhizal plants, however, was too small to elicit differences in ant attendance. In spite of the lack of a significant indirect effect of mycorrhizal fungi on ant attendance, mycorrhizal plants suffered damage to a significantly greater proportion of their leaves compared to non-mycorrhizal plants. This result likely stems from other (non-ant-mediated) indirect effects of mycorrhizal fungi on herbivores.  相似文献

Premature death has been defined as a growth stoppage linked to the accumulation of specific deletions of the mitochondrial genome (mtDNA) inPodospora anserina. This occurs only in strains carrying theAS1-4mutation which lies in a gene encoding a cytosolic ribosomal protein. Here we describe the isolation and genetic characterization of 10 nuclear mutations which either delay the appearance of this syndrome (respite from premature death) or cause a switch to the classical senescence process (repeal of premature death). These mutations lie in at least six genes. Some cause defects at the levels of ascospore germination, growth rates, and/or sensitivity toward inhibitors of protein syntheses. All modify the onset of senescence in wild-type (AS1+) strains. The role played by these genes is discussed with respect to the control of diseases due to mtDNA rearrangements in filamentous fungi.  相似文献

An adult male owl monkey (Aotus nancymai) had hypercalcemia associated with a disseminated case of histoplasmosis (Histoplasma capsulatum var. capsulatum). Histopathologic examination of tissues at necropsy showed moderate to marked numbers of small granulomas involving multiple organs. The granulomas contained numerous round, basophilic, intracellular structures, 1-2 microns in diameter, that stained positive in Periodic acid-Schiff and Gomori's methenamine silver tests. The hypercalcemia in this case was attributed to enhance conversion of dietary vitamin D to 1,25-dihydroxycholecalciferol by the granulomas.  相似文献

 We used arrays of artificial flowers with and without fragrance to determine the importance of olfactory and visual cues in attracting insects to a floral mimic. The mimic is a fungus, Puccinia monoica Arth., which causes its crucifer hosts (here, Arabis drummondii Gray) to form pseudoflowers that mimic co-occurring flowers such as the buttercup, Ranunculus inamoenus Greene. Although pseudoflowers are visually similar to buttercups, their sweet fragrance is distinct. To determine whether visitors to pseudoflowers were responding to fragrance we performed an experiment in which we removed the visual cues, but allowed fragrance to still be perceived. In this experiment we found that pseudoflower fragrance can attract visitors by itself. In other experiments we found that the relative importance of olfactory and visual cues depended on the species of visitor. Halictid bees (Dialictus sp.) had a somewhat greater visual than olfactory response, whereas flies (muscids and anthomyiids) were more dependent on olfactory cues. We also used bioassays to determine which of the many compounds present in the natural fragrance were responsible for attraction. We found that halictid bees were equally attracted to pseudoflowers and to a blend containing phenylacetaldehyde, 2-phenylethanol, benzaldehyde and methylbenzoate in the same relative concentrations as in pseudoflowers. Flies, on the other hand, only responded to pseudoflower scent, indicating that we have not yet identified the compound(s) present in pseudoflowers that are attracting them. The ability of insects to differentiate pseudoflowers from true flowers by their fragrance may be important in the evolution of the mimicry system. Different fragrances may facilitate proper transfer of both fungal spermatia and pollen, and thus make it possible for the visual mimicry to evolve. Root colonisation byGlomus fistulosum BEG 31 of a plant population from the outbreeding wild strawberry (Fragaria vesca) ranged from 42 to 80%; this was correlated with a significant increase in fruit trusses, berries and leaves. The mycorrhizal plants produced significantly more primary and secondary runners but less lateral runners. One hundred randomly selected seeds were multiplied in vitro and 4 clonal populations were selected. Root colonisation in the clones varied from 16 to 75%, with significant difference in the % colonisation between some of the clones. Percentage mycorrhizal colonisation was positively correlated with effects on host growth.The positive correlation between mycorrhizal root colonisation and growth effects in the clones was not reflected in their respective susceptibility, in the non-mycorrhizal state, toPhytophthora fragariae infection. Clones showing the highest and lowest mycorrhizal root colonisation showed high disease susceptibility whereas a clone with intermediate colonisation was resistant. When the clones were colonised with mycorrhizal fungi, the two susceptible clones became resistant toP. fragariae whereas the resistant and partially-resistant clones were less affected.
