Selected strains of ectomycorrhizal fungi, such as the basidiomycete Laccaria bicolor , are currently being used as inoculants in nurseries to improve growth of forest trees after outplanting. Information is needed on the survival of these introduced strains in forests and their impact on indigenous biodiversity. Dissemination and persistence of an American strain, L. bicolor S238N, were studied 10 years after outplanting in a Douglas fir plantation located at Saint-Brisson (Morvan, France). About 430 Laccaria spp. sporophores were collected over 3 years. Inheritance of nuclear ribosomal DNA, as well as RAPD markers, was characterized in L. bicolor S238N, using a haploid progeny set of 91 monokaryons. More than 50 markers were identified (19 heterozygous and 33 homozygous or cytoplasmic markers), which unambiguously confirmed that the introduced strain was still present in the inoculated plots. Neither selfing ( P < 0.0008) nor introgression with indigenous strains was detected although in vitro interfertility between the American strain and indigenous L. bicolor was identified. No ingress of the introduced genet into adjacent uninoculated plots colonized by various local Laccaria genets was detected. It is proposed that the spatial distributions identified have developed through mycelial propagation of the introduced strain and intraspecific competition with native genets. Although longer-term data is still lacking, the stability of the inoculant strain and the limited disturbance to indigenous populations described support large-scale nursery production of this host-fungal combination. 相似文献

The ability of arbuscular mycorrhizal (AM) fungi from a metal-tolerant plant (Viola calaminaria, violet) to colonise and reduce metal uptake by a non-tolerant plant (Trifolium subterraneum, subterranean clover) in comparison to a metal-tolerant AM fungus isolated from a non-tolerant plant was studied. AM spores
from the violet rhizosphere and from violet roots were characterised by polymerase chain reaction (PCR) amplification of the
SSU rDNA, and sequencing. Subterranean clover was grown in pots containing a soil supplemented with Cd and Zn salts and inoculated
either with a mixture of spores extracted from the violet rhizosphere or with spores of a Cd-tolerant Glomus mosseae P2 (BEG 69), or non-inoculated. The diversity of fungi, including AM fungi, colonising clover roots was assessed and analysed
using terminal-restriction fragment length polymorphism. At least four different Glomus species were found in the violet rhizosphere. After 8 weeks in a growth chamber, colonisation of clover roots with spores
from the violet rhizosphere increased Cd and Zn concentrations in clover roots without significantly affecting the concentrations
of metals in the shoot and plant growth. G. mosseae P2 reduced plant growth and slightly increased the Cd concentration. Only one AM fungus (Glomus b) from the violet rhizosphere colonised clover roots, but other fungi were present. AM fungi from heavy metal-contaminated
soils and associated with metal-tolerant plants may be effective in accumulating heavy metals in roots in a non-toxic form.

Diversity of endophytic fungi of single Norway spruce needles and their role as pioneer decomposers 总被引:6,自引:0,他引:6
The diversity of endophytic fungi within single symptomless Norway spruce needles is described and their possible role as pioneer decomposers after needle detachment is investigated. The majority (90%) of all 182 isolates from green intact needles were identified as Lophodermium piceae. Up to 34 isolates were obtained from single needles. Generally, all isolates within single needles had distinct randomly amplified microsatellite (RAMS) patterns. Single trees may thus contain a higher number of L. piceae individuals than the number of their needles. To investigate the ability of needle endophytes to act as pioneer decomposers, surface-sterilized needles were incubated on sterile sand inoculated with autoclaved or live spruce forest humus layer. The dry weight loss of 13-17% found in needles after a 20-week incubation did not significantly differ between the sterilized and live treatments. Hence, fungi surviving the surface sterilization of needles can act as pioneer decomposers. A considerable portion of the needles remained green during the incubation. Brown and black needles, in which the weight loss had presumably taken place, were invaded throughout by single haplotypes different from L. piceae. Instead, Tiarasporella parca, a less common needle endophyte, occurred among these invaders of brown needles. Needle endophytes of Norway spruce seem thus to have different abilities to decompose host tissues after needle cast. L. piceae is obviously not an important pioneer decomposer of Norway spruce needles. The diversity of fungal individuals drops sharply when needles start to decompose. Thus, in single needles the decomposing mycota is considerably less diverse than the endophytic mycota. 相似文献

We tested the hypotheses that responses to the mountain pine beetle fungal associate Grosmannia clavigera will differ between the evolutionarily co‐evolved host lodgepole pine (Pinus contorta var. latifolia) and the naïve host jack pine (Pinus banksiana) and that these responses will be influenced by water availability. G. clavigera inoculation resulted in more rapid stem lesion development in lodgepole than in jack pine; water deficit delayed lesion development in both species. Decreased hydraulic conductivity was observed in inoculated lodgepole pine seedlings, likely because of tracheid occlusion by fungal hyphae and/or metabolite accumulation. Drought but not inoculation significantly impacted bark abscisic acid levels. Jasmonic and salicylic acid were implicated in local and systemic responses of both species to G. clavigera, with salicylic acid appearing to play a greater role in jack pine response to G. clavigera than lodgepole pine. Water deficit increased constitutive levels and/or attenuated induced responses to G. clavigera for several monoterpenes in lodgepole but not jack pine. Instead, inoculation of well‐watered but not water deficit jack pine resulted in a greater number of xylem resin ducts. These findings reveal mechanisms underlying differences in G. clavigera‐induced responses between lodgepole and jack pine hosts, and how water availability modulates these responses. 相似文献

An isolate (G15) of a bacterium, frequently isolated from roots of various plant species, was identified asSerratia plymuthica. At low temperature (viz. 2–8°C), the studied isolated readily produced a red pigment which proved useful in recognizing the bacteria on reisolation.
In laboratory tests it exhibited strong antagonism againstBotrytis cinerea andGerlachia nivalis and moderate antagonism againstRhizoctonia solani, Fusarium culmorum andPythium sp. The bacterium significantly increased growth of lettuce plants when applied to the roots under non-sterile conditions

Although arbuscular mycorrhizal fungi (AMF) form spatially complex communities in terrestrial ecosystems, the scales at which this diversity manifests itself is poorly understood. This information is critical to the understanding of the role of AMF in plant community composition. We examined small-scale (submetre) variability of AMF community composition (terminal restriction fragment length polymorphism fingerprinting) and abundance (extraradical hyphal lengths) in two 1 m(2) plots situated in a native grassland ecosystem of western Montana. Extraradical AMF hyphal lengths varied greatly between samples (14-89 m g soil(-1)) and exhibited spatial structure at scales <30 cm. The composition of AMF communities was also found to exhibit significant spatial autocorrelation, with correlogram analyses suggesting patchiness at scales <50 cm. Supportive of overall AMF community composition analyses, individual AMF ribotypes corresponding to specific phylogenetic groups exhibited distinct spatial autocorrelation. Our results demonstrate that AMF diversity and abundance can be spatially structured at scales of <1 m. Such small-scale heterogeneity in the soil suggests that establishing seedlings may be exposed to very different, location dependent AMF communities. Our results also have direct implications for representative sampling of AMF communities in the field. 相似文献

对从药渣中分离的真菌ZYJHYZ254进行鉴定及产酶活性研究,从转录组分析菌株不同生长时期差异表达基因对其生长发育及产酶调控的影响,以筛选高产木质纤维素水解酶真菌,寻找调控关键基因。鉴定ZYJHYZ254为拟暗射脉菌Phaeophlebiopsis sp.,产酶在第5-7天最高。从生长3 d与7 d的菌丝中共检测到1 232个差异基因,以3 d的菌丝为对照,显著上调、下调基因分别有826及406个,基因注释和GO、KEGG功能富集分析结果表明差异表达基因主要与蛋白质合成、代谢及酶合成相关。此外,共有387个CAZymes基因表达,GH数量最多,约占49.61%,其次为AA (97)与GT (62),约占25.06%与16.02%。GH16 (24个)占GH的12.50%,含量最多,主要编码葡萄糖苷酶、木聚糖酶等,AA中AA3 (37个)占比38.14%,编码氧化酶、脱氢酶等。结果表明ZYJHYZ254中生长3 d与7 d的菌丝经功能富集分析发现差异表达基因主要与蛋白质合成、代谢,以及酶合成相关。进一步研究发现在两个生长时期中CAZymes基因表达最多的是GH16与AA3,预示了该菌葡萄糖苷酶、木聚糖酶、β-半乳糖苷酶、氧化酶与脱氢酶含量最丰富,对降解特殊生物质中的木质纤维素具有重要意义。 相似文献

通过对文字衣科黑文衣属形态学、解剖学、化学与分子生物学的研究,发现中国新记录种1种,即变黄黑文衣(Phaeographis flavescens Dal-Forno&Eliasaro),该种主要特征为地衣体壳状,表面浅黄绿色,无光泽,光滑;子囊盘弯曲,多分支,聚生于假子座中;盘缘黑色,相邻线盘之间有小缝隙,盘面较平坦,黑色,覆有白色粉霜;子囊层无色透明,侧丝顶端分支;子囊含8个子囊孢子;子囊孢子褐色,横隔透镜型,(4~)6胞室,大小19~25.5μm×6~7μm,I+紫红色。该种分布于云南和福建,该研究丰富了对中国黑文衣属物种组成与分布的认知。 相似文献

Airborne fungal counts and types were examined in three selected regions in Egypt. Two of the sampling sites are rural areas, one cultivated with chamomile and the second with vegetable. The third site is located in an urban area. A sedimentation method was used to isolate airborne fungal spores. Airborne fungal spore counts averaged 71\pm 19, 64\pm 14 and 175\pm 79 cfu/p/h in the urban, vegetable and chamomile growing areas, respectively. A total of 1486 fungal colonies belonging to 32 genera were identified. Alternaria (7.5–59.9%), Aspergillus (11.2–38.9%), Penicillium (9.5–15%) and Cladosporium (7.78–17.5%) were the predominant fungal genera found in all sampling sites. Alternaria (42–59.9%) and Aspergillus (38.9%) were the common fungal genera in the cultivated and urban areas, respectively. Vegetation is considered the main source of Alternaria, whereas Aspergillus, Penicillium and Cladosporium are related to local microenvironments and urbanization. Acremonium, Aureobasidium, Botrytis, Beauveria, Chlamydomyces, Chalara, Curvularia, Fusarium, Geotrichum, Trichothecium, Oidiodendron, Scopulariopsis, Spicaria, Stachybotrys chartarum, Torula and Thamnidium, were only detected in low percentages (0.11–1.8%) in the cultivated areas. Vegetation adds different fungal types into the air and their numbers vary according to vegetation type and weather conditions. Airborne fungal counts increased with temperature and decreased with rainfall and relative humidity. Airborne fungal spores have many implications in the spread of human and plant diseases. The presence of fungal spores in air, in spite of their counts, may raise arguments about their role in health complaints in a particular region, „i.e., the fungal concentration may be low but the predominant aeroallergen may be dangerous”. 相似文献
