The 13/12C ratio in plant roots is likely dynamic depending on root function (storage versus uptake), but to date, little is known about the effect of season and root order (an indicator of root function) on the isotopic composition of C‐rich fractions in roots. To address this, we monitored the stable isotopic composition of one evergreen (Picea abies) and one deciduous (Fagus sylvatica), tree species' roots by measuring δ13C of bulk, respired and labile C, and starch from first/second and third/fourth order roots during spring and fall root production periods. In both species, root order differences in δ13C were observed in bulk organic matter, labile, and respired C fractions. Beech exhibited distinct seasonal trends in δ13C of respired C, while spruce did not. In fall, first/second order beech roots were significantly depleted in 13C, whereas spruce roots were enriched compared to higher order roots. Species variation in δ 13C of respired C may be partially explained by seasonal shifts from enriched to depleted C substrates in deciduous beech roots. Regardless of species identity, differences in stable C isotopic composition of at least two root order groupings (first/second, third/fourth) were apparent, and should hereafter be separated in belowground C‐supply‐chain inquiry.  相似文献

Products of the reaction between dialdehyde starch and Y-NH2 compounds (e.g. semicarbazide or hydrazine) are effective ligands for metal ions. The usefulness of these derivatives was tested in the experiment, both in terms of the immobilization of heavy metal ions in soil and the potential application in phytoextraction processes. The experimental model comprised maize and the ions of such metals as: Zn(II), Pb(II), Cu(II), Cd(II), and Ni(II). The amount of maize yield, as well as heavy metal content and uptake by the aboveground parts and roots of maize, were studied during a three-year pot experiment. The results of the study indicate the significant impact of heavy metals on reduced yield and increased heavy metal content in maize. Soil-applied dialdehyde starch derivatives resulted in lower yields, particularly disemicarbazone (DASS), but in heavy metal-contaminated soils they largely limited the negative impact of these metals both on yielding and heavy metal content in plants, particularly dihydrazone (DASH). It was demonstrated that the application of dihydrazone (DASH) to a soil polluted with heavy metals boosted the uptake of Zn, Pb, Cu, and Cd from the soil, hence there is a possibility to use this compound in the phytoextraction of these metals from the soil. Decreased Ni uptake was also determined, hence the possibility of using this compound in the immobilization of this metal. The study showed that dialdehyde starch disemicarbazone was ineffective in the discussed processes.  相似文献

The study was conducted to investigate the nutritional value of five samples of raw and extruded pea seeds (Pisum sativum L., Tarachalska cv.) from different experimental fields. The study included 150 male 1-day-old Ross 308 chickens, which were randomly assigned to three dietary treatments (50 replications each) and kept in individual cages. From days 1 to 16, all birds received only the basal diets. From days 17 to 21, the control group received still the basal diet, but for the two other groups, 20% of basal diet was replaced by raw or extruded peas. Furthermore, the groups receiving raw or extruded peas were divided into five subgroups of 10 animals each, where the diets contained one of the five pea samples of the same cultivar grown at different locations, respectively. On days 19 and 20, excreta were individually collected, and then all chickens were sacrificed and ileal digesta were sampled for determination of ileal digestibility, which was calculated by the difference method. Extrusion of pea seeds decreased the contents of crude fibre, acid and neutral detergent fibre, trypsin inhibitor activity (TIA), phytic P and resistant starch (RS) (≤ 0.05), but increased the contents of apparent metabolisable energy (AMEN) by approximately 2.25 MJ/kg dry matter (DM). Furthermore, extrusion improved the DM and crude protein digestibility significantly by about 21.3% and 11.6%, respectively. Similar results were observed for the digestibility of all analysed amino acids. In conclusion, extrusion markedly influenced the chemical composition of peas, reduced their contents of phytic P, TIA and RS and consequently had a positive impact on nutrient digestibility and AMEN values.  相似文献

NT-1 cells of tobacco (Nicotiana tabacum L.) were transformed with pGBSSHBS and pGBSSHER expression cassettes wherein expression of hepatitis B surface antigen (HBsAg) was driven by potato granule-bound starch synthase (GBSS) promoter. The transformed nature of the cells was confirmed by PCR analysis. Expression of HBsAg was confirmed by RT-PCR and Western blotting and levels of expression were assayed by ELISA. Transformed cell lines exhibited a sucrose-inducible pattern of HBsAg expression. NT-1 medium supplemented with 175 mmol L−1 sucrose gave the highest HBsAg expression of 198 ng g−1 FW after 8 days of induction. Different sugars, for example glucose, fructose, and palatinose, were also tested to study the inducible nature of GBSS promoter. The results demonstrate that potato GBSS promoter can be used in heterologous host systems like tobacco NT-1 cell suspension cultures for sucrose-inducible expression of recombinant proteins.  相似文献

目的:探讨以改性聚乳酸为细胞外基质网架构建组织工程皮肤的可行性。方法:采用盐溶法制备机械性能得到部分改进的聚乳酸多孔泡沫网架,向改进的聚乳酸网架接种真皮成纤维细胞和表皮角质形成细胞,以普通聚乳酸支架作为对照,构建组织工程皮肤。体外培养一周,对网架进行形态学观察。主要观察指标:①一般形态观察②组织学观察。结果:复层组织工程皮肤在结构上与正常皮肤相似,具有真皮、表皮双层结构。改性聚乳酸网架上有双层细胞生长,生长的细胞与网架接触,并且在其表面形成较为明显而连续的细胞层。随着培养时间的延长,发生了一系列变化:表皮部分细胞层数逐渐增多,真皮部分细胞也逐渐增多,并向表皮层深入,位于表皮与网架之间。结论:双醛淀粉作为良好的增柔剂在改善聚乳酸网架的机械性能的同时,也具有良好的细胞相容性,不影响细胞的生长增殖和代谢,可以进一步用作组织工程皮肤的支架材料。  相似文献

Barley α-amylase 1 mutant (AMY) and Lentinula edodes glucoamylase (GLA) were cloned and expressed in Saccharomyces cerevisiae. The purified recombinant AMY hydrolyzed corn and wheat starch granules, respectively, at rates 1.7 and 2.5 times that of GLA under the same reaction conditions. AMY and GLA synergistically enhanced the rate of hydrolysis by ∼3× for corn and wheat starch granules, compared to the sum of the individual activities. The exo-endo synergism did not change by varying the ratio of the two enzymes when the total concentration was kept constant. A yield of 4% conversion was obtained after 25 min 37°C incubation (1 unit total enzyme, 15 mg raw starch granules, pH 5.3). The temperature stability of the enzyme mixtures was ≤50°C, but the initial rate of hydrolysis continued to increase with higher temperatures. Ca++ enhanced the stability of the free enzymes at 50°C incubation. Inhibition was observed with the addition of 10 mM Fe++ or Cu++, while Mg++ and EDTA had lesser effect. 以2个高淀粉和2个低淀粉玉米自交系为材料,分析了玉米籽粒淀粉的动态积累规律,同时对高低淀粉玉米籽粒灌浆过程中淀粉生物合成关键酶活性的动态变化及其与淀粉积累动态的相关性进行讨论分析。研究结果表明:灌浆过程中4个自交系淀粉含量变化趋势均呈sigmoid型曲线。灌浆过程中ADPG-PPase(腺苷二磷酸葡萄糖焦磷酸化酶)、SSS(可溶性淀粉合成酶)、GBSS(颗粒结合淀粉合成酶)活性均呈单峰曲线变化,峰值都出现在20~30DAP(授粉后天数)。2个高淀粉自交系的Q酶(淀粉分支酶)活性也呈单峰曲线变化,峰值也出现在20DAP,而2个低淀粉自交系的Q酶活性则呈双峰曲线变化,2个峰值分别出现在15~20DAP和30~35DAP。4个自交系籽粒淀粉的积累速率与各自交系ADPG-PPase、SSS和GBSS的活性变化呈极显著正相关。各自交系关键酶活性之间,ADPG-PPase、SSS和GBSS三者间活性变化呈极显著正相关,这3种酶活性变化与Q酶活性变化也呈不同程度的正相关。  相似文献

【目的】考察泸型酒发酵过程中酒醅细菌群落的演替规律,探讨菌群演替与环境因素变化的相关性。【方法】采用高通量测序技术分析泸型酒酒醅细菌群落的演替规律,并运用Mantel test分析不同发酵阶段的细菌群落演替与环境因素变化的相关性。【结果】酒醅发酵过程中有397个属的微生物,其中Lactobacillus、Bacillus、Weissella、Dysgonomonas、Comamonas以及Ruminococcaceae为优势属(相对丰度1.0%)。通过聚类分析可将酒醅发酵过程划分为3个阶段:阶段I (0–5 d),阶段II (6–17 d)和阶段III(18–40d),且3个阶段的酒醅菌群结构差异显著(P0.05)。Metastats分析结果表明,与阶段I相比,阶段II酒醅细菌群落中Lactobacillus和unclassifiedLactobacillaceae相对丰度显著升高(P0.05),而unclassifiedBacillaceae、 Staphylococcus、 Bacillus、 unclassified Enterobacteriaceae、 Lactococcus、Pseudomonas、Thermoactinomyces、Leuconostoc、Staphylococcus相对丰度显著降低(P0.05)。与阶段II相比,阶段III酒醅细菌群落中Lactobacillus相对丰度显著增长(P0.05),Comamonas、Acetobacter、unclassified Bacilli、Clostridium、Bacillus、Ruminococcus、unclassified Porphyromonadaceae和unclassified Streptophyta相对丰度显著下降(P0.05)。结果表明,阶段I的细菌菌群演替与酒醅温度、水分和乙醇浓度变化线性相关(P0.05);阶段II和阶段III的细菌菌群演替与酒醅温度、水分、酸度、乙醇浓度均没有相关性(P0.05)。【结论】泸型酒酒醅中细菌群落在不同发酵阶段结构差异显著,且温度、水分以及乙醇浓度对酒醅发酵前期(0–5 d)细菌群落演替具有重要作用。  相似文献
