Zn是植物必需的营养元素,同时也是一种常见的有毒重金属元素.由于长期的环境选择和适应进化,植物相应对Zn~(2+)产生了耐性,可减轻或避免Zn~(2+)的毒害.植物对锌耐性机制有:菌根和细胞膜对Zn~(2+)吸收的阻止和控制,其中控制Zn~(2+)的细胞膜跨膜转运器主要有(ZIP)类、阳离子扩散促进器(CDF)类和B-type ATPase (HMA)类;金属硫蛋白(MTs)、植物螯合素(PCs)和有机酸等Zn~(2+)螯合物质的体内螯合解毒;体内区室化分隔以及通过抗氧化系统和渗透调节物质的代谢调节等.本文从生理和分子水平上综述了植物对Zn~(2+)耐性机理的研究进展,并在此基础上提出目前存在的问题和今后研究的重点领域,为该领域的相关研究提供资料和借鉴.  相似文献

Relationships between growth of osmotically stressed intact seedlings and polyribosome levels and water status of growing tissues were examined. Sudden exposure of barley (Hordeum vulgare L. cv. Arivat) roots to a solution of ?0.8 MPa polyethylene glycol caused leaf growth to stop almost immedately, but growth resumed at a much lower rate after 0.5–1 h. In the growing region of leaves, the polyribosome: total ribosome ratio of free (non-membrane-bound) ribosomes was significantly reduced after 15 min stress, but a decrease in the large polyribosome:total polyribosome ratio occurred only after 1–2 h. Membrane-bound and free polyribosome levels both decreased to 70% of unstressed control values after 4 h stress. Recovery of total polyribosomes occurred within 1 h after relief of 4 h stress, but required 3 h after relief of 24 h stress. Stress detectably reduced the water potential and osmotic potential of growing tissue within 0.5–1.0 h, and osmotic adjustment continued for up to 10 h. Recovery of water status was incomplete after 1 h relief of a 4 h stress. In contrast, expanded blade tissues of stressed plants underwent minor changes in water status and slow decreases in polyribosomes levels. These results confirm that growing tissues of barley leaves are selectively responsive to stress, and suggest that changes in growth, water status and polyribosome levels may be initiated by the same signal. Measurements of seedling growth, polyribosome levels and water status of growing tissues of barley and wheat (Triticum aestivum L. cv. Zaragoza) leaves, etiolated pea (Pisum sativum L. cv. Alaska) epicotyl and etiolated squash (Cucurbita pepo L. cv. Elite) hypocotyl stressed with polyethylene glycol solutions of ?0.3 to ?0.8 MPa for 12 h or more showed that polyribosome levels were highly correlated with seedling growth rate as well as with tissue water and osmotic potentials, while turgor remained unchanged. These results suggest that long-term growth of osmotically stressed plants may be limited by a reduced capacity for protein synthesis in growing tissues and is not dictated by turgor loss.  相似文献

When testing for genetic differentiation the joint null hypothesis that there is no allele frequency difference at any locus is of interest. Common approaches to test this hypothesis are based on the summation of χ2 statistics over loci and on the Bonferroni correction, respectively. Here, we also consider the Simes adjustment and a recently proposed truncated product method (TPM) to combine P‐values. The summation and the TPM (using a relatively large truncation point) are powerful when there are differences in many or all loci. The Simes adjustment, however, is powerful when there are differences regarding one or a few loci only. As a compromise between the different approaches we introduce a combination between the Simes adjustment and the TPM, i.e. the joint null hypothesis is rejected if at least one of the two methods, Simes and TPM, is significant at the α/2‐level. Simulation results indicate that this combination is a robust procedure with high power over the different types of alternatives.  相似文献

This paper investigates the dependence of monoterpene emissions at the canopy scale on total leaf area and leaf distribution. Simulations were carried out for a range of hypothetical but realistic forest canopies of the evergreen Quercus ilex (holm oak). Two emission models were applied that either did (SIM-BIM2) or did not (G93) account for cumulative responses to temperature and light. Both were embedded into a canopy model that considered spatial and temporal variations of foliage properties. This canopy model was coupled to a canopy climate model (CANOAK) to determine the micrometeorological conditions at the leaf scale. Structural properties considerably impacted monoterpene emission. The sensitivities to changes in total leaf area and to leaf area distribution were found to be of similar magnitude. The two different models performed similarly on a whole-year basis but showed clear differences during certain episodes. The analysis showed that structural indices have to be carefully evaluated for proper scaling of emission from leaves to canopy. Further research is encouraged on seasonal dynamics of emission potentials.  相似文献

Macrophage metalloelastase or matrix metalloproteinase-12 (MMP-12) appears to exacerbate atherosclerosis, emphysema, aortic aneurysm, rheumatoid arthritis, and inflammatory bowel disease. An inactivating E219A mutation, validated by crystallography and NMR spectra, prevents autolysis of MMP-12 and allows us to determine its NMR structure without an inhibitor. The structural ensemble of the catalytic domain without an inhibitor is based on 2813 nuclear Overhauser effects (NOEs) and has an average RMSD to the mean structure of 0.25 Å for the backbone and 0.61 Å for all heavy atoms for residues Trp109-Gly263. Compared to crystal structures of MMP-12, helix B (hB) at the active site is unexpectedly more deeply recessed under the β-sheet. This opens a pocket between hB and β-strand IV in the active-site cleft. Both hB and an internal cavity are shifted toward β-strand I, β-strand III, and helix A on the back side of the protease. About 25 internal NOE contacts distinguish the inhibitor-free solution structure and indicate hB's greater depth and proximity to the sheet and helix A. Line broadening and multiplicity of amide proton NMR peaks from hB are consistent with hB undergoing a slow conformational exchange among subtly different environments. Inhibitor-binding-induced perturbations of the NMR spectra of MMP-1 and MMP-3 map to similar locations across MMP-12 and encompass the internal conformational adjustments. Evolutionary trace analysis suggests a functionally important network of residues that encompasses most of the locations adjusting in conformation, including 18 residues with NOE contacts unique to inhibitor-free MMP-12. The conformational change, sequence analysis, and inhibitor perturbations of NMR spectra agree on the network they identify between structural scaffold and the active site of MMPs.  相似文献

Abstract: Snowshoe hares (Lepus americanus) are an important prey species and a dominant herbivore across much of their North American range, and researchers have questioned the influences of forestry practices that alter habitat for hares and the potential community-level effects on carnivores. We examined the effects of precommercial thinning (PCT) from 1 to 11 years posttreatment on snowshoe hares. In the commercial forests of northern Maine, USA, we counted and cleared hare pellets twice a year during 2001 and 2002 on >46 km of pellet transects across 30 regenerating conifer stands (17 treated with PCT) previously treated with an aerial application of herbicide. We compared densities of snowshoe hare pellets among 3 development classes with (1 yr after thinning, 6 yr after thinning, and 11 yr after thinning) and without thinning (stands with a similar history of clearcut and herbicide treatment but no thinning). During both years, densities of hares were lower in stands treated with PCT than in similar unthinned stands across the 3 development classes and during both leaf-off and leaf-on seasons (P < 0.001). Within both thinned and unthinned stands, hare density was greatest in stands in the 1-year development class when compared to the 6-year and 11-year development classes, but a statistical difference (P = 0.048) among classes was evident only during leaf-off seasons. Precommercial thinning was associated with densities of snowshoe hares that were approximately half of those in similar unthinned stands up to at least 11 years posttreatment; however, thinned stands may retain densities of hares greater than stands managed using other forest harvesting regimes. Our results apply to core portions of stands with crop trees spaced at 1.8–2.4-m intervals following complete overstory removal and herbicide treatment. We advocate caution when applying our results to other thinning regimes or across broader spatial scales.  相似文献

In this study, 26 healthy young volunteers were submitted to 900 MHz (2 W) GSM cellular phone exposure and to sham exposure in separate sessions. The study was designed to assess cardiac regulatory mechanism in different autonomic nervous system (ANS) states during exposure to low-intensity EMF. Rest-to-stand protocol was applied to evaluate ANS in quiet condition (rest, vagal prevalence) and after a sympathetic activation (stand). The procedure is conducted twice in a double-blind design: once with a genuine EMF exposure and once with a sham exposure (at least 24 h apart). During each session three-leads electrocardiograms were recorded and RR series extracted off-line. Time domain and frequency domain HRV parameters were calculated in every phase of the protocol and during different exposures. The analysis of the data show there was no statistically significant effect due to EMF exposure both on main (i.e., RR mean) and most of the other HRV parameters. A weak interaction between some HRV parameters (i.e., SDNN, TINN, and triangular index in time domain and LF power in frequency domain analysis) and RF exposure was observed and this effect seems to be gathered around the sympathetic response to stand.  相似文献

比较了内蒙古高原荒漠区4种锦鸡儿属(Caragana)优势植物——柠条锦鸡儿、狭叶锦鸡儿、垫状锦鸡儿和荒漠锦鸡儿的生理适应特性。研究发现:4种锦鸡儿属植物中,荒漠锦鸡儿的叶片含水量、束缚水/自由水比值、水势以及水分利用效率最低,气孔导度日变化表现为与气温变化相似的早晚低、中午高的单峰曲线。4种锦鸡儿蒸腾速率日变化状况相似,均在10:00达到最大,以后逐渐降低,但日蒸腾积累值垫状锦鸡儿<荒漠锦鸡儿<柠条锦鸡儿<狭叶锦鸡儿。相比较,柠条锦鸡儿、狭叶锦鸡儿和垫状锦鸡儿对当地的水分条件有良好的适应性,能够保持较好的水分状况,水分亏缺在15%以下,而荒漠锦鸡儿叶水分亏缺日变幅较大,保水能力不及其它3种。渗透调节比较研究发现:荒漠锦鸡儿的渗透势最低,细胞质离子浓度最高,无机渗透调节物产生的渗透势所占的比例也更大,说明其低渗透势的维持主要来自无机渗透调节物质的较多积累。保护酶和自由基比较研究发现:POD和SOD活性荒漠锦鸡儿明显高于其它3个种,但CAT活性在4种植物中无显著差别;叶片自由基含量狭叶锦鸡儿>荒漠锦鸡儿>柠条锦鸡儿>垫状锦鸡儿。这些结果表明:(1)荒漠锦鸡儿对干旱环境的适应方式与其它3种不同,柠条锦鸡儿、狭叶锦鸡儿和垫状锦鸡儿以强保水能力、维持稳定的水分而适应,而荒漠锦鸡儿以更负的渗透势、更高细胞质离子浓度弥补其更多的水分消耗和对水分变化的强耐性而适应,较高的保护酶活性可能是其强耐性的生理基础之一。(2)保水能力弱的锦鸡儿种主要通过无机离子的积累,调节细胞质渗透势,保持水分平衡,这是一种相对节省能量的适应对策。  相似文献

Sesuvium portulacastrum is a halophytic species well adapted to salinity and drought. In order to evaluate the physiological impact of salt on water deficit-induced stress response, we cultivated seedlings for 12 days, in the presence or absence of 100 mmol l−1 NaCl, on a nutrient solution containing either 0 mmol l−1 or 25 mmol l−1 mannitol. Mannitol-induced water stress reduced growth, increased the root/shoot ratio, and led to a significant decrease in water potential and leaf relative water content, whereas leaf Na+ and K+ concentrations remained unchanged. The addition of 100 mmol l−1 NaCl to 25 mmol l−1 mannitol-containing medium mitigated the deleterious impact of water stress on growth of S. portulacastrum, improved the relative water content, induced a significant decrease in leaf water potential and, concomitantly, resulted in enhancement of overall plant photosynthetic activity (i.e. CO2 net assimilation rate, stomatal conductance). Presence of NaCl in the culture medium, together with mannitol, significantly increased the level of Na+ and proline in the leaves, but it had no effect on leaf soluble sugar content. These findings suggest that the ability of NaCl to improve plant performance under mannitol-induced water stress may be due to its effect on osmotic adjustment through Na+ and proline accumulation, which is coupled with an improvement in photosynthetic activity. A striking recovery in relative water content and growth of the seedlings was also recorded in the presence of NaCl on release of the water stress induced by mannitol.  相似文献
