We examine the risk model in sperm competition games for cases where female fertility increases significantly with sperm numbers (sperm limitation). Without sperm competition, sperm allocation increases with sperm limitation. We define 'average risk' as the probability q that females in the population mate twice, and 'perceived risk' as the information males gain about the sperm competition probability with individual females. If males obtain no information from individual females, sperm numbers increase with q unless sperm limitation is high and one of the two competing ejaculates is strongly disfavoured. If males can distinguish between virgin and mated females, greater sperm allocation to virgins is favoured by high sperm limitation, high q, and by the second male's ejaculate being disfavoured. With high sperm limitation, sperm allocation to virgins increases and to mated females decreases with q at high q levels. With perfect information about female mating pattern, sperm allocation (i) to virgins that will mate again exceeds that to mated females and to virgins that will mate only once, (ii) to virgins that mate only once exceeds that for mated females if q is high and there is high second male disadvantage and (iii) to each type of female can decrease with q if sperm limitation is high, although the average allocation increases at least across low q levels. In general, higher sperm allocation to virgins is favoured by: strong disadvantage to the second ejaculate, high sperm limitation, high average risk and increased information (perceived risk). The analysis of carbon isotope discrimination (Δ) in crop plant remains from archaeological sites may help to assess water availability for early agriculture. This study presents the analysis of Δ in seeds of naked wheat (Triticum aestivum/durum), lentil (Lens orientalis/culinaris), and flax (Linum sp.) found at the archaeological site of Tell Halula in the valley of the Middle Euphrates (Syria). This Neolithic site is the oldest in this region of the Fertile Crescent where the cultivation of domesticated plants has been reported, with seed remains ranging from 9550 to 8465 BP. Most of the seeds analysed showed Δ values greater than 16 ‰, reaching 20 ‰ for some samples of flax. For wheat, Δ values were much higher than those reported in present-day (1996) durum wheat crops cultivated under rainfed conditions in north-west Syria under environments with somewhat higher rainfall than Tell Halula. Similarly, grains of present-day (1997) barley cultivated in the archaeological site also showed lower values than those found in archaeological kernels. An empirical relationship between Δ of mature kernels and total precipitation (plus irrigation where applicable) from heading to maturity (r2 = 0.82, n = 11) was established for durum wheat, currently cultivated in different environments of the Mediterranean basin. The resulting relationship was applied to the data on Δ of wheat fossil kernels from Tell Halula to estimate the accumulated water inputs during the time (about 6 weeks) the kernels were produced. Calculated water inputs for wheat during early agriculture were (over 110 mm) at least 5 times higher than current-day rainfall accumulated in Tell Halula during the same phenological period. These results strongly suggest that early agriculture wheat was cultivated at Tell Halula under much wetter conditions than are currently to be found in the area. The presence of flax and its very high Δ values also support this conclusion. Whether such humid conditions during cultivation were due to moister conditions prevailing at this time, by planting in alluvial areas or by irrigation works is discussed.  相似文献

A major challenge in managing depression is that antidepressant drugs take a long time to exert their therapeutic effects. For the development of faster-acting treatments, it is crucial to get an improved understanding of the molecular mechanisms underlying antidepressant mode of action. Here, we used a novel mass spectrometry-based workflow to investigate how antidepressant treatment affects brain protein turnover at single-cell and subcellular resolution. We combined stable isotope metabolic labeling, quantitative Tandem Mass Spectrometry (qTMS) and Multi-isotope Imaging Mass Spectrometry (MIMS) to simultaneously quantify and image protein synthesis and turnover in hippocampi of mice treated with the antidepressant paroxetine. We identified changes in turnover of individual hippocampal proteins that reveal altered metabolism-mitochondrial processes and report on subregion-specific turnover patterns upon paroxetine treatment. This workflow can be used to investigate brain protein turnover changes in vivo upon pharmacological interventions at a resolution and precision that has not been possible with other methods to date. Our results reveal acute paroxetine effects on brain protein turnover and shed light on antidepressant mode of action.  相似文献

Cat gene expression has been investigated following PEG-mediated plasmid uptake into barley protoplasts. The uptake conditions optimised for transient expression were employed for stable transformation. Transformed protoplast-derived calli of the cvs. Dissa and Igri, were selected on medium containing G418 at 40 g ml–1 or kanamycin sulphate at 250 g ml–1. Absolute transformation frequencies of 28.9×10–5 and 21.3×10–5 were recorded for Dissa with kanamycin sulphate and G418 selection, respectively. The frequency for Igri was 11.5×10–5 with G418 selection. An extracellular protease was produced by Arthrobacter ramosus isolated from the alkaline lake of Lonar, Buldhana District of Maharashtra, India when grown on a synthetic medium of pH 10 containing casein. The optimum conditions for production were 3.0% initial casein concentration, 2% inoculum of 1 × 108 cells/ml, pH 9.0, temperature 30 °C and shaken culture conditions. The protease was purified by ammonium sulphate precipitation followed by Sephadex G-100 chromatography. Two proteases viz. Arthro I and Arthro II, having molecular weights 21 and 11.4 kDa respectively were isolated. The Arthro II fraction had K m 395 g/ml and V max 10.55 g/min for azocasein. The maximum activity of enzyme was at 55 °C and pH 8. It was thermostable (up to 80 °C), alkali stable (pH 12) and stable in commercial detergent. The enzyme may contain a thiol group at the active site.  相似文献

It is generally well understood that some ecological factors select for increased and others for decreased dispersal. However, it has remained difficult to assess how the evolutionary dynamics are influenced by the spatio-temporal structure of the environment. We address this question with an individual-based model that enables habitat structure to be controlled through variables such as patch size, patch turnover rate, and patch quality. Increasing patch size at the expense of patch density can select for more or less dispersal, depending on the initial configuration. In landscapes consisting of high-quality and long-lived habitat patches, patch degradation selects for increased dispersal, yet patch loss may select for reduced dispersal. These trends do not depend on the component of life-history that is affected by habitat quality or the component of life-history through which density-dependence operates. Our results are based on a mathematical method that enables derivation of both the evolutionary stable strategy and the stationary genotype distribution that evolves in a polymorphic population. The two approaches generally lead to similar predictions. However, the evolutionary stable strategy assumes that the ecological and evolutionary time scales can be separated, and we find that violation of this assumption can critically alter the evolutionary outcome.  相似文献

Salt marshes and shallow-water macroalgal beds are known to provide nursery habitat for many species of fish and invertebrates. The role of these habitats as refuge from predation is well established, but the degree to which indigenous primary production within the nursery provides food for growth and development of estuarine species remains unresolved. In this study, we tested the hypothesis that juvenile blue crabs depend on indigenous primary production, directly or indirectly, during their entire stay within the nursery. To test this hypothesis, we conducted isotopic studies and stomach content analyses of juveniles from habitats near the mouth of Delaware Bay and from an adjacent lagoonal estuary (ca. 39.5° N, 75.1° W). Primary producers, marsh detritus, various life-history stages of blue crabs and potential prey species were sampled in the main estuary and in an adjacent marsh during the summer and early fall of two consecutive years. Newly settled juveniles (<15 mm carapace width) from the marsh were about 1.8‰ lighter in carbon (−17.2‰) relative to larger juveniles from the marsh (15–30 mm carapace width) and appeared to have retained a carbon isotopic signature indicative of the phytoplankton-based food web associated with larval stages. However, the signature of juveniles changed as a function of size. Large juveniles and crabs >60 mm were enriched in δ13C (−14.7 ± 0.1‰) compared to small crabs, suggesting a gradual shift in diet from a planktonic to a detritus-based food web with increasing size. As with crabs from Delaware Bay, the δ13C signature of juvenile crabs sampled from macroalgal beds in the lagoonal estuary (Rehoboth Bay) changed as a function of size. Also, δ13C ratios of crabs varied among the various species of macroalgae. The δ15N composition of primary producers in the marsh and main estuary also was reflected in the δ15N values of crabs and other benthic consumers in the respective habitats. Results of stomach-content analysis in this study were consistent with isotope data. Observed changes in prey preferences were related to changes in size of juvenile crabs and also differed among habitats. Gut content analyses of the three size classes of juveniles in macroalgal beds from Rehoboth Bay indicated that the crabs depend heavily on various amphipod species that occur on the seaweeds. These amphipods graze directly on the macroalgae and are among the most abundant invertebrates in the macroalgal beds. This implies a direct trophic relationship between the juvenile crabs and the macroalgae. In summary, our study provides strong evidence that the value of nursery areas such as salt marshes and macroalgal beds goes beyond that of providing refuge from predation, and that species using these nurseries (e.g. juvenile blue crabs) are ultimately dependent on primary production originating in benthic plants indigenous to the nursery.  相似文献
