The effect of time and spatial averaging on 15N chemical shift/1H-15N dipolar correlation spectra, i.e., PISEMA spectra, of -helical membrane peptides and proteins is investigated. Three types of motion are considered: (a) Librational motion of the peptide planes in the -helix; (b) rotation of the helix about its long axis; and (c) wobble of the helix about a nominal tilt angle. A 2ns molecular dynamics simulation of helix D of bacteriorhodopsin is used to determine the effect of librational motion on the spectral parameters. For the time averaging, the rotation and wobble of this same helix are modelled by assuming either Gaussian motion about the respective angles or a uniform distribution of a given width. For the spatial averaging, regions of possible 15N chemical shift/1H-15N dipolar splittings are computed for a distribution of rotations and/or tilt angles of the helix. The computed spectra show that under certain motional modes the 15N chemical shift/1H-15N dipolar pairs for each of the residues do not form patterns which mimic helical wheel patterns. As a result, the unambiguous identification of helix tilt and helix rotation without any resonance assignments or on the basis of a single assignment may be difficult.  相似文献

槭属(AcerL)属槭树科(Aceraceae),200种,分布于亚、欧、北美和非洲北缘。本文研究了槭属的系统演化、地理分布、起源与扩散。认为:(1)槭树科与无患子科关系密切,槭属是槭树科2属中较进化的类群。(2)在原始而典型的槭属植物的基础上,槭属沿花的各部减少,有的器官甚至向完全退化的方向演化,但也有少数向增加数目的方向特化。(3)讨论了槭属4亚属23组的演化趋势,并绘制出其系统演化图。(4)槭属起源于侏罗纪的中国四川东部、湖北、湖南及其邻近地区,并向西、东北和南方扩散而进入西亚、欧洲、非洲北缘、北美洲和马来半岛至印尼。  相似文献

Progress curves of cassava mosaic virus disease (CMD) and populations of the whitefly vector (Bemisia tabaci) were assessed using four cassava varieties grown alone and as a random mixture in two experiments established under epidemic conditions at a site near Kampala in southern Uganda. There were significant differences in final CMD incidence and in the areas under the disease progress curves between varieties when grown alone and as a mixture in both experiments. Variety Ebwanateraka had the highest incidence and SS4 the lowest, even though it supported the largest populations of adult whiteflies. The overall incidence of CMD in the mixture was similar to that in pure stands of the partially resistant Nase 2 and greater than in the resistant Migyera and SS4. Compared to pure stands, incidence of CMD in each component of the mixture was reduced significantly only in Ebwanateraka, whereas vector populations were less only in SS4 and Nase 2. On several observation dates the actual incidence of CMD and populations of adult whiteflies in the mixture were significantly less than expected values estimated from the results for the four varieties when each was grown alone. A highly significant positive relationship was established for each variety between peak populations of adult whitefly and leaf area index at the time. The implications of the findings and the scope for future research on the use of varietal mixtures for the management of CMD are discussed.  相似文献

In three field experiments in 1985 and 1986, we studied the effect of the date of primary infection on the spread of beet yellows closterovirus (BYV) and beet mild yellowing luteovirus (BMW) from artificially inoculated sugar beet plants. Laboratory-reared vector aphids, Myzus persicae, were placed on these sources of virus. There was no substantial natural immigration of vectors or viruses. In two experiments, one with BMYV in 1985 and the other in BYV in 1986, populations of vector aphids remained low and there was little virus spread, i.e. c. 50 infected plants from one primarily infected source. The cause of this small amount of spread was the low number of vector aphids. In the third experiment, with BYV in 1986, large populations of M. persicae developed and there was substantial virus spread: c. 2000 infected plants in the plots which were inoculated before canopy closure. In later-inoculated plots in the same experiment, there was much less spread: c. 100 infected plants per virus source plant. Differences between fields in predator impact are implicated as the most probable factor causing differences in vector establishment and virus spread between these three experiments. Virus spread decreased with later inoculation in all three experiments. A mathematical model of virus spread incorporating results from our work has been used to calculate how the initial proportion of infected plants in a crop affects the final virus incidence. This model takes into account the effect of predation on the development of the aphid populations. The processes underlying the spread and its timing are discussed.  相似文献

Two field experiments examined the effect of straw spread on the soil surface on the incidence of bean yellow mosaic potyvirus (BYMV) in plots of narrow-leafed lupin (Lupinus angustifolius) sown at narrow (17.5 cm) vs wide (35 cm) row spacing and low (25–30 kg/ha) vs medium (50–60 kg/ha) seeding rates. Virus ingress was by vector aphids flying from adjacent pastures dominated by subterranean clover. In Expt 1, in which BYMV infection was extensive, straw greatly decreased the rate and amount of virus spread regardless of row spacing or plant density, decreasing infection more than 70% by the final assessment date. This effect of straw was attributed to decreased landing rates of incoming vector alates. In the plots without added straw, narrow row spacing decreased BYMV % infection by 38% by the last assessment date. Sowing at the medium seeding rate also decreased infection. The effect of wide row spacing seemed due to delayed canopy closure between rows which is likely to have increased the landing of aphids while the effect of medium seeding rate was attributed partly to the dilution effect of greater plant numbers and partly to the effects of partial canopy development in decreasing landing rates. In Expt 2, in which the incidence of BYMV infection was low, added straw again decreased BYMV spread, but by only 25–27% at final assessment; there were no effects of row spacing or seeding rate. In both experiments, an additional “reference” treatment was included which had a high (90–100 kg/ha) seeding rate, narrow rows and no straw. The dense canopy it developed also decreased BYMV incidence but less than in the plots with added straw in Expt 1. In Expt 1, adding straw and the resulting decrease in plants killed by BYMV, were associated with an overall increase in lupin grain yield of 20%. The greater plant densities resulting from the medium seeding rate also increased grain yield but row spacing did not affect it significantly. These results indicate that retaining stubble on the soil surface at seeding will assist in management of BYMV infection in lupin crops but that wide row spacing in the absence of retained stubble is undesirable.  相似文献

1. Stream flow is a major vector for zebra mussel spread among inland lakes. Veligers have been found tens to hundreds of km from upstream source lakes in unvegetated stream and river systems. It has been suggested, however, that the downstream transport of zebra mussels is restricted by wetland ecosystems. We hypothesized that vegetated waterways, (i.e. wetland streams) would hinder the downstream dispersal of zebra mussels in connected inland lake systems. 2. Veliger abundance, recruitment and adult mussels were surveyed in four lake‐wetland systems in southeastern Michigan, U.S.A. from May to August 2006. Sampling was conducted downstream of the lakes invaded by zebra mussels, beginning at the upstream edge of aquatic vegetation and continuing downstream through the wetland streams. 3. Veliger abundance decreased rapidly in vegetated waterways compared to previously reported rates of decrease in non‐vegetated streams. Veligers were rarely found more than 1 km downstream from where vegetation began. Newly recruited individuals and adults were extremely rare beyond open water in the wetland systems. 4. Densely vegetated aquatic ecosystems limit the dispersal of zebra mussels downstream from invaded sources. Natural, remediated and constructed wetlands may therefore serve as a protective barrier to help prevent the spread of zebra mussels and other aquatic invasive species to other lakes and ecosystems.  相似文献

In this paper we reconstruct the location of the last glacial refugia and postglacial spread of Abies throughout Europe based on combined pollen and macrofossil data. More than 208 pollen sequences available in the European Pollen Database (EPD) and 38 macrofossil sites are used to produce distribution maps encompassing a time span between 38000 and 5500 years B.P. The investigation excludes more recent periods, because these could be strongly influenced by human impact. The pollen data presented here confirm long-lasting refugial areas such as southern Italy and Greece already described in previous studies. The combined pollen/macrofossil dataset identifies the Pyrenees as a further important refugium. In addition the pollen data indicate potential refugia in south-east France and north-west Italy. Possible migration tracks of Abies are discussed by comparing the palaeobotanical evidence with isozyme studies on gene markers of recent fir populations.Electronic Supplementary Material Supplementary material is available in the online version of this article at http://dx.doi.org/10.1007/s00334-004-0049-4.  相似文献

Severe acute respiratory syndrome coronavirus (SARS-CoV) is the etiological agent of SARS. Analysis of SARS-CoV spike glycoprotein (S) using recombinant plasmid and virus infections demonstrated that the S-precursor (proS) exists as a approximately 190 kDa endoplasmic reticulum form and a approximately 210 kDa Golgi-modified form. ProS is subsequently processed into two C-terminal proteins of approximately 110 and approximately 80 kDa. The membrane-bound proprotein convertases (PCs) furin, PC7 or PC5B enhanced the production of the approximately 80 kDa protein. In agreement, proS processing, cytopathic effects, and viral titers were enhanced in recombinant Vero E6 cells overexpressing furin, PC7 or PC5B. The convertase inhibitor dec-RVKR-cmk significantly reduced proS cleavage and viral titers of SARS-CoV infected cells. In addition, inhibition of processing by dec-RVKR-cmk completely abrogated the virus-induced cellular cytopathicity. A fluorogenically quenched synthetic peptide encompassing Arg(761) of the spike glycoprotein was efficiently cleaved by furin and the cleavage was inhibited by EDTA and dec-RVKR-cmk. Taken together, our data indicate that furin or PC-mediated processing plays a critical role in SARS-CoV spread and cytopathicity, and inhibitors of the PCs represent potential therapeutic anti-SARS-CoV agents.  相似文献

Epidemics of marine pathogens can spread at extremely rapid rates. For example, herpes virus spread through pilchard populations in Australia at a rate in excess of 10 000 km year?1, and morbillivirus infections in seals and dolphins have spread at more than 3000 km year?1. In terrestrial environments, only the epidemics of myxomatosis and calicivirus in Australian rabbits and West Nile Virus in birds in North America have rates of spread in excess of 1000 km year?1. The rapid rates of spread of these epidemics has been attributed to flying insect vectors, but flying vectors have not been proposed for any marine pathogen. The most likely explanation for the relatively rapid spread of marine pathogens is the lack of barriers to dispersal in some parts of the ocean, and the potential for long‐term survival of pathogens outside the host. These findings caution that pathogens may pose a particularly severe problem in the ocean. There is a need to develop epidemic models capable of generating these high rates of spread and obtain more estimates of disease spread rate.  相似文献
