Summary DNA fingerprints generated by the Jeffreys' probes, 33.6 and 33.15, indicated the presence of minisatellite-like sequences in the red clover genome. The fingerprints generated by probe 33.6 gave less background and fewer but better defined bands than those obtained with probe 33.15. Assay of a regenerative somaclonal variant (F49R) by DNA fingerprinting with probe 33.6 detected mutation that was unlinked to the regenerative trait. The fingerprints obtained under the applied conditions also demonstrated genetic stability of consecutive generations of the regenerants in tissue culture. DNA fingerprints of F1 plants revealed that each polymorphic band was inherited from either one or the other parent. Both probes distinguished individual-specific genotypes in seven cultivars of red clover. Greater variability in DNA fingerprints was detected between (V=0.899) than within (0.417≤V≤0.548) cultivars.  相似文献

The nonstructural protein NSm of tomato spotted wilt virus (TSWV) has been identified as the avirulence determinant of the tomato single dominant Sw-5 resistance gene. Although Sw-5 effectiveness has been shown for most TSWV isolates, the emergence of resistance-breaking (RB) isolates has been observed. It is strongly associated with two point mutations (C118Y or T120N) in the NSm viral protein. TSWV-like symptoms were observed in tomato crop cultivars (+Sw-5) in the Baja California peninsula, Mexico, and molecular methods confirmed the presence of TSWV. Sequence analysis of the NSm 118–120 motif and three-dimensional protein modelling exhibited a noncanonical C118F substitution in seven isolates, suggesting that this substitution could emulate the C118Y-related RB phenotype. Furthermore, phylogenetic and molecular analysis of the full-length genome (TSWV-MX) revealed its reassortment-related evolution and confirmed that putative RB-related features are restricted to the NSm protein. Biological and mutational NSm 118 residue assays in tomato (+Sw-5) confirmed the RB nature of TSWV-MX isolate, and the F118 residue plays a critical role in the RB phenotype. The discovery of a novel TSWV-RB Mexican isolate with the presence of C118F substitution highlights a not previously described viral adaptation in the genus Orthotospovirus, and hence, the necessity of further crop monitoring to alert the establishment of novel RB isolates in cultivated tomatoes.  相似文献

A survey of the parasitoids ofIllinoia liriodendri (Monell) in northern California conducted from 1988–1990 revealed the presence of 12 primary and 14 hyper-parasitoid species. The most common primary parasitoid wasAphidius polygonaphis (Fitch), which was imported from the eastern United States in the 1970's and is now established throughout the area. New host records were noted forA. ervi Haliday,A. avenaphis (Fitch), Praon occidentale Baker,P. unicum Smith,Diaeretiella rapae M'Intosh,Lysiphlebus testaceipes (Cresson), andMonoctonus nervosus (Haliday) (all Hymenoptera: Braconidae: Aphidiinae), andAphelinus sp. nr.asychis Walker (Hymenoptera: Aphelinidae). The most common hyperparasitoid species werePachyneuron aphidis (Bouché) andAsaphes californicus Girault (both Hymenoptera: Pteromalidae). Cotyledonary-stage embryos of Haifa white clover, collected 13 days after cross-pollination, were induced to form adventitious shoots primarily from the hypocotyl region. The culture medium used for the production of adventitious shoots contained 5 M thidiazuron and 0.5 M -naphthaleneacetic acid. Numerous shoot meristems were produced within the first week, discrete shoots developed by week three, small plantlets by week eight, and whole plants in soil by week ten. 95–100% of all embryos, regardless of genotype, produced adventitious shoots within four weeks with an average production of 17.5 shoots per embryo. The majority of shoots (on average 77%) were easily converted to whole plants in soil. Reproduction and development of Pratylenchus penetrans were studied on genetically transformed ladino clover roots. Solitary females developing on transformed roots in nutrient gellan gum medium (pH 5.5) deposited 1.2, 1.5, 1.6, 1.8, and 2.0 eggs per day at the respective temperatures of 17, 20, 25, 27, and 30 °C. The number of eggs deposited was highly correlated with temperature. A reduction in egg-laying rates at the start of hatching was observed at all temperatures. Juvenile mortality was higher at 17 °C (50.4%), 20 °C (50.3%), and 30 °C (58.4%) than at 25 °C (34.6%) and 27 °C (37.6%). Life-cycle (egg deposition to egg deposition) duration was 46, 38, 28, 26, and 22 days at the respective temperatures. The developmental zero degrees (°C) and the effective accumulative temperatures (degree-days) required for hatching, female emergence, and onset of oviposition (completion of one generation) of P. penetrans were estimated to be 2.7 and 200, 4.2 and 548, and 5.1 and 564, respectively. Pratylenchus penetrans reproduces over a wide range of temperatures.  相似文献

To test the hypothesis that early maturation of barley affords it some resistance to cereal aphids, the colonisation, survival, growth, reproduction and emigration of M. dirhodum on barley was measured and compared with previously published results for this aphid on wheat.
Barley plants were colonised as readily as wheat plants. Survival to maturity was similar on both hosts. Relative growth rate was higher on wheat than on barley for most of the season while reproduction was similar on both. Emigration, measured alate production, was significantly higher on mature wheat than on mature barley.
It is proposed that the increase of M. dirhodum populations on barley is normally terminated early in the year directly plant maturation and consequent drying out of the leaves, resulting in the death of aphids. This is different to the situation on wheat, which matures later, and where emigration following plant induced alate induction has been shown to be the major factor affecting the decline in population growth.
These results support the hypothesis that early maturation of barley provides some resistance to M. dirhodum.  相似文献

Contact of adjacent root hairs of seedlings of white clover ( Trifolium repens L. cv. Ladino and Louisiana Nolin) led to cell-cell adhesion of root hair tips. The involvement of the root lectin, trifoliin A, in this phenomen was examined in slide cultures of axenically grown seedlings. Trifoliin A was detected by indirect immunofluorescence on root hair tips, which had adhered to one another. Seedlings grown under conditions which specifically reduce the levels of this lectin on the root surface (e.g., in the presence of 15 m M NO3– or 5 m M 2-deoxy- d -glucose) had significantly fewer adhesions of root hair tips. In addition, flushing the slide cultures with 20 m M 2-deoxy- d -glucose resulted in an immediate 4-fold reduction in frequency of tip adhesions. These results are consistent with the lectin cross-bridging model, which predicts that cell-cell adhesions would occur when trifoliin A on root hair tips contacts complementary glycosylated receptors on neighboring root hairs.  相似文献

Feeding experiments have demonstrated that 7,2′-dihydroxy-4′-methoxy-isoflavone-[14C-Me] and -isoflavanone-[14C-Me] are extremely efficient precursors of the phytoalexin demethylhomopterocarpin in Cu2+-treated red clover seedlings. Neither of these compounds, nor demethylhomopterocarpin-[14C-Me], was incorporated into a second pterocarpan phytoalexin, maackiain. 3-Hydroxy-9-methoxypterocarp-6a-ene-[14C-Me] was a poor precursor of both pterocarpans. A biosynthetic pathway to demethylhomopterocarpin via 2′-hydroxylation of formononetin (7-hydroxy-4′-methoxyisoflavone) and subsequent reduction to the isoflavanone is proposed. The conversion of this isoflavanone into the pterocarpan may involve the corresponding isoflavanol and a carbonium ion intermediate. The branch-point to maackiain is probably at the formononetin stage. The presence of two coumestans, 9-O-methylcoumestrol and medicagol, previously unreported in red clover, is demonstrated. Biosynthetic implications are discussed.  相似文献

Oviposition by a parasitoid wasp in an already parasitized host (= superparasitism) may result in larval competition and the loss of offspring. Consequently, in solitary species, the decision to superparasitize should be based on the probability of offspring survival. Females of Aphidius ervi Haliday and A. smithi Sharma & Subba Rao (Hymenoptera: Aphidiidae), two parasitoids of the pea aphid, discriminated between conspecific-and heterospecific-parasitized hosts. Both species showed partial preference for aphids previously attacked by A. smithi, a result predicted from the fact that A. ervi is superior to A. smithi in larval competition under most conditions. Females of A. smithi selectively attacked conspecific-parasitized pea aphids when given a choice between these and self-parasitized hosts. Results indicated that females of both species responded to an external pheromone-like marker to discriminate between selfconspecific-and heterospecific-parasitized aphids. In addition, A. smithi may use internal cues to recognize, and avoid oviposition in, aphids previously parasitized by A. ervi.

Par conséquent, dans les espèces solitaires, la décision de superparasiter devrait être basée sur la probabilité de survie de la progéniture. Les femelles de Aphidius ervi Haliday et de A. smithi Sharma & Subba Rao (Hymenoptera: Aphidiidae), deux parasites du puceron du pois, ont discriminé entre les hôtes parasités par des individus de la même espèce et ceux parasités par des individus d'une espèce différente. Les deux espèces de parasite ont montré une préférence partielle pour les pucerons préalablement parasités par A. smithi, résultat qui était prévisible du fait que, dans la majorité des cas, A. ervi est supérieur à A. smithi dans la compétition entre larves. Les femelles de A. smithi ont sélectivement attaqué les pucerons parasités par un individu de la même espèce lorsqu'il leur était donné de choisir parmi ceux-ci et ceux parasités par les même individu. Les résultats indiquaient que les femelles des deux espèces répondaient à une sorte de phéromone externe pour distinguer les pucerons parasités par le même individu de ceux parasités par un individu de la même espèce et ceux parasités d'une autre espèce. En plus, A. smithi pourrait utiliser des signaux internes pour reconnaître et éviter l'oviposition dans les pucerons préalablement parasités par A. ervi.

The reproductive potential of Meloidogyne graminicola was compared with that of M. incognita on Trifolium species in greenhouse studies. Twenty-five Trifolium plant introductions, cultivars, or populations representing 23 species were evaluated for nematode reproduction and root galling 45 days after inoculation with 3,000 eggs of M. graminicola or M. incognita. Root galling and egg production by the two root-knot nematode species was similar on most of the Trifolium species. In a separate study, the effect of initial population densities (Pi) of M. graminicola and M. incognita on the growth of white clover (T. repens) was determined. Reproductive and pathogenic capabilities of M. graminicola and M. incognita on Trifolium spp. were similar. Pi levels of both root-knot nematode species as low as 125 eggs per 10-cm-d pots severely galled white clover plants after 90 days. Meloidogyne graminicola has the potential to be a major pest of Trifolium species in the southeastern United States.  相似文献
