The hippocampus is a center for learning and memory as well as a target of Alzheimer's disease in aged humans. Synaptic modulation by estrogen is essential to understand the molecular mechanisms of estrogen replacement therapy. 过氧亚硝酸阴离子(ONOO-)是一种性质活泼的自由基,可引起强的氧化性损伤,介导了一氧化氮(NO)的大部分毒性作用.应用全细胞膜片钳技术,探讨ONOO-对脑片海马神经元电压门控钠通道电流(INa)和神经元兴奋性的影响.结果表明,ONOO-供体SIN-1(10,500,2000μmol/L)可浓度依赖性抑制INa电流峰值.SIN-1与ONOO-清除剂尿酸共处理,并不影响INa.500μmol/L的SIN-1可使INa的I-V曲线上移,并可抑制其失活后恢复过程,但对INa的激活和失活过程无影响.SIN-1还可抑制动作电位发放频率和幅值.脑片预处理腺苷酸环化酶(adenylate cyclase,AC)抑制剂MDL-12,330A(25μmol/L)和NEM(50μmol/L)对SIN-1的作用无影响.然而,预处理鸟苷酸环化酶(CG)抑制剂ODQ可抑制SIN-1对INa的作用.以上结果提示,ONOO-通过cGMP-INa-AP信号级联系统作用于海马神经元,与PKA和蛋白巯基亚硝化途径无关,这可能是ONOO-神经毒性的机制之一.  相似文献

应用双色荧光原位杂交的方法,国内首次报道一例特殊inv(Y)异常的性质,探讨Y染色体倒位结构异常的形成机理以及与习惯性流产临床表型的关系。应用 Biotin-11-dUTP标记的Y染色体短臂断裂点Yp11.3探针(编号889)和CY3标记的Y染色体长臂断裂点Yq12远端异染色质区探针(编号PY3.4),对1例G显带核型分析为[46, XY(90%) / 46, X, inv(Y)(p11.3;q12)]的平衡易位携带者进行双色荧光原位杂交研究。双色FISH结果显示,该易位携带者异常核型比例为22%,稍高于G显带分析中确定的比例。而且,除G显带检测出的倒位类型外,又有两种类型的倒位,其中涉及到常规显带技术难以检测出的染色单体型倒位。3种倒位类型的存在说明该患者inv(Y)断裂点呈不均一性。FISH技术是一种能准确可靠检测出染色体倒位形成的重要手段。   相似文献

Uptake ofl-[35S]cysteic acid (L-CA) in rat synaptic membrane vesicles was investigated. Preincubation with either 10 mMl-glutamic acid (L-Glu), 25 mM L-CA, 10 mMdl-homocysteic acid, or 25 mMdl-2-amino-4-phosphonobutyrate on membrane vesicles enhanced L-[35S]CA and L-[3H]Glu uptake. Na+ (5 mM) and omission of Cl from the assay medium decreased L-[35S]CA uptake into both 10 mM L-Glu-loaded and non-loaded membrane vesicles. The anion transport blockers, 4-acetamide-4-isothiocyano-2,2-disulfonic acid stibene (SITS) and 4,4-diisothiocyano-2,2-disulfonic acid stilbene (DIDS), inhibited L-[35S]CA uptake in a dose-dependent manner. The maximal uptake rate for L-[35S]CA was decreased by 50 M SITS, while the apparent Km value of L-CA was not changed. SITS increased the EC50 value of Cl for L-[35S]CA uptake from 5 mM to 10 mM with reduction of the maximal effect. 目的 :从离子通道角度研究吗啡对中枢神经系统兴奋性及抑制性突触传递的作用并探讨其机制。方法 : 原代培养新生Wistar大鼠的海马神经元。采用膜片钳技术研究吗啡对其兴奋性及抑制性突触后电流及谷氨酸诱发电流的影响。结果 :①吗啡可明显增强海马神经元兴奋性突触传递 ,加吗啡后自发兴奋性突触后电流 (sEPSC)的发放频率增加了 ( 2 0 7.8± 2 0 .9) %。此作用可被阿片受体阻断剂纳洛酮阻断 (P <0 .0 1) ;②吗啡对微小兴奋性突触后电流 (mEPSC)的发放频率及谷氨酸诱发电流的幅度没有明显影响 (P >0 .0 5 ) ;③吗啡可明显抑制神经元自发抑制性突触后电流 (sIPSC) ,纳洛酮可拮抗吗啡作用 (n =13 ,P <0 .0 1)。结论 :实验结果提示吗啡对海马神经元的兴奋作用不是由于吗啡直接作用于兴奋性氨基酸—谷氨酸突触传递过程 ,而是可能由于抑制了抑制性中间神经元 ,间接产生的兴奋作用。  相似文献

In most nonneural systems, platelet-activating factor (PAF) receptor effects are mediated by G-proteins that are often pertussis toxin-sensitive. The activation of pertussis toxin-sensitive G-proteins linked to PAF receptors results in the mobilization of intracellular calcium, at least in part, through the second messenger inositol triphosphate. We have sought to determine if a pertussis toxin-sensitive G-protein is involved in the PAF receptor-mediated phenomena of growth cone collapse and of synaptic enhancement in primary neuronal culture. Using infrared differential interference contrast microscopy and patch-clamp recording techniques, pertussis toxin, but not the inactive B oligomer of the toxin, was found to block both the growth cone collapse and the enhanced synaptic release of excitatory transmitter induced by a nonhydrolyzable PAF receptor agonist, making it likely that Go, Gq, or Gi is the G-protein transducer of PAF receptors in primary neurons. We believe that PAF acts directly on neuronal receptors, which are linked to pertussis toxin-sensitive G-proteins, on the tips of developing neurites, and on presynaptic nerve terminals, leading to growth cone collapse and enhanced synaptic release of transmitter.  相似文献

The group III metabotropic glutamate receptor subtype 7 (mGlu7) is an important regulator of glutamatergic and GABAergic neurotransmission and known to mediate emotionality and male social behavior. However, a possible regulatory role in maternal behavior remains unknown to date. Adequate expression of maternal behavior is essential for successful rearing and healthy development of the young. By understanding genetic and neural mechanisms underlying this important prosocial behavior, we gain valuable insights into possible dysregulations. Using genetic ablation as well as pharmacological modulation, we studied various parameters of maternal behavior in two different mouse strains under the influence of mGlu7. We can clearly show a regulatory role of mGlu7 in maternal behavior. Naïve virgin female C57BL/6 mGlu7 knockout mice showed more often nursing postures and less spontaneous maternal aggression compared to their heterozygous and wildtype littermates. In lactating C57BL/6 wildtype mice, acute central activation of mGlu7 by the selective agonist AMN082 reduced arched back nursing and accelerated pup retrieval without affecting maternal aggression. In addition, in lactating CD1 wildtype mice the selective mGlu7 antagonist XAP044 increased both pup retrieval and maternal aggression. With respect to receptor expression levels, mGlu7 mRNA expression was higher in lactating vs virgin C57BL/6 mice in the prefrontal cortex, but not hypothalamus or hippocampus. In conclusion, these findings highlight a significant role of the mGlu7 receptor subtype in mediating maternal behavior in mice. Region‐dependent studies are warranted to further extend our knowledge on the specific function of the brain glutamate system in maternal behavior.  相似文献

As practitioners promote passive restoration as a complementary approach to technical reclamation, it is imperative to know its drivers. Although the consequences of endozoochory can be crucial to passive restoration success, few experimental studies assess the use of heavily disturbed sites by seed dispersers such as carnivores and how the seeds they bring in emerge and survive. Using an indoor sowing experiment conducted in a quarry located within a natural park in Portugal, we examined for the first time how carnivore endozoochorous seeds collected in the quarry potentially influence its passive restoration, through effects on plant emergence and survival. Also, we tested whether sowing date and water soaking, relevant factors when sowings are to be carried out, would affect seedling emergence and mortality rates when compared with the effect of endozoochory. Our target species were included in the revegetation plan of the quarry, of which endozoochorous seeds of Carob tree (Ceratonia siliqua) were collected in sufficient number for analysis. Irrespective of the carnivore species, endozoochorous carob seeds performed similarly to untreated seeds regarding emergence rates. Endozoochorous carob seedlings showed greater mortality rates but the net result for the plant can still be the colonization of recently vacant habitats by a large proportion of viable seeds. Concerning sowing date, the later carob seeds were sown over the fruit‐ripening season the faster seedlings emerged. Water soaking increased emergence rate by 6.5 times. Broadly, sowings with previous soaking and carnivore‐mediated seed dispersal of this dry‐fruited tree can jointly enhance quarry restoration.  相似文献
