Gene expression throughout the different stages of Alzheimer's disease was analysed in samples from cerebral cortex. The gene encoding the voltage-gated potassium channel Kv3.4 was already overexpressed in early stages of the disease, and in advanced stages Kv3.4 was present at high levels in neurodegenerative structures. This subunit regulates delayed-rectifier currents, which are primary determinants of spike repolarization in neurones. In unique samples from a patient with Alzheimer's disease whose amount of amyloid plaques was decreased by beta amyloid immunization, Kv3.4 was overexpressed. The channel subunit was expressed in the neuropil, in the remaining conventional plaques in the frontal cortex and in collapsed plaques in the orbitary cortex. Therefore, amyloid deposition in plaques does not seem to be responsible for the increase in Kv3.4 levels. Nevertheless, Kv3.4 up-regulation is related to amyloid pathology, given that transgenic mice with the Swedish mutation of amyloid precursor protein showed increased expression of Kv3.4. Up-regulation of voltage-gated potassium channel subunits alters potassium currents in neurones and leads to altered synaptic activity that may underlie the neurodegeneration observed in Alzheimer's disease. Thus, Kv3.4 likely represents a novel therapeutic target for the disease. 相似文献

We cloned from a rat brain cDNA library a novel cDNA and named it a potential synaptic guanine nucleotide exchange factor (GEF) for Arf (synArfGEF (Po)) (GenBank Accession no. AB057643) based on its domain structure and localization. The cloned gene was 7410 bases long with a 3585-bp coding sequence encoding a protein of 1194 amino acids. The deduced protein contained a coiled-coil structure in the N-terminal portion followed by Sec7 and Plekstrin homology (PH) domains. Thus, the protein was a member of the Sec7 family of proteins, GEFs. Conservation of the ADP-ribosylation factor (Arf)-binding sequence suggested that the protein was a GEF for Arf. The gene was expressed specifically in the brain, where it exhibited region-specific expression. The protein was highly enriched in the postsynaptic density (PSD) fraction prepared from the rat forebrain. Uniquely, the protein interacted with PSD-95, SAP97 and Homer/Vesl 1/PSD-Zip45 via its C-terminal PDZ-binding motif and co-localized with these proteins in cultured cortical neurons. These results supported its localization in the PSD. The postsynaptic localization was also supported by immunohistochemical examination of the rat brain. The mRNA for the synArfGEF was also localized to dendrites, as well as somas, of neuronal cells. Thus, both the mRNA and the protein were localized in the postsynaptic compartments. These results suggest a postsynaptic role of synArfGEF in the brain. 相似文献

In an attempt to prepare a library of short oligoadenylate analogues featuring both the enzyme‐stable internucleotide linkage and the 5′‐O‐methylphosphonate moiety and thus obtain a pool of potential RNase L agonists/antagonists, we studied the spontaneous polycondensation of the adenosin‐5′‐O‐ylmethylphosphonic acid (pcA), an isopolar AMP analogue, and its imidazolide derivatives employing N,N′‐dicyclohexylcarbodiimide under nonaqueous conditions and uranyl ions under aqueous conditions, respectively. The RP LC–MS analyses of the reaction mixtures per se, and those obtained after the periodate treatment, along with analyses and separations by capillary zone electrophoresis, allowed us to characterize major linear and cyclic oligoadenylates obtained. The structure of selected compounds was supported, after their isolation, by NMR spectroscopy. Ab initio calculation of the model structures simulating the AMP‐imidazolide and pcA‐imidazolide offered the explanation why the latter compound exerted, in contrast to AMP‐imidazolide, a very low stability in aqueous solutions. © 2009 Wiley Periodicals, Inc. Using electrophysiological methods, effects of temperature on rhythmic impulsation of spontaneously active neurons of thoracic ganglia was studied in grasshoppers Tettigonia cantans and Metrioptera roeselii. Several types of response to temperature changes were observed. Its elevation caused an increase or decrease of the discharge dispersion and a gradual, proportional or abrupt rise of the impulse rate. On decreasing temperature, the impulsation usually returned to its original state, although sometimes at different rate. In neurons whose impulse rate was close to that of pulse repetition rate in male advertisement calls, this dependence was preserved at temperature changes. On the whole, there is reason to believe that changes of temperature in CNS are accompanied not only by a simple change of the neuronal activity level, but also by a complex readjustment in the neuronal network functioning, which allow preserving optimal level of insect metabolic processes and adequate behavior. 相似文献

Since chromium(III) was demonstrated to have antioxidative action, we have decided to study the effect of this element on V-induced LPO in liver and kidney of rats. Outbred 2-month-old, albino male Wistar rats received daily, for a period of 12 weeks: group I (control), deionized water to drink; group II, sodium metavanadate (SMV) solution at a concentration of 0.100mgV/mL; group III, chromium chloride (CC) solution at a concentration of 0.004mgCr/mL and group IV, SMV-CC solution at a concentration of 0.100mgV and 0.004mgCr/mL. The particular experimental groups took up with drinking water about 8.6mgV/kg b.w./24h (group II), 0.4mgCr/kg b.w./24h (group III), 9mgV and 0.36mgCr/kg b.w./24h (group IV). The V- or Cr-treated groups had higher concentrations of these two elements in liver and kidney compared to the controls. The administration of vanadium alone caused a significant decrease in fluid intake and in body weight gain compared to the controls. In liver supernatants obtained from all tested rats a statistically significant increase in MDA concentration was demonstrated in spontaneous LPO in comparison with the control rats. Moreover, in rats intoxicated with vanadium alone a statistically significant increase in liver MDA level was observed in the presence of 100microM NaVO(3). Instead, in supernatants of liver received from rats treated with chromium alone, a statistically significant increase in MDA concentration in comparison with the controls was found in the presence of 400microM NaVO(3). In kidney supernatants obtained from rats treated with chromium alone, a statistically significant increase in lipid peroxidation was shown in the presence of 30microM FeSO(4) and 400microM NaVO(3). These results show that the tested doses of vanadium(V) and chromium(III) ingested by rats with their drinking water caused significant alterations in internal organs, especially in liver. In this study, I analyzed the natural recovery of grasslands on abandoned agricultural fields in the Transylvanian Lowland (Câmpia Transilvaniei), Romania. I examined fields that were abandoned 1–40 years ago and considered how successful they have been in recovering spontaneously as compared to reference grassland, especially in relation to species composition and dominance structure. Diverse, nondegraded seminatural grasslands from the surroundings were chosen as targets for the recovery. Five such grasslands were analyzed in order to have multiple references that accounted for site heterogeneity and different land use history. This study found that the number of natural and seminatural habitat species increased, whereas the number of weeds and aliens decreased with age. Old-fields had become very similar in species composition and dominance structure to reference grasslands over a 14- to 20-year time interval, with the only failure being the unsuccessful or slow colonization of a few grassland species. Because spontaneous succession is efficient, human interventions are not needed to restore target communities on the old-fields. Propagule pressure expressed by the area of potential seed source in a 500-m-radius buffer was found to have a strong influence on recovery success of old-fields. The success of grassland species in colonizing postagricultural fields was not affected by their dispersal mode but by their frequency in the landscape, this being another evidence for the importance of propagule availability in the course of recovery. The contractile vacuole complex is a membrane-bound osmoregulatory organelle of fresh water protozoa such as Paramecium. In Paramecium it consists of a central vacuole (the contractile vacuole) and 5-10 arms that radially extend from the vacuole into the cytosol (the radial arms). Excess cytosolic water, acquired osmotically, is segregated by the radial arms and enters the vacuole, so that the vacuole swells (the fluid-filling phase). The vacuole then rounds (the rounding phase) and the radial arms sever from the vacuole. The vacuole membrane then fuses with the plasma membrane at the pore region and the pore opens. The vacuole shrinks as its fluid is discharged through the pore (the fluid-discharging phase). The pore closes when the fluid has been discharged. The radial arms then reattach to the vacuole, so that the vacuole swells again as the fluid enters from the arms (the next fluid-filling phase). We found that the vacuole continued to show rounding and slackening even after it together with a small amount of cytosol had been isolated from the cell. Using a microcantilever placed on the surface of the vacuole the tension of the in vitro vacuole increased to 5 x 10(-3)N m(-1) as the vacuole rounds, and its lowest value was 1 x 10(-4)N m(-1) during slackening. We propose a hypothesis that an increase in the spontaneous curvature of the organelle's membrane leads to an increase in membrane tension and thus to the vacuole's rounding, severing of the radial arms from the vacuole, and opening of the pore. Conversely, a decrease in the spontaneous curvature accompanied by a decrease in membrane tension could lead to the closing of the pore and reattachment of the radial arm at the start of the fluid-filling phase. 相似文献

A single autonomous video camera was used to record the abundances of Chromis viridis over a branching Acropora sp. colony eight times per day over a period of 50 days. The poor explanatory power of global radiation suggests the need for recording the light really available to the fish, especially in the UV range. The increasing number of C. viridis observed with increasing wind along shore and water level may correspond to individuals swimming further from their shelter in order to get closer to the food carried by the water currents. 相似文献
