X-linked adrenoleukodystrophy (ALD; MIM #300100) is a neurodegenerative disorder caused by mutations in the ABCD1 adrenoleukodystrophy protein gene. The ABCD1 gene mutations have been reported by laboratories in China and Japan, but not in Korea. This case report describes a Korean boy diagnosed with X-ALD. Direct sequencing for the ABCD1 gene in this boy and his mother detected Tyr620His missense mutation, caused by cDNA nucleotide change 1858 T>C in exon 8 (c.1858T>C). This missense variant was novel and predicted to be possibly damaging by the PolyPhen and SIFT prediction software. Moreover, this is the first report in Korean. 相似文献

目的 研究重庆地区成人感染性腹泻患者的病原学特点.方法 采用MICROSCAN系统进行粪便细菌培养,用ELISA和多重PCR方法进行病毒检测.结果 130例腹泻标本中,检出细菌阳性17例,包括沙门菌属7例,志贺菌属5例,副溶血弧菌3例和嗜水气单胞菌2例.病毒检测中,单重感染30例,双重感染10例,三重感染1例.A组轮状病毒检出率为3.85% (5/130),B轮状病毒检出率为3.85% (5/130),C组轮状病毒检出率为15.4% (20/130),诺如病毒检出率为9.23% (12/130),星状病毒检出率为4.62%(6/130),札如病毒检出率为3.08% (4/130),腺病毒检出率为0.77%(1/130).结论 重庆地区成人腹泻患者中,沙门菌和志贺菌为细菌感染的主要菌种,轮状病毒和诺如病毒为病毒感染的主要病原体. 相似文献

NELL2 is a neuron-specific secreted glycoprotein containing an N-terminal thrombospondin I-like domain (TSP-N). In this study, we describe NELL2-Tsp, a novel alternative splice variant of rat NELL2. NELL2-Tsp uses an alternate stop codon resulting in a C-terminal truncated form of NELL2, containing a signal peptide and a TSP-N domain. NELL2-Tsp is a glycosylated protein specifically expressed in brain tissue. NELL2-Tsp and NELL2 are secreted, likely due to the putative signal peptide. However, due to the truncation, the secreted portion of NELL2-Tsp is smaller than that of NELL2. Immunoprecipitation analysis confirmed that NELL2-Tsp was able to associate with NELL2 and with itself. In addition, expression of NELL2-Tsp notably reduced secretion of NELL2 and inhibited NELL2-mediated neurite outgrowth. These results suggest that NELL2-Tsp may act as a negative regulator of wild-type NELL2. 相似文献

A human nuclear actin-related protein, hArpNβ/Baf53, is a component of chromatin remodeling and histone acetyltransferase complexes. We identified two alternative splicing products of the gene for hArpNβ/Baf53. They encoded a protein isoform, hArpNβS; and its fusion product with green fluorescent protein was to be found in the cytoplasm, not the nucleus. The known or suspected hazards of polycyclic aromatic hydrocarbons (PAHs) have provoked enormous concentration and endeavours to relieve or eliminate these precarious compounds from miscellaneous environments including soil, water and air. Among various interventions, biodegradation is an appealing approach for its comparative high efficiency and preferable safety. Microorganisms played crucial role in biodegradation of PAHs. Traditional access mainly including culture-dependent procedures has discovered and isolated PAHs-degrading microorganisms which could be subsequently applied to specific contaminated locus. Although certain progress has been achieved owing to traditional methods, much details in PAHs bioremedation leave pending because of the complexity nature of this process. As the rapid development of biology, molecular techniques such as PCR, fingerprinting technique (mainly DGGE), DNA hybridization technique and gene reporters technique have been intensively applied to gain further insight into the mechanism of PAHs degradation. These techniques not only proved the existence and role of uncultivable microorganisms in the whole population of PAHs degrading related microbials, but also made it possible to revealed the otherwise undetectable complex relationships between multi-microorganism concerned in PAHs biodegradation. Even though the efficiency of the polymerase chain reaction (PCR) reaction decreases, analyses are made in terms of Galton-Watson processes, or simple deterministic models with constant replication probability (efficiency). Recently, Schnell and Mendoza have suggested that the form of the efficiency, can be derived from enzyme kinetics. This results in the sequence of molecules numbers forming a stochastic process with the properties of a branching process with population size dependence, which is supercritical, but has a mean reproduction number that approaches one. Such processes display ultimate linear growth, after an initial exponential phase, as is the case in PCR. It is also shown that the resulting stochastic process for a large Michaelis-Menten constant behaves like the deterministic sequence x(n) arising by iterations of the function f(x)=x+x/(1+x). 相似文献

随着合成生物学的兴起和发展,基因克隆和DNA大片段组装成为了常规操作。利用人工智能和液体操作机器人进行高通量的DNA组装和功能筛选已被广泛应用。传统的依赖于限制性内切酶识别位点的克隆技术对序列有选择性、步骤繁琐、实验人员的培训周期长,不利于以流水线形式进行工程化使用,已经逐步在生物工程领域内被淘汰。文中论述了一系列适于机械化操作的新一代分子克隆技术,即不依赖基因序列和连接反应克隆方法、Gibson组装、聚合酶环形延伸克隆、细胞裂解物体外无痕连接和细胞体内组装克隆。对这些方法的建立、基本原理及应用前景等方面进行了总结,并对其优缺点进行了比较。 相似文献
