斜纹夜蛾Spodoptera litura(Fabricius)是重要的农业害虫之一。为了控制斜纹夜蛾,使农作物免受危害,前人研究了化学防治、杀虫灯和性诱捕器诱杀、植物诱集和驱避、转基因抗虫品种以及生物防治对其种群控制作用。化学防治在压制斜纹夜蛾种群暴发、减轻农作物遭受损失方面作出了巨大贡献,但日益增强的抗药性、农药残留造成的环境污染、对天敌的杀伤和人畜安全的影响等问题不断受到关注,人们开始对该害虫的一些新的治理策略进行了探讨。在这些无公害的控制技术中,植物诱集和生物防治是2种值得提倡的有效措施。不少植物源提取物和昆虫生长调节剂对斜纹夜蛾幼虫具有极好的效果,是值得研发的2种新型生物制剂。香芋和蓖麻等诱集作物对斜纹夜蛾有较好的诱集效果,如果能将作物诱集和释放天敌昆虫或病原微生物制剂结合,使之成为一套有效的生态控制体系,将可现实斜纹夜蛾种群的持续控制。  相似文献

摘要 目的:探究外周血中性粒细胞胞外诱捕网(NETs)、TP53、信号转导与转录因子3(STAT3)表达与弥漫性大B细胞淋巴瘤(DLBCL)临床病理及预后的关系。方法:选取2020年3月-2021年12月收治的71例DLBCL患者作为研究对象,抽取患者外周静脉血,采用R-CHOP方案进行治疗,记录患者外周血NETs、TP53、STAT3表达情况并分析DLBCL患者外周血NETs、TP53、STAT3表达与其临床病理及预后的关系。结果:髓细胞组织增生蛋白(MYC)阳性在TP53阳性中的占比显著高于TP53阴性,差异有统计学意义(x2=28.844,P<0.001);Hans分型生发中心B细胞(GCB)在STAT3阳性中的占比显著高于STAT3阴性(x2=4.331,P=0.037),其余差异无统计学意义(P>0.05);随访截止至2022年6月,随访时长8~28个月,71例患者中共53例缓解DLBCL患者,其余18例为R/R DLBCL患者;NETs阳性、TP53阳性、STAT3阳性患者无进展生存期(PFS)显著低于NETs阴性、TP53阴性、STAT3阴性患者,差异有统计学意义(P<0.05)且NETs阳性、TP53阳性、STAT3阳性患者存活率均低于NETs阴性、TP53阴性、STAT3阴性患者(P<0.05);单因素分析结果显示Ann Arbor分期、NETs、TP53、STAT3为DLBCL患者的影响因素(P<0.05);以患者预后情况(R/R DLBCL=1,缓解DLBCL=0)为因变量,将Ann Arbor分期、NETs、TP53、STAT3单因素分析有统计学意义的因素纳入COX回归模型中,结果显示:NETs、TP53、STAT3为DLBCL患者预后的危险因素(P<0.05)。结论:TP53、STAT3表达与DLBCL临床病理存在一定相关性,临床应对DLBCL患者TP53、STAT3表达情况引起重视;NETs、TP53、STAT3表达为DLBCL预后的危险因素,可作为DLBCL患者不良预后的预测指标。  相似文献

Pitfall trapping is a sampling technique extensively used to sample surface foraging invertebrates for biological diversity studies and ecological monitoring. To date, very few invertebrate studies have considered what trap size is optimal for sampling spiders. This study presents preliminary findings from a single short sampling period on the role of trap size in sampling spiders in a Western Australian Jarrah forest. Four different trap diameters (4.3, 7.0, 11.1 and 17.4 cm) were examined (4 trap sizes × 15 replicates = 60 traps). Two-way ANOVAs revealed no significant interaction effects between trap size or the spatial positioning of transects within the study site along which the pitfall traps were arranged. Post-hoc tests revealed abundance, family richness and species richness increased with increasing trap sizes for traps 7.0 cm. No significant differences in these dependent variables occurred between 4.3 and 7.0 cm traps, or for species richness between 11.1 and 17.4 cm traps. Determination of an optimal trap size was undertaken by bootstrapping and calculating species accumulation curves for increasing numbers of traps used. Three different criteria were considered: equivalent number of traps (15), standardized sampling intensity (cumulative trap circumference, approximately 207 cm) and standardized cumulative handling time (approximately 1 hour 17 minutes). The largest trap size (17.4 cm) was most efficient in terms of number of traps and trap circumference. For the same number of traps, it caught 19 species whereas all other trap sizes caught ten species. At the standardized circumference, it caught seven species whereas all other trap sizes caught five. For handling time, however, the two largest trap sizes (17.4 and 11.1 cm) were optimal. The quality of molecular dynamics (MD) simulations of proteins depends critically on the biomolecular force field that is used. Such force fields are defined by force-field parameter sets, which are generally determined and improved through calibration of properties of small molecules against experimental or theoretical data. By application to large molecules such as proteins, a new force-field parameter set can be validated. We report two 3.5 ns molecular dynamics simulations of hen egg white lysozyme in water applying the widely used GROMOS force-field parameter set 43A1 and a new set 45A3. The two MD ensembles are evaluated against NMR spectroscopic data NOE atom–atom distance bounds, 3JNH and 3J coupling constants, and 15N relaxation data. It is shown that the two sets reproduce structural properties about equally well. The 45A3 ensemble fulfills the atom–atom distance bounds derived from NMR spectroscopy slightly less well than the 43A1 ensemble, with most of the NOE distance violations in both ensembles involving residues located in loops or flexible regions of the protein. Convergence patterns are very similar in both simulations atom-positional root-mean-square differences (RMSD) with respect to the X-ray and NMR model structures and NOE inter-proton distances converge within 1.0–1.5 ns while backbone 3JHN-coupling constants and 1H– 15N order parameters take slightly longer, 1.0–2.0 ns. As expected, side-chain 3J-coupling constants and 1H– 15N order parameters do not reach full convergence for all residues in the time period simulated. This is particularly noticeable for side chains which display rare structural transitions. When comparing each simulation trajectory with an older and a newer set of experimental NOE data on lysozyme, it is found that the newer, larger, set of experimental data agrees as well with each of the simulations. Two dimensional (2D) solid-state (13)C.(13)C dipolar recoupling experiments are performed on a series of model compounds and on the visual pigment rhodopsin to establish the most effective method for long range distance measurements in reconstituted membrane proteins. The effects of uniform labeling, inhomogeneous B(1) fields, relaxation and dipolar truncation on cross peak intensity are investigated through NMR measurements of simple amino acid and peptide model compounds. We first show that dipolar assisted rotational resonance (DARR) is more effective than RFDR in recoupling long-range dipolar interactions in these model systems. We then use DARR to establish (13)C-(13)C correlations in rhodopsin. In rhodopsin containing 4'-(13)C-Tyr and 8,19-(13)C retinal, we observe two distinct tyrosine-to-retinal correlations in the DARR spectrum. The most intense cross peak arises from a correlation between Tyr268 and the retinal 19-(13)CH(3), which are 4.8 A apart in the rhodopsin crystal structure. A second cross peak arises from a correlation between Tyr191 and the retinal 19-(13)CH(3), which are 5.5 A apart in the crystal structure. These data demonstrate that long range (13)C em leader (13)C correlations can be obtained in non-crystalline integral membrane proteins reconstituted into lipid membranes containing less than 150 nmoles of protein. In rhodopsin containing 2-(13)C Gly121 and U-(13)C Trp265, we do not observe a Trp-Gly cross peak in the DARR spectrum despite their close proximity (3.6 A) in the crystal structure. Based on model compounds, the absence of a (13)C em leader (13)C cross peak is due to loss of intensity in the diagonal Trp resonances rather than to dipolar truncation.  相似文献

Relaxation in methyl groups is strongly influenced by cross-correlated interactions involving the methyl dipoles. One of the major interference effects results from intra-methyl (1)H-(13)C, (1)H-(1)H dipolar interactions, leading to significant differences in the relaxation of certain multiplet components that contribute to double- and zero-quantum (1)H-(13)C spectra. NMR experiments are presented for the measurement of this differential relaxation effect. It is shown that this difference in relaxation between double- and zero-quantum multiplet components can be used as a sensitive reporter of side chain dynamics and that accurate methyl axis order parameters can be measured in proteins that tumble with correlation times greater than approximately 5 ns.  相似文献

It is believed that reactive oxygen species (ROS) play a very important role in the pathogenesis of acute respiratory distress syndrome (ARDS), but the mechanism has not been so clear, owing to the absence of direct measurable (experimental) data. In majority of the medical studies on free radicals, the analysis of ROS has generally been done by the way of measuring their secondary and breakdown products. In our study, we used electron spin resonance (ESR), a sensitive and accurate technique to detect ROS directly and also used some other sensitive techniques including ultra-weak luminescence and chemical luminescence to identify the species and relative amount of ROS. Furthermore, superoxide dismutase (SOD) was pre-administrated in ARDS rats to verify the results from the above studies and explore the possibility of the clinical application of SOD in ARDS. The spectra of ESR showed that the concentration of ROS increased at 10 min and reached a summit at 30 min after injection of oleic acid (OA), then dropped gradually. The scavenging effects of different scavenging agents on ROS by the analysis of ultra-weak luminescence proved that superoxide anion was the main species of ROS in the development of OA-induced ARDS. Moreover, the results of quantified measure of superoxide anion by chemical luminescence also showed the accordant tendency exhibited in ESR measurement. The pre-treatment of SOD might distinctly inhibit the production of superoxide anion, obviously improve the blood gas status, lung wet/dry ratio and lung/body ratio in ARDS rats. It is suggested that ROS may play a key role in the initiation phase of ARDS, while superoxide anion may be a leading actor in this process and SOD could effectively protect rats from ARDS. These results may provide helpful information for the treatment and prevention of ARDS.  相似文献

Photosensitizers newly developed for photodynamic therapy of cancer need to be assessed using accurate methods of measuring reactive oxygen species (ROS). Little is known about the characteristics of the reaction of singlet oxygen (1O2) with spin traps, although this knowledge is necessary in electron spin resonance (ESR)/spin trapping. In the present study, we examined the effect of various reductants usually present in biological samples on the reaction of 1O2 with 5,5-dimethyl-1-pyrroline-N-oxide (DMPO). The ESR signal of the hydroxyl radical (•OH) adduct of DMPO (DMPO-OH) resulting from 1O2-dependent generation of •OH strengthened remarkably in the presence of reduced glutathione (GSH), 6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid (Trolox), ascorbic acid, NADPH, etc. A similar increase was observed in the photosensitization of uroporphyrin (UP), rose bengal (RB) or methylene blue (MB). Use of 5-(diethoxyphosphoryl)-5-methyl-1-pyrroline-N-oxide (DEPMPO) as a spin trap significantly lessened the production of its •OH adduct (DEPMPO-OH) in the presence of the reductants. The addition of DMPO to the DEPMPO-spin trapping system remarkably increased the signal intensity of DEPMPO-OH. DMPO-mediated generation of •OH was also confirmed utilizing the hydroxylation of salicylic acid (SA). These results suggest that biological reductants enhance the ESR signal of DMPO-OH produced by DMPO-mediated generation of •OH from 1O2, and that spin trap-mediated •OH generation hardly occurs with DEPMPO.  相似文献

We conducted field and semi-field experiments to determine the most visually stimulating and acceptable version of a branch-mimicking trap (PVC cylinder topped with an inverted screen funnel) to capture adult plum curculios, Conotrachelus nenuphar (Herbst) (Coleoptera: Curculionidae) foraging within canopies of host trees. Cylinder traps (25 cm tall × 6 cm diam) coated with different colors of gloss or flat latex paint captured similar numbers of feral adults in field tests. Tall cylinder traps (50 cm tall × 6 cm diam) captured significantly more feral adults in field tests in 1999 than cylinder traps of lesser height and/or different diameter, and significantly more released adults arrived at traps with these dimensions compared to cylinder traps of lesser height in semi-field tests. In field and semi-field tests, respectively, significantly more adults were captured by and numerically more adults arrived at tall cylinder traps (50 cm tall × 6 cm diam) attached in an upright, standard position to a horizontal limb than attached at angles of 45°, 90°, 135°, or 180° to horizontal limbs.  相似文献

The Lucitrap (Miazma Pty Ltd, Queensland, Australia) combined with a synthetic odour bait, Lucilure (Miazma Pty Ltd, Queensland, Australia), is a commercially available trap for sampling and control of Lucilia cuprina (Wiedemann) in Australia. It was tested in Hungary against Lucilia sericata (Meigen) (Diptera: Calliphoridae), a cause of sheep strike throughout temperate Europe. The standard Lucitrap was tested against black or yellow sticky target traps. Both trap types were baited with either Lucilure or liver and 10% w/v sodium sulphide solution. With Lucilure as bait, L. sericata were caught on sticky traps but not in Lucitraps. With liver and sodium sulphide as bait, sticky traps caught 500-1500 times more L. sericata than Lucitraps. An adhesive sheet fitted to the top of a Lucitrap captured 30-300 times more L. sericata then were captured inside an unaltered Lucitrap. Direct observation of metallic green calliphorids (92.1% L. sericata) alighting on Lucitraps indicated that most flies stayed for a short while (modal class 2-4 s) and only a few stayed longer, to an observed maximum of 28 s. Flies explored a mean of 1.5 entry holes (range 0-7) during a visit but only 6% entered the trap. Size of L. sericata was not a physical barrier to Lucitrap entry, because many larger species were captured. However, L. sericata captured inside Lucitraps were significantly smaller than those captured on sticky traps, demonstrating that size was of behavioural importance. The data demonstrate that the Lucitrap is not effective as a trap for L. sericata in Hungary, due mainly to a failure of flies to enter the trap in large numbers. In Australia and South Africa, L. sericata is commonly caught in Lucitraps baited with Lucilure, although L. cuprina is more numerous. Our study highlights the potential for diversity of fly behaviour between different geographical populations of the same species. Such diversity can have a significant effect on the functioning of systems for fly sampling and control, when these systems depend for their success on certain behavioural responses of the target species.  相似文献
