Although fungi have contributed tremendously to understanding biological phenomena common to all eukaryotic organisms, some of their properties testify as to their uniqueness. Among these are growth by apical extension of hyphae, the manufacture of hydrophobins for emergence into the air, and the possession of an extended somatic heterokaryon in basidiomycetes. This justifies studies on the molecular basis of development aimed particularly at this group of organisms, which are of great importance to life on earth and human society.  相似文献

Sugar analysis of isolated cell walls from three formae speciales of Fusarium oxysporum showed that they contained not only glucose and (N-acetyl)-glucosamine, but also mannose, galactose, and uronic acids, presumably originating from cell wall glycoproteins. Cell wall glycoproteins accounted for 50–60% of the total mass of the wall. X-ray diffraction studies showed the presence of α-1,3-glucan in the alkali-soluble cell wall fraction and of β-1,3-glucan and chitin in the alkali-insoluble fraction. Electron microscopy and lectin binding studies indicated that glycoproteins form an external layer covering an inner layer composed of chitin and glucan.  相似文献

Females of all species belonging to the family Drosophilidae have two kinds of sperm-storage organs: paired spherical spermathecae and a single elongate tubular seminal receptacle. We examined 113 species belonging to the genus Drosophila and closely allied genera and describe variation in female sperm-storage organ use and morphology. The macroevolutionary pattern of organ dysfunction and morphological divergence suggests that ancestrally both kinds of organs stored sperm. Loss of use of the spermathecae has evolved at least 13 times; evolutionary regain of spermathecal function has rarely if ever occurred. Loss of use of the seminal receptacle has likely occurred only once; in this case, all descendant species possess unusually elaborate spermathecae. Data further indicate that the seminal receptacle is the primary sperm-storage organ in Drosophila. This organ exhibits a pattern of strong correlated evolution with the length of sperm. The evolution of multiple kinds of female sperm-storage organs and the rapidly divergent and correlated evolution of sperm and female reproductive tract morphology are discussed.  相似文献

Pollen samples of 6 varieties of Zea mays L. were used to isolate the viable sperm cells. After being probed with N-hydroxysuccinimido-biotin (NHS-biotin), the sperm cell plasma membrane proteins were compared with each other using the method of Western blotting. Results showed that there was no significant difference among varieties. The molecular weights of probed plasma membrane proteins were concentrated on 91,60,43,30 and 17 kD. Immunochemical method was adopted for further purification of sperm plasma membrane protein preparation which was some- what contaminated with cell organelles. After the cell organelles were isolated from etiolated seedlings of Zea mays by sucrose density gradient super centrifugation, the crude membrane proteins of organelles, endoplasm reticulum, mitochondria, Golgi body and plasmolemma were respectively used as antigen to immunize Guinea pig. The antibody was obtained from respective antiserum, then further used to produce immuno-affinity absorbent. After the solution of membrane proteins of sperm cells passed through the column, some proteins probed whth NHS-biotin were identified. Two major proteins probed with NHS-biotin were considered to be sperm cell specific. The size of these proteins in SDS-PAGE was about 65 kD, 22 kD, respectively.  相似文献

The techniques of transmission electron microscopy (TEM), electron microscopy (EM) cytochemical visualization of polysaccharide, cell wall flourescence labelling of cell wall and inhibition of wall formation by coumarin treatment were used to explore the cell wall regeneration and its chemical characteristics in mesophyll protoplasts of Astragalus melilotoides var. tenuis. The results showed that after 24 h in culture a number of protruding vesicles, as well as a small amount of fibrillar component were formed on the surface of protoplasts. On day 3, the amount of fibrils increased significantly. On day 5, regenerated primary wall composed of fibrils and granules were observed, in which polysacchaides were detected as result of the periodic acid-silver methenamine reaction. In addition, after 36 h in culture, the protoplasts tended to coalesce, flourescence staining and coumarin treatment demonstrated that the protoplast adhesion was the result of cell wall formation. Based on these data, problems such as the structure of regenerated wall and its chemical nature, etc. were discussed.  相似文献

In a previous report we have shown that the arrangement of callus cell wail fibrils of Abutilon avicennae could be induced to change under IAA (2 ppm) and kinetin (10 ppm) treatments. Kinetin at this concentration was shown to be able to induce callus cell differentiation and form tracheary elements by changing the orientation of the wall fibrils. It was thus assumed that the hormonal induction of cellular differentiation and structual change of the cell wall may possibly be accompanied by the simultaneous changes of chemical composition of the wall. Attempt was therefore made to investigate if such changes do occur in vitro under the influence of phytohormones. Suspension cell-culture of Abutilon avicennae was used in this experiment to study the hormonal effect on the incorporation of H3-glucose into the cell wall polysaccharides. Analysis of neutral sugars of the cell wall following IAA (2ppm) and kinetin (10ppm) treatments was carried out with a gas chromatography. The results obtained in this experiment are shown in tables 1-2 and figures 1, It was found that the auxin was capable of promoting the synthesis of all neutral sugars, among which the glucose and the maunose in particular, increased tremendously. When H3-glucose was added to the culture medium, IAA was found to enhance the incorporation of the isotopes into the matrix polysaccharides (hemiceUulose and pectin). The result demonstrates clearly that the primary function of IAA is to stimulate the synthesis of hemicellulose composition and, as a consequence, the cell wall retained at the primary growth stage. Kinetin, on the other hand, showed an inhibitory effect on most of the neutral sugars except glucose and mannose. It appeared to have a striking inhibitory action on the synthesis of arabinose and rhanmose (a special composition of pectic substance). It also limited the incorporation of H3-glucose into the pectic substance. It is, therefore, suggested that the action of kinetin may mainly be inhibitory on the synthesis of pectic composition. The decreased rate of pectin synthesis would implicate that the cell wall has been advan ced into the phase of secondary growth. The results presented here agree fairly well with our connotation that there is a parallel relationship between cellular morphology and biochemical characteristics during cell wall differentiation and growth.  相似文献

The enzyme horseradish peroxidase (EC 1.11.1.7) catalyses oxidation of NADH. NADH oxidation is prevented by addition of the enzyme superoxide dismutase (EC 1.15.1.1) to the reaction mixture before adding peroxidase but addition of dismutase after peroxidase has little inhibitory effect. Catalase (EC 1.11.1.6) inhibits peroxidase-catalysed NADH oxidation when added at any time during the reaction. Apparently the peroxidase uses hydrogen peroxide (H2O2) generated by non-enzymic breakdown of NADH to catalyse oxidation of NADH to a free-radical, NAD., which reduces oxygen to the superoxide free-radical ion, O2 .-. Some of the O2 .- reacts with peroxidase to give peroxidase compound III, which is catalytically inactive in NADH oxidation. The remaining O2 .- undergoes dismutation to O2 and H2O2. O2 .- does not react with NADH at significant rates. Mn2+ or lactate dehydrogenase stimulate NADH oxidation by peroxidase because they mediate a reaction between O2 .- and NADH. 2,4-Dichlorophenol, p-cresol and 4-hydroxycinnamic acid stimulate NADH oxidation by peroxidase, probably by breaking down compound III and so increasing the amount of active peroxidase in the reaction mixture. Oxidation in the presence of these phenols is greatly increased by adding H2O2. The rate of NADH oxidation by peroxidase is greatest in the presence of both Mn2+ and those phenols which interact with compound III. Both O2 .- and H2O2 are involved in this oxidation, which plays an important role in lignin synthesis.  相似文献

Melanocarpus albomyces, a thermophilic fungus isolated from compost by enrichment culture in a liquid medium containing sugarcane bagasse, produced cellulase-free xylanase in culture medium. The fungus was unusual in that xylanase activity was inducible not only by hemicellulosic material but also by the monomeric pentosan unit of xylan but not by glucose. Concentration of bagasse-grown culture filtrate protein followed by size-exclusion and anion-exchange chromatography separated four xylanase activities. Under identical conditions of protein purification, xylanase I was absent in the xylose-grown culture filtrate. Two xylanase activities, a minor xylanase IA and a major xylanase IIIA, were purified to apparent homogeneity from bagasse-grown cultures. Both xylanases were specific forβ-1,4 xylose-rich polymer, optimally active, respectively, at pH 6.6 and 5.6, and at 65°C. The xylanases were stable between pH 5 to 10 at 50°C for 24 h. Xylanases released xylobiose, xylotriose and higher oligomers from xylans from different sources. Xylanase IA had a Mr of 38 kDa and contained 7% carbohydrate whereas xylanase IIIA had a Mr of 24 kDa and no detectable carbohydrate. The Km for larchwood xylan (mg ml−1) and Vmax (μmol xylose min−1 mg−1 protein) of xylanase IA were 0.33 and 311, and of xylanase IIIA 1.69 and 500, respectively. Xylanases IA, II and IIIA showed no synergism in the hydrolysis of larchwood glucuronoxylan or oat spelt and sugarcane bagasse arabinoxylans. They had different reactivity on untreated and delignified bagasse. The xylanases were more reactive than cellulase on delignified bagasse. Simultaneous treatment of delignified bagasse by xylanase and cellulase released more sugar than individual enzyme treatments. By contrast, the primary cell walls of a plant, particularly from the region of elongation, were more susceptible to the action of cellulase than xylanase. The effects of xylanase and cellulase on plant cell walls were consistent with the view that hemicellulose surrounds cellulose in plant cell walls.  相似文献

1. To examine the relationship between male–female emergence patterns and ejaculate dynamics, patterns producing sperm and seminal fluids in male internal reproductive organs, the size of a spermatophore transferred at mating and the fate of spermatophore contents moved into female storage organs were compared among the fishflies (Megaloptera: Corydalidae: Chauliodinae), Parachauliodes continentalis, P. japonicus and Neochauliodes sinensis .
2. Spermatophore contents moved into female storage organs decreased rapidly in P. japonicus and N. sinensis , but hardly at all in P. continentalis . This suggests that the females of the former two species may remate sooner than the latter species as it is known in insects that material remaining in the storage organs mechanically inhibits receptivity to mating.
3. Male P. japonicus and N. sinensis increased in internal reproductive organ mass continuously after adult eclosion, and the spermatophore size produced at the first mating increased with male age. In contrast, the internal reproductive organs of P. continentalis were relatively small and did not increase in mass after emergence. P. continentalis transferred a constantly smaller spermatophore at any copulation than the former two species.
4. Males of P. japonicus and N. sinensis emerged earlier than females, while P. continentalis showed a nearly simultaneous emergence pattern between the sexes. It seems that males of P. japonicus and N. sinensis (more polyandrous than P. continentalis ) inhibit female receptivity for a longer time period by emerging earlier and transferring larger ejaculates. In these two species, the number of sperm ejaculated at the first mating also increased with male age. The increase in sperm number by emerging earlier may be adaptive for the males owing to numerical sperm competition when the female remates.  相似文献
