以玉米淀粉糖渣为原料制备米曲,米曲中的中性蛋白酶活力能够达到4000 U/g(干基),糖化酶比活力可达到120 U/g左右,满足酿造酱油所需要求。用糖渣米曲发酵酱油,氨基态氮和总氮质量浓度分别能够达到7.1 g/L和11.9 g/L,还原糖质量浓度为13.2 g/L,各项理化指标均可达到国家二级酱油标准。  相似文献

本实验以灵芝、金耳以及冬虫夏草作为发酵菌种,选用豆浆为基本原料配制培养基,在发酵过程中定时取样并测定其中几种生物活性物质的含量,分析各生物活性物质在发酵过程中的变化。研究结果表明,三种食用真菌发酵豆浆的发酵液中总游离型大豆异黄酮含量在发酵过程中均缓慢上升并分别在发酵72 h、24 h和36 h达到最高,与发酵初期相比分别增加了31.81μg/m L、19.05μg/m L和15.71μg/m L;三种食用真菌发酵液中蛋白质含量随着发酵时间的推移而降低的同时氨基酸含量均有增加,且发酵液中的肽主要是分子量范围在300 Da~1 500 Da之间的具有较高生物活性的小分子肽,发酵液中的必需氨基酸含量以及占总氨基酸含量的比值均有增加。本实验探究了食用真菌发酵豆浆过程中生物活性物质的变化,为日后的富集游离型大豆异黄酮、多肽以及氨基酸等生物活性物质实验提供依据,并为工业化生产提供实践基础。  相似文献

【目的】对酱油酱醪来源的嗜盐四联球菌进行分类,并比较菌株的生理特性。【方法】采用多位点序列分型和糖类代谢聚类分析相结合的方法对菌株进行分类,通过菌株对环境耐受性分析研究菌株的生理特性。【结果】酱醪来源的12株嗜盐四联球菌被分为两个类群,菌株所属类别与其分离的原生环境存在一定的关联性。嗜盐四联球菌C25、C33、C3、R55耐高盐及高糖能力均较强,菌株C1、R44和R23耐高盐能力较强。虽然嗜盐四联球菌普遍不耐酸,但是菌株C33和R44在酸性培养体系(pH 5.0)中仍能正常生长。较低的培养温度(15℃和25℃)对嗜盐四联球菌的生长有显著抑制。【结论】采用基因型与表型结合的方式成功将酱醪来源的嗜盐四联球菌归为2个类群,不同类群的菌株在耐高渗等生理特性上具有差异性。  相似文献

PEGylation is frequently used to improve the efficacy of protein and peptide drugs. Recently, we investigated its effects on the action mechanism of the cyclic β-sheet antimicrobial peptide tachyplesin I isolated from Tachypleus tridentatus [Y. Imura, M. Nishida, Y. Ogawa, Y. Takakura, K. Matsuzaki, Action Mechanism of Tachyplesin I and Effects of PEGylation, Biochim. Biophys. Acta 1768 (2007) 1160-1169]. PEGylation did not change the basic mechanism behind the membrane-permeabilizing effect of the peptide on liposomes, however, it decreased the antimicrobial activity and cytotoxicity. To obtain further information on the effects of PEGylation on the activities of antimicrobial peptides, we designed another structurally different PEGylated antimicrobial peptide (PEG-F5W, E19Q-magainin 2-amide) based on the α-helical peptide magainin 2 isolated from the African clawed frog Xenopus laevis. The PEGylated peptide induced the leakage of calcein from egg yolk l-α-phosphatidylglycerol/egg yolk l-α-phosphatidylcholine large unilamellar vesicles, however, the activity was weaker than that of the control peptides. The PEGylated peptide induced lipid flip-flop coupled to the leakage and was translocated into the inner leaflet of the bilayer, indicating that PEGylation did not alter the basic mechanism of membrane permeabilization of the parent peptide. The cytotoxicity of the non-PEGylated peptides was nullified by PEGylation. At the same time, the antimicrobial activity was weakened only by 4 fold. The effects of PEGylation on the activity of magainin were compared with those for tachyplesin.  相似文献

Phosphoglucose isomerase (PGI) with a novel lysyl aminopeptidase (LysAP) activity was recently isolated and partially characterized from the human pathogen, Vibrio vulnificus. This PGI is a heterodimer consisting of 60.8- and 23.4-kDa subunits, which together provide LysAP activity. The present study further characterizes the complex structure and functions of Vibrio PGI and draws parallels with rabbit and human PGI. A Proscan search of Vibrio PGI revealed 194 different structural motifs of which 124 and 127 were also found in rabbit and human PGI, respectively. Vibrio PGI contains motifs for the serine, histidine and aspartic acid active sites of the subtilase family of serine proteases which form a putative catalytic triad consisting of His534 and Ser159 on the 60.8-kDa subunit and Asp53 on the 23.4-kDa subunit. Together, they form one LysAP site for each heterodimer. Each active site motif is overlapped by motifs for EF-hand calcium binding domains. The LysAP activity was inhibited by the addition of ≥10 μM Ca2+, suggesting that the EF-hand calcium-binding domain may be a natural regulatory region for LysAP activity. In contrast, PGI's isomerase activity was enhanced at Ca2+ concentrations >100 μM. PGI-LysAP cleaved the amino-terminal lysyl residue from des-Arg10-kallidin producing des-Arg9-bradykinin; therefore, Vibrio PGI-LysAP may serve as a virulence factor to enhance Vibrio invasiveness. Together, these data provide a framework to account for PGI's LysAP activity and further demonstrate the structural complexity and functional importance of this molecule.  相似文献

Stingless honey bees form a large group of bees that lack of a sting and are found among Meliponinae species indigenous to various tropical and subtropical regions. They are able to produce “stingless bee honey” that contains divergent categories of phenolic and flavonoid compounds and have been associated with antioxidant and antibacterial activity. This study examines the physicochemical properties, antioxidant-activity and anti-microbial activity of stingless bee honey from Malaysia that was produced by Geniotrigona thoracica, Heterotrigona itama and Heterotrigona erythrogastra. The results show that G. thoracica honey has the highest concentration of the total phenolic context (99.04?±?5.14?mg/ml) and the greatest reducing power (19.05?±?0.79%), while flavonoids (17.67?±?0.75?mg/ml), reducing power (18.10?±?0.35%), DPPH (47.40?±?3.18%) and FRAP (50.66?±?5.77?mM of Fe2+/100?g) of H. itama honey is significantly higher than those of the other honeys. In addition, G. thoracica honey has the highest antibacterial activity against Staphylococcus xylosus (2.10?±?0.10?cm), which is Gram-positive bacterium, and against Pseudomonas aeruginosa (1.60?±?0.10?cm) and Vibrio parahaemolyticus (2.03?±?0.06?cm), which are Gram-negative bacteria. These results suggest that stingless bee honeys possess useful amounts of phenolic and flavonoid compounds that are able to act as natural anti-oxidants and also have significant anti-microbial activity.  相似文献

This paper promotes the biometric classification system of plant cultivars, unique characteristics, in terms of the uranium (U) uptake, primarily in the function of the application for phytoremediation. It is known that the degree of adoption of U depends on the plant species and its morphological and physiological properties, but it is less known what impact have plants cultivars, sorts, and hybrids. Therefore, we investigated the U adoption in four cultivars of three plant species (corn, sunflower and soy bean). “Vegetation experiments were carried out in a plastic-house filled with soil (0.66 mgU) and with tailing (15.3 mgU kg?1) from closed uranium mine Gabrovnica-Kalna southeast of Serbia”. Principal Component Analysis (PCA), Cluster Analysis (CA) and analysis of variance (ANOVA) were used for assessing the effect of different substrates cultivars, plant species and plant organs (root or shoot) on U uptake. Obtained results showed that a difference in U uptake by three investigated plant species depends not only of the type of substrate types and plant organs but also of their cultivars. Biometrics techniques provide a good opportunity for a better understanding the behavior of plants and obtaining much more useful information from the original data.  相似文献

The role of the sesquiterpene botrydial in the interaction of the phytopathogenic fungus Botrytis cinerea and plant-associated bacteria was analyzed. From a collection of soil and phyllospheric bacteria, nine strains sensitive to growth-inhibition by B. cinerea were identified. B. cinerea mutants unable to produce botrydial caused no bacterial inhibition, thus demonstrating the inhibitory role of botrydial. A taxonomic analysis showed that these bacteria corresponded to different Bacillus species (six strains), Pseudomonas yamanorum (two strains) and Erwinia aphidicola (one strain). Inoculation of WT and botrydial non-producing mutants of B. cinerea along with Bacillus amyloliquefaciens strain MEP218 in soil demonstrated that both microorganisms exert reciprocal inhibitory effects; the inhibition caused by B. cinerea being dependent on botrydial production. Moreover, botrydial production was modulated by the presence of B. amyloliquefaciens MEP218 in confrontation assays in vitro. Purified botrydial in turn, inhibited growth of Bacillus strains in vitro and cyclic lipopeptide (surfactin) production by B. amyloliquefaciens MEP218. As a whole, results demonstrate that botrydial confers B. cinerea the ability to inhibit potential biocontrol bacteria of the genus Bacillus. We propose that resistance to botrydial could be used as an additional criterion for the selection of biocontrol agents of plant diseases caused by B. cinerea.  相似文献
