The proper organization of cortical microtubule arrays is essential for anisotropic growth in plants but how distinct array patterns are formed is not understood. Here, we report a relationship between microtubule dynamics and array organization using transgenic plants expressing modified tubulins. When green fluorescent protein (GFP) or a hemaglutinin epitope tag was fused to the N-terminus of tubulins and expressed in Arabidopsis plants, these tubulins were incorporated into microtubules along with endogenous tubulins. Plants expressing the modified beta-tubulins were phenotypically normal and possessed transversely oriented cortical arrays in the epidermal cells of the root elongation zone; however, the expression of modified alpha-tubulins caused right-handed helical growth, increased trichome branching, and a shallow left-handed (S-form) helical array organization. In cells expressing the modified alpha-tubulins, microtubule dynamicity was suppressed and polymerization was promoted, and GFP-EB1 (End Binding 1) labeled larger regions of the microtubule end more frequently, when compared with control cells. We propose that the N-terminal appendage introduced into alpha-tubulin inhibits GTP hydrolysis, thus producing polymerization-prone microtubules with an extended GTP cap. Consistent with this interpretation, plants expressing an alpha-tubulin mutated in the GTPase-activating domain exhibited similar microtubule properties, with regard to dynamics and the localization of GFP-EB1, and showed right-handed helical growth.  相似文献

Three new species of BrazilianDendropanax from the rain forests of Bahia state are described and illustrated.Dendropanax amorimii, endemic to the rain forests near Boa Nova, in southern Bahia, is characterized by compound umbellate inflorescences with elongated primary branches each having a proximal whorl of bracts, and by leaves with well-developed tertiary venation.Dendropanax bahiensis, endemic to the Atlantic rain forests of southern Bahia, is distinguished by its large and membranous leaves with reddish schizogenous glands on the abaxial surface and by the reduced and branched inflorescences with short-pedicellate flowers and large fruits.Dendropanax geniculatus, an endemic species from the “matas de grotão” of Serra da Chapadinha, Chapada Diamantina, is characterized by simple and usually geniculate inflorescences bearing long-pedicellate flowers and fruits.  相似文献

Christmas Island is a remote Australian territory located close to the main Indonesian island of Java. Y-chromosome and mitochondrial DNA (mtDNA) markers were used to investigate the genetic structure of the population, which comprises communities of mixed ethnic origin. Analysis of 12 Y-chromosome biallelic polymorphisms revealed a high level of gene diversity and haplotype frequencies that were consistent with source populations in southern China and Southeast Asia. mtDNA hypervariable segment I (HVS-I) sequences displayed high levels of haplotype diversity and nucleotide diversity that were comparable to various Asian populations. Genetic distances revealed extremely low mtDNA differentiation among Christmas Islanders and Asian populations. This was supported by the relatively high proportion of sequence types shared among these populations. The most common mtDNA haplogroups were M* and B, followed by D and F, which are prevalent in East/Southeast Asia. Christmas Islanders of European descent were characterized by the Eurasian haplogroup R*, and a limited degree of admixture was observed. In general, analysis of the genetic data indicated population affinities to southern Chinese (in particular from the Yunnan Province) and Southeast Asia (Thailand, Malaysia, and Cambodia), which was consistent with historical records of settlement. The combined use of these different marker systems provides a useful and appropriate model for the study of contemporary populations derived from different ethnic origins.  相似文献

The southern root-knot nematode (Meloidogyne incognita) is a major parasite of cotton in the U.S., and management tactics for this nematode attempt to minimize population levels. We compared three post-harvest practices for their ability to reduce nematode population levels in the field, thereby reducing initial nematode population for the next year's crop. The three practices tested were: 1) chemical defoliation before harvest plus cutting cotton stalks after harvest, 2) chemical defoliation plus applying a herbicide to kill plants prior to cutting the stalks, and 3) chemical defoliation without cutting stalks. Experiments were conducted in both the greenhouse and in the field. The greenhouse experiments demonstrated that M. incognita reproduction (measured as egg counts and root gall rating indices) was significantly greater when stalks were not cut. Cutting stalks plus applying herbicide to kill cotton roots did not significantly reduce nematode reproduction compared to cutting stalks alone. In field experiments, cutting stalks reduced egg populations and root galling compared to defoliation without stalk cutting. In a greenhouse bioassay which used soil from the field plots, plants grown in soil from the defoliation only treatment had greater root gall ratings and egg counts than in the stalk cutting plus herbicide treatment. Therefore, we conclude that cutting cotton stalks immediately after harvest effectively reduces M. incognita reproduction, and may lead to a lower initial population density of this nematode in the following year.  相似文献

Recent collections of tetrasporangiate “Heterosiphonia” japonica Yendo from Watch Hill to Point Judith, Rhode Island, represent the first report of this nonnative alga in the western Atlantic. Native to the Pacific Ocean, this species was unintentionally introduced into European waters by 1984 and has subsequently invaded the eastern Atlantic Ocean widely from France to Norway and south into the Mediterranean Sea. Thus far, all western Atlantic collections of this species are confined to the outer coast of Rhode Island, and at present are not found in Narragansett Bay or in Long Island Sound along the Connecticut coast. Molecular and morphological studies confirm the identity of this newly introduced invasive species.  相似文献

This is the first study on the south eastern Pacific coast of South America which details long term, interannual variability in the structure of subtidal rocky-bottom kelp-dominated communities before, during, and after the El Niño Southern Oscillation (ENSO) event of 1997–1998 in northern Chile (23°S). The temporal patterns of the main components of these ecosystems, which included Macrocystis integrifolia, Lessonia trabeculata, echinoids and asteroids, were evaluated seasonally between 1996 and 2004. M. integrifolia demonstrated high interannual variability in temporal patterns of abundance. The 1997–1998 ENSO did not significantly modify the temporal patterns of Macrocystis, although local extinction of M. integrifolia beds occurred during negative thermal anomalies in 1999–2000 (La Niña event), facilitating the establishment of urchin dominated “barren grounds”. The abundance of Lessonia trabeculata showed little temporal variability, and this species dominated the deeper regions of the kelp assemblage (8–13 m depth). The structure of the kelp communities in the study area is regulated by a trophic cascade which modulates alternation between kelp dominated areas and sea urchin barrens. In this context, frequent and intense upwelling of cold water high in nutrients favors the establishment and persistence of kelp assemblages. During ENSO, coastal upwellings can mitigate superficial warming of coastal water and increase the nutrient concentration in the water column. Superficial warming during the 1997–1998 ENSO induced spawning by different species of echinoderms, which resulted in major recruitment of these species during 1999. Top-down events, such as the decrease in densities of the asteroids after the 1997–1998 ENSO event, favored increases in densities of benthic grazers, which caused significant decreases in abundance of M. integrifolia. The re-establishment of the adult fraction of the carnivore (starfish) guild coincided with a decrease in the density of sea urchins and thus re-establishment of the kelp. In the temperate south eastern Pacific, oceanographic events, which act on different spatial-temporal scales, trigger trophic cascades that act at local levels, producing interannual variability in the structure of kelp communities. On the other hand, considering the high macroinvertebrate diversity associated with kelp assemblages, the transitions between kelp-dominated areas and sea urchin barrens do not appear to significantly affect the biodiversity of these assemblages of benthic invertebrates.  相似文献

Several mass strandings of beaked whales have recently been correlated with military exercises involving mid-frequency sonar highlighting unknowns regarding hearing sensitivity in these species. We report the hearing abilities of a stranded juvenile beaked whale (Mesoplodon europaeus) measured with auditory evoked potentials. The beaked whale’s modulation rate transfer function (MRTF) measured with a 40-kHz carrier showed responses up to an 1,800 Hz amplitude modulation (AM) rate. The MRTF was strongest at the 1,000 and 1,200 Hz AM rates. The envelope following response (EFR) input–output functions were non-linear. The beaked whale was most sensitive to high frequency signals between 40 and 80 kHz, but produced smaller evoked potentials to 5 kHz, the lowest frequency tested. The beaked whale hearing range and sensitivity are similar to other odontocetes that have been measured.  相似文献

The type species of the genera Echinospirifer Ljaschenko, 1973 Komispirifer Ljaschenko, 1973 and Mennespirifer Ljaschenko, 1973 were revised based on material from the memorial collection of A.I. Ljaschenko. Mennespirifer is recognized as a junior synonym of the genus Komispirifer; the genera Echinospirifer and Komispirifer differ in fine ornament and the structure of the shell wall.  相似文献
